    Temocillin: a new candidate antibiotic for local antimicrobial delivery in orthopaedic surgery?

    Objectives - To assess the performance of the Gram-negative-specific antibiotic temocillin in polymethylmethacrylate bone cement pre-loaded with gentamicin, as a strategy for local antibiotic delivery. Methods - Temocillin was added at varying concentrations to commercial gentamicin-loaded bone cement. The elution of the antibiotic from cement samples over a 2 week period was quantified by LC-MS. The eluted temocillin was purified by fast protein liquid chromatography and the MICs for a number of antibiotic-resistant Escherichia coli were determined. The impact strength of antibiotic-loaded samples was determined using a Charpy-type impact testing apparatus. Results - LC-MS data showed temocillin eluted to clinically significant concentrations within 1 h in this laboratory system and the eluted temocillin retained antimicrobial activity against all organisms tested. Impact strength analysis showed no significant difference between cement samples with or without temocillin. Conclusions - Temocillin can be added to bone cement and retains its antimicrobial activity after elution. The addition of up to 10% temocillin did not affect the impact strength of the cement. The results show that temocillin is a promising candidate for use in antibiotic-loaded bone cement.</p

    Multiple innovations underpinned branching form diversification in mosses

    International audienceBroad-scale evolutionary comparisons have shown that branching forms arose by convergencein vascular plants and bryophytes, but the trajectory of branching form diversificationin bryophytes is unclear. Mosses are the most species-rich bryophyte lineage andtwo sub-groups are circumscribed by alternative reproductive organ placements. In one,reproductive organs form apically, terminating growth of the primary shoot (gametophore)axis. In the other, reproductive organs develop on very short lateral branches. Aswitch from apical to lateral reproductive organ development is proposed to have primedbranching form diversification. Moss gametophores have modular development and each module develops from a singleapical cell. Here we define the architectures of 175 mosses by the number of module classes,branching patterns and the pattern in which similar modules repeat. Using ancestral characterstate reconstruction we identify two stages of architectural diversification. During a first stage there were sequential changes in the module repetition pattern, reproductiveorgan position, branching pattern and the number of module classes. During a secondstage, vegetative changes occurred independently of reproductive fate.     Structural neural networks subserving oculomotor function in first-episode schizophrenia

    BACKGROUND: Smooth pursuit and antisaccade abnormalities are well documented in schizophrenia, but their neuropathological correlates remain unclear. METHODS: In this study, we used statistical parametric mapping to investigate the relationship between oculomotor abnormalities and brain structure in a sample of first-episode schizophrenia patients (n = 27). In addition to conventional volumetric magnetic resonance imaging, we also used magnetization transfer ratio, a technique that allows more precise tissue characterization. RESULTS: We found that smooth pursuit abnormalities were associated with reduced magnetization transfer ratio in several regions, predominantly in the right prefrontal cortex. Antisaccade errors correlated with gray matter volume in the right medial superior frontal cortex as measured by conventional magnetic resonance imaging but not with magnetization transfer ratio. CONCLUSIONS: These preliminary results demonstrate that specific structural abnormalities are associated with abnormal eye movements in schizophrenia

    Changes in the Frontotemporal Cortex and Cognitive Correlates in First-Episode Psychosis

    Background: Loss of cortical volume in frontotemporal regions has been reported in patients with schizophrenia and their relatives. Cortical area and thickness are determined by different genetic processes, and measuring these parameters separately may clarify disturbances in corticogenesis relevant to schizophrenia. Our study also explored clinical and cognitive correlates of these parameters.Methods: Thirty-seven patients with first-episode psychosis (34 schizophrenia, 3 schizoaffective disorder) and 38 healthy control subjects matched for age and sex took part in the study. Imaging was performed on an magnetic resonance imaging 1.5-T scanner. Area and thickness of the frontotemporal cortex were measured using a surface-based morphometry method (Freesurfer). All subjects underwent neuropsychologic testing that included measures of premorbid and current IQ, working and verbal memory, and executive function.Results: Reductions in cortical area, more marked in the temporal cortex, were present in patients. Overall frontotemporal cortical thickness did not differ between groups, although regional thinning of the right superior temporal region was observed in patients. There was a significant association of both premorbid IQ and IQ at disease onset with area, but not thickness, of the frontotemporal cortex, and working memory span was associated with area of the frontal cortex. These associations remained significant when only patients with schizophrenia were considered.Conclusions: Our results suggest an early disruption of corticogenesis in schizophrenia, although the effect of subsequent environmental factors cannot be excluded. In addition, cortical abnormalities are subject to regional variations and differ from those present in neurodegenerative diseases

    Tomato: a crop species amenable to improvement by cellular and molecular methods

    Tomato is a crop plant with a relatively small DNA content per haploid genome and a well developed genetics. Plant regeneration from explants and protoplasts is feasable which led to the development of efficient transformation procedures. In view of the current data, the isolation of useful mutants at the cellular level probably will be of limited value in the genetic improvement of tomato. Protoplast fusion may lead to novel combinations of organelle and nuclear DNA (cybrids), whereas this technique also provides a means of introducing genetic information from alien species into tomato. Important developments have come from molecular approaches. Following the construction of an RFLP map, these RFLP markers can be used in tomato to tag quantitative traits bred in from related species. Both RFLP's and transposons are in the process of being used to clone desired genes for which no gene products are known. Cloned genes can be introduced and potentially improve specific properties of tomato especially those controlled by single genes. Recent results suggest that, in principle, phenotypic mutants can be created for cloned and characterized genes and will prove their value in further improving the cultivated tomato.

    Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage.     Vegetation responses to abrupt climatic changes during the Last Interglacial Complex (Marine Isotope Stage 5) at Tenaghi Philippon, NE Greece

    The discovery that climate variability during the Last Glacial shifted rapidly between climate states has intensified efforts to understand the distribution, timing and impact of abrupt climate change under a wide range of boundary conditions. In contribution to this, we investigate the nature of abrupt environmental changes in terrestrial settings of the Mediterranean region during the Last Interglacial Complex (Marine Isotope Stage [MIS] 5) and explore the relationships of these changes to high-latitude climate events. We present a new, temporally highly resolved (mean: 170 years) pollen record for the Last Interglacial Complex from Tenaghi Philippon, north-east Greece. The new pollen record, which spans the interval from 130,000 to 65,000 years ago, forms part of an exceptionally long polleniferous sediment archive covering the last 1.35 million years. The pollen data reveal an interglacial followed by alternating forest and steppe phases representing the interstadials and stadials of the Early Glacial. Superimposed on these millennial-scale changes is evidence of persistent sub-millennial-scale variability. We identify ten high-amplitude abrupt events in the pollen record, characterised by rapid contractions of closed forest to open steppe environment and interpreted to indicate major changes in moisture availability and temperature. The contractions in forest cover on millennial timescales appear associated with cooling events in the Mediterranean Sea, North Atlantic and Greenland regions, linked to the Dansgaard-Oeschger (DO) cycles of the Early Glacial. On sub-millennial timescales, the pattern of changes in forest cover at Tenaghi Philippon display a structure similar to the pattern of short-lived precursor and rebound-type events detected in the Greenland ice-core record. Our findings indicate that persistent, high-amplitude environmental variability occurred throughout the Early Glacial, on both millennial and submillennial timescales. Furthermore, the similarity of the pattern of change between Tenaghi Philippon and Greenland on sub-millennial timescales suggests that teleconnections between the high-latitudes and the Mediterranean region operate on sub-millennial timescales and that some terrestrial archives, such as Tenaghi Philippon, are particularly sensitive recorders of these abrupt climate changes
