    The early stages of transform margin formation are associated with crustal fragmentation and block rotation. The restricted size of the resultant microcontinental blocks precludes palaeogeographical reconstructions and reliable estimations of the amount of rotation they can undergo. An example considered here is the Falkland Plateau. This is located adjacent to the Agulhas–Falkland Fracture Zone and its westernmost province is the Falkland Islands microcontinent. The position of the plateau and the islands prior to Gondwana break-up remains contentious. This study integrates seismic reflection and gravity data to propose a revised position of the Falkland Islands microcontinent constrained by (1) the presence of a mega-décollement, controlling the Gondwanide Orogen, described north of the Falkland Islands and underneath South Africa and the Outeniqua Basin, and (2) the similar architecture of fault networks mapped north of the islands and in the northernmost Outeniqua Basin. This revised position requires a re-evaluation of the timing and rate of rotation of the Falkland Islands microcontinent and affects the expected crustal architecture adjacent to the islands. Our model yields rotation rates between 5.5° and 8° Ma−1 and two potential times for rotation, and predicts more unstretched crust beneath the basin east of the Falkland Islands than previous model

    A secondary data analysis of a cluster randomized controlled trial: improved cookstoves associated with reduction in incidence of low birthweight in rural Malawi.

    BACKGROUND: In northern rural Malawi, the majority of households cook using open fires and there is also a high burden of adverse birth outcomes. The use of open fires or highly polluting cookstoves is associated with low birthweight in babies. There is mixed evidence on whether implementation of cleaner burning cookstoves reduces the number of babies born with low birthweight. METHODS: This is a secondary analysis of a cluster randomized control trial in Malawi, conducted over 2014-17. Households were randomized to receive improved cookstoves or to continue current practices. For this analysis, the primary outcome was low birthweight in households under routine demographic surveillance, among births occurring within the trial time frame (N = 4010). A subset of data with stricter exposure definitions respecting the original randomized allocation was also analysed (N = 1050). A causal, forwards modelling approach was used. RESULTS: The main dataset showed evidence of effect of the intervention on low birthweight [adjusted odds ratio (aOR) 0.69; 95% CI 0.48-0.99, n = 2788). The subset analysis lacked power to provide evidence of association between improved cookstoves and low birthweight in the stricter exposure definition (aOR 0.62; 95% CI 0.35-1.09, n = 932). CONCLUSIONS: This study provides some evidence that an improved cookstove intervention in rural Malawi reduced the number of babies born with low birthweight by 30%. This direction of the effect was also seen in the subset analysis. The analysis suggests that the intervention reduced the number of infants born prematurely or with intra-uterine growth restriction, indicating that improved cookstoves could be a useful maternal health intervention

    Spatio-temporal trends in normal-fault segmentation recorded by low-temperature thermochronology: Livingstone fault scarp, Malawi Rift, East African Rift System

    The evolution of through-going normal-fault arrays from initial nucleation to growth and subsequent interaction and mechanical linkage is well documented in many extensional provinces. Over time, these processes lead to predictable spatial and temporal variations in the amount and rate of displacement accumulated along strike of individual fault segments, which should be manifested in the patterns of footwall exhumation. Here, we investigate the along-strike and vertical distribution of low-temperature apatite (U–Th)/He (AHe) cooling ages along the bounding fault system, the Livingstone fault, of the Karonga Basin of the northern Malawi Rift. The fault evolution and linkage from rift initiation to the present day has been previously constrained through investigations of the hanging wall basin fill. The new cooling ages from the footwall of the Livingstone fault can be related to the adjacent depocentre evolution and across a relay zone between two palaeo-fault segments. Our data are complimented by published apatite fission-track (AFT) data and reveal significant variation in rock cooling history along-strike: the centre of the footwall yields younger cooling ages than the former tips of earlier fault segments that are now linked. This suggests that low-temperature thermochronology can detect fault interactions along strike. That these former segment boundaries are preserved within exhumed footwall rocks is a function of the relatively recent linkage of the system. Our study highlights that changes in AHe (and potentially AFT) ages associated with the along-strike displacement profile can occur over relatively short horizontal distances (of a few kilometres). This is fundamentally important in the assessment of the vertical cooling history of footwalls in extensional systems: temporal differences in the rate of tectonically driven exhumation at a given location along fault strike may be of greater importance in controlling changes in rates of vertical exhumation than commonly invoked climatic fluctuations

    Abstract This paper examines the welfare effects of market segmentation using the example of the video rental and sales markets for VHS and DVD formats. I consider a market with two types of buyers: institutional and individual buyers. Institutional buyers rent the good to low-value individuals, while relatively high-value individuals purchase the good directly. Copyright law in the U.S. effectively prevents market segmentation according to use of the product. As a result, rental and sales markets for VHS cassettes are traditionally segmented by pricing VHS cassettes intertemporally: first setting a very high price for the institutional buyers, and lowering the price six months later for sales to end-users. When pricing the DVD format over the past three years, firms have chosen to engage in no price discrimination. Using data on the rental and sales activities of video rental stores for both VHS and DVD formats, I estimate a model of intertemporal price discrimination which rationalizes the use of such different pricing policies for the same movie on two different formats. I use the estimated parameter values to predict whether or not the elimination of intertemporal price discrimination will remain the optimal strategy as DVD hardware penetration evolves, and to estimate the effects of the restrictive copyright law for firms and consumers

    Phase I/II Study of the WEE1 Inhibitor Adavosertib (AZD1775) in Combination with Carboplatin in Children with Advanced Malignancies: Arm C of the AcSé-ESMART Trial

    International audienceAbstract Purpose: AcSé-ESMART Arm C aimed to define the recommended dose and activity of the WEE1 inhibitor adavosertib in combination with carboplatin in children and young adults with molecularly enriched recurrent/refractory malignancies. Patients and Methods: Adavosertib was administered orally, twice every day on Days 1 to 3 and carboplatin intravenously on Day 1 of a 21-day cycle, starting at 100 mg/m2/dose and AUC 5, respectively. Patients were enriched for molecular alterations in cell cycle and/or homologous recombination (HR). Results: Twenty patients (median age: 14.0 years; range: 3.4–23.5) were included; 18 received 69 treatment cycles. Dose-limiting toxicities were prolonged grade 4 neutropenia and grade 3/4 thrombocytopenia requiring transfusions, leading to two de-escalations to adavosertib 75 mg/m2/dose and carboplatin AUC 4; no recommended phase II dose was defined. Main treatment-related toxicities were hematologic and gastrointestinal. Adavosertib exposure in children was equivalent to that in adults; both doses achieved the cell kill target. Overall response rate was 11% (95% confidence interval, 0.0–25.6) with partial responses in 2 patients with neuroblastoma. One patient with medulloblastoma experienced unconfirmed partial response and 5 patients had stable disease beyond four cycles. Seven of these eight patients with clinical benefit had alterations in HR, replication stress, and/or RAS pathway genes with or without TP53 alterations, whereas TP53 pathway alterations alone (8/10) or no relevant alterations (2/10) were present in the 10 patients without benefit. Conclusions: Adavosertib–carboplatin combination exhibited significant hematologic toxicity. Activity signals and identified potential biomarkers suggest further studies with less hematotoxic DNA-damaging therapy in molecularly enriched pediatric cancers
