    Lignin content analysis in oil palm frond juice base medium: effect on bacterial cellulose production by Acetobacter xylinum 0416

    Extensive work on bacterial cellulose (BC) cultivation is crucial in optimizing BC performance and versatility in application. Herein, this study aims to investigate the lignin content in oil palm frond (OPF) juice medium that could affect the BC production by Acetobacter xylinum 0416 in static culture. The effects of lignin content were observed by the performances of BC production from the raw and pre-treated OPF juice medium fermentation with Hestrin-Schramm (HS) medium as a point of comparison. The TAPPI methods analysis outlined up to 3.719 g/L of total lignin content in raw OPF juice and FTIR analysis depicted the functional group of a typical lignin compound, thus confirming its presence as a component inhibitor. The raw OPF juice was further pre-treated using an alkaline precipitation method resulting in acid-insoluble lignin removal up to 75.47% of the total lignin content. The results revealed that the BC yield from the raw OPF juice fermentation was lower compared to the HS medium with dried BC content of 0.152 g/L and 2.52 g/L, respectively. The BC cultivation using the pre-treated OPF juice medium was improved as the BC yield significantly increased to 1.91 g/L. Therefore, a proper pre-treatment strategy on OPF juice medium for lignin content removal could improve the medium capability in BC cultivation for higher-scale production

    Associations between diet and disease activity in ulcerative colitis patients using a novel method of data analysis

    BACKGROUND: The relapsing nature and varying geographical prevalence of ulcerative colitis (UC) implicates environmental factors such as diet in its aetiology. METHODS: In order to determine which foods might be related to disease activity in UC a new method of dietary analysis was developed and applied. Eighty-one UC patients were recruited at all stages of the disease process. Following completion of a 7 d diet diary, clinical assessment including a sigmoidoscopic examination (scale 0 (normal mucosa) to 6 (very active disease)) was conducted. Food weights for each person were adjusted (divided) by the person's calorific intake for the week. Each food consumed was given a food sigmoidoscopy score (FSS) calculated by summing the products of the (adjusted) weight of food consumed and sigmoidoscopy score for each patient and occurrence of food and dividing by the total (adjusted) weight of the food consumed by all 81 patients. Thus, foods eaten in large quantities by patients with very active disease have high FSSs and vice versa. Foods consumed by <10 people or weighing <1 kg for the whole group were excluded, leaving 75 foods. RESULTS: High FSS foods were characterized by high levels of the anti-thiamin additive sulfite (Mann-Whitney, p < 0.001), i.e. bitter, white wine, burgers, soft drinks from concentrates, sausages, lager and red wine. Caffeine also has anti-thiamin properties and decaffeinated coffee was associated with a better clinical state than the caffeine containing version. Beneficial foods (average intake per week) included pork (210 g), breakfast cereals (200 g), lettuce (110 g), apples and pears (390 g), milk (1250 ml), melon (350 g), bananas (350 g), bacon (120 g), beef and beef products (500 g), tomatoes (240 g), soup (700 g), citrus fruits (300 g), fish (290 g), yogurt (410 g), cheese (110 g), potatoes (710 g) and legumes (120 g). CONCLUSIONS: The dietary analysis method described provides a new tool for establishing relationships between diet and disease and indicates a potentially therapeutic diet for UC

    Integrated genomic characterization of oesophageal carcinoma

    Oesophageal cancers are prominent worldwide; however, there are few targeted therapies and survival rates for these cancers remain dismal. Here we performed a comprehensive molecular analysis of 164 carcinomas of the oesophagus derived from Western and Eastern populations. Beyond known histopathological and epidemiologic distinctions, molecular features differentiated oesophageal squamous cell carcinomas from oesophageal adenocarcinomas. Oesophageal squamous cell carcinomas resembled squamous carcinomas of other organs more than they did oesophageal adenocarcinomas. Our analyses identified three molecular subclasses of oesophageal squamous cell carcinomas, but none showed evidence for an aetiological role of human papillomavirus. Squamous cell carcinomas showed frequent genomic amplifications of CCND1 and SOX2 and/or TP63, whereas ERBB2, VEGFA and GATA4 and GATA6 were more commonly amplified in adenocarcinomas. Oesophageal adenocarcinomas strongly resembled the chromosomally unstable variant of gastric adenocarcinoma, suggesting that these cancers could be considered a single disease entity. However, some molecular features, including DNA hypermethylation, occurred disproportionally in oesophageal adenocarcinomas. These data provide a framework to facilitate more rational categorization of these tumours and a foundation for new therapies.ope

    Incident type 2 diabetes attributable to suboptimal diet in 184 countries

    The global burden of diet-attributable type 2 diabetes (T2D) is not well established. This risk assessment model estimated T2D incidence among adults attributable to direct and body weight-mediated effects of 11 dietary factors in 184 countries in 1990 and 2018. In 2018, suboptimal intake of these dietary factors was estimated to be attributable to 14.1 million (95% uncertainty interval (UI), 13.814.4 million) incident T2D cases, representing 70.3% (68.871.8%) of new cases globally. Largest T2D burdens were attributable to insufficient whole-grain intake (26.1% (25.027.1%)), excess refined rice and wheat intake (24.6% (22.327.2%)) and excess processed meat intake (20.3% (18.323.5%)). Across regions, highest proportional burdens were in central and eastern Europe and central Asia (85.6% (83.487.7%)) and Latin America and the Caribbean (81.8% (80.183.4%)); and lowest proportional burdens were in South Asia (55.4% (52.160.7%)). Proportions of diet-attributable T2D were generally larger in men than in women and were inversely correlated with age. Diet-attributable T2D was generally larger among urban versus rural residents and higher versus lower educated individuals, except in high-income countries, central and eastern Europe and central Asia, where burdens were larger in rural residents and in lower educated individuals. Compared with 1990, global diet-attributable T2D increased by 2.6 absolute percentage points (8.6 million more cases) in 2018, with variation in these trends by world region and dietary factor.     Children's and adolescents' rising animal-source food intakes in 1990-2018 were impacted by age, region, parental education and urbanicity

    Animal-source foods (ASF) provide nutrition for children and adolescents physical and cognitive development. Here, we use data from the Global Dietary Database and Bayesian hierarchical models to quantify global, regional and national ASF intakes between 1990 and 2018 by age group across 185 countries, representing 93% of the worlds child population. Mean ASF intake was 1.9 servings per day, representing 16% of children consuming at least three daily servings. Intake was similar between boys and girls, but higher among urban children with educated parents. Consumption varied by age from 0.6 at <1 year to 2.5 servings per day at 1519 years. Between 1990 and 2018, mean ASF intake increased by 0.5 servings per week, with increases in all regions except sub-Saharan Africa. In 2018, total ASF consumption was highest in Russia, Brazil, Mexico and Turkey, and lowest in Uganda, India, Kenya and Bangladesh. These findings can inform policy to address malnutrition through targeted ASF consumption programmes. (c) 2023, The Author(s)

    Pan-cancer analysis of whole genomes

    Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale(1-3). Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4-5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter(4); identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation(5,6); analyses timings and patterns of tumour evolution(7); describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity(8,9); and evaluates a range of more-specialized features of cancer genomes(8,10-18).Peer reviewe

    We performed an extensive immunogenomic anal-ysis of more than 10,000 tumors comprising 33diverse cancer types by utilizing data compiled byTCGA. Across cancer types, we identified six im-mune subtypes\u2014wound healing, IFN-gdominant,inflammatory, lymphocyte depleted, immunologi-cally quiet, and TGF-bdominant\u2014characterized bydifferences in macrophage or lymphocyte signa-tures, Th1:Th2 cell ratio, extent of intratumoral het-erogeneity, aneuploidy, extent of neoantigen load,overall cell proliferation, expression of immunomod-ulatory genes, and prognosis. Specific drivermutations correlated with lower (CTNNB1,NRAS,orIDH1) or higher (BRAF,TP53,orCASP8) leukocytelevels across all cancers. Multiple control modalitiesof the intracellular and extracellular networks (tran-scription, microRNAs, copy number, and epigeneticprocesses) were involved in tumor-immune cell inter-actions, both across and within immune subtypes.Our immunogenomics pipeline to characterize theseheterogeneous tumors and the resulting data areintended to serve as a resource for future targetedstudies to further advance the field

    Influence of drying method on the crystal structure and thermal property of oil palm frond juice-based bacterial cellulose

    Dehydration of wet bacterial cellulose (BC) mainly focuses on the physical characteristic or mechanical strength, but its polymorph crystal structure and thermal properties were rarely discussed. Herein, this study utilizes the drying method of hot-pressed and freeze-dried to the wet BC to elucidate the BC polymorph crystal structure in correlation with the BC crystallinity and thermal properties. The results indicate that dehydration of BC using hot-pressed (BC-HP) method reduced the cellulose Iα allomorph fraction by ± 5.0%, increased the crystallinity by ± 5.0%, but lower in thermal stability compared to freeze-dried (BC-FD) method. Additionally, BC-HP shows an exothermic reaction of melting at lower enthalpy value which is contradict to BC-FD and plant cellulose that presented an endothermic reaction with higher enthalpy values. Thus, investigation on BC polymorph crystal structure and thermal properties may offer an important role to regulate the desired characteristic of the dried BC

    Production and Characterization of Bacterial Cellulose Nanofiber by Acetobacter Xylinum 0416 Using Only Oil Palm Frond Juice as Fermentation Medium

    Renewable fermentation feedstock from a freshly cut oil palm frond (OPF) is a promising medium for bacterial cellulose (BC) production. Herein, this study assessed its suitability as a solitary medium for the production of BC using Acetobacter xylinum 0416 in a static culture. The proximate composition and components analysis revealed up to 36.2 g/L of fermentable sugars and vital nutrients were available in OPF juice. Meanwhile, the effect of OPF juice concentration and initial pH arrangement shows that the maximum yield of BC (2.33 g/L) was obtained using 60% v/v of OPF juice concentration at initial pH 3.5 and incubated at 30℃ for 14 days. Besides, pure OPF juice showed a minimum in BC production due to the presence of inhibitory component suspected to be the lignin content. The kinetic study was elucidated for BC produced from OPF juice and compared with coconut water medium. The FT-IR and XRD spectra, and FE-SEM micrograph analysis for the BC showed the typical peaks represent in cellulose with a crystallinity index of 82.4% and threadlike microfibrils structure ranging from 35.6.5 nm to 77.5 nm. Thus, OPF juice is a solely sufficient medium that offers a positive alternative to BC production
