    Efficiency for Large Fleets: An Analysis of How Industrial Customers Use Electric and Hybrid Vehicles

    This paper presents methods and results which allow an analysis of relevant driving parameters of hybrid and electric vehicles. In order to gain crucial insights into how industrial customers use hybrid and electric vehicles, the paper investigates the following parameters: information about odometer, charging processes and battery charging levels. The data used for this purpose was provided by the Canadian fleet management company Geotab Inc. They were evaluated by means of ‘Google BigQuery’ and the statistics programme ‘IBM SPSS Statistics’. It turned out that correlations between ‘charging time’ and ‘battery charging level’ exist, as well as between ‘battery level’ and ‘distance per day’. One of the main questions in the present study asks whether long charging times of car batteries lead to decreased average battery charging levels. As a result of this study, the longer a hybrid vehicle is charged per day, the lower sink its average battery charging level. The findings of this research help managers of car fleets to enhance their existing fleet management for establishing more efficient fleets with respect to ecological and economical aspects. Our research is particularly significant as this will save money across such fleets worldwide, and at the same time, preserve the environment as much as possible.     Buried alive: Aquatic plants survive in ‘ghost ponds’ under agricultural fields

    The widespread loss of wetlands due to agricultural intensification has been highlighted as a major threat to aquatic biodiversity. However, all is not lost as we reveal that the propagules of some aquatic species could survive burial under agricultural fields in the sediments of ‘ghost ponds’ - ponds in-filled during agricultural land consolidation. Our experiments showed at least eight aquatic macrophyte species to germinate from seeds and oospores, following 50–150 years of dormancy in the sediments of ghost ponds. This represents a significant proportion of the expected macrophyte diversity for local farmland ponds, which typically support between 6 and 14 macrophyte species. The rapid (< 6 months) re-colonisation of resurrected ghost ponds by a diverse aquatic vegetation similarly suggests a strong seed-bank influence. Ghost ponds represent abundant, dormant time capsules for aquatic species in agricultural landscapes around the globe, affording opportunities for enhancing landscape-scale aquatic biodiversity and connectivity. While reports of biodiversity loss through agricultural intensification dominate conservation narratives, our study offers a rare positive message, demonstrating that aquatic organisms survive prolonged burial under intensively managed agricultural fields. We urge conservationists and policy makers to consider utilizing and restoring these valuable resources in biodiversity conservation schemes and in agri-environmental approaches and policies

    Assertive community treatment for elderly people with severe mental illness

    Background: Adults aged 65 and older with severe mental illnesses are a growing segment of the Dutch population. Some of them have a range of serious problems and are also difficult to engage. While assertive community treatment is a common model for treating difficult to engage severe mental illnesses patients, no special form of it is available for the elderly. A special assertive community treatment team for the elderly is developed in Rotterdam, the Netherlands and tested for its effectiveness.Methods: We will use a randomized controlled trial design to compare the effects of assertive community treatment for the elderly with those of care as usual. Primary outcome measures will be the number of dropouts, the number of patients engaged in care and patient's psychiatric symptoms, somatic symptoms, and social functioning. Secondary outcome measures are the number of unmet needs, the subjective quality of life and patients' satisfaction. Other secondary outcomes include the number of crisis contacts, rates of voluntary and involuntary admission, and length of stay. Inclusion criteria are aged 65 plus, the presence of a mental disorder, a lack of motivation for treatment and at least four suspected problems with functioning (addiction, somatic problems, daily living activities, housing etc.). If patients meet the inclusion criteria, they will be randomly allocated to either assertive community treatment for the elderly or care as usual. Trained assessors will use mainly observational instruments at the following time points: at baseline, after 9 and 18 months.Discussion: This study will help establish whether assertive community treatment for the elderly produces better results than care as usual in elderly people with severe mental illnesses who are difficult to engage. When assertive community treatment for the elderly proves valuable in these respects, it can be tested and implemented more widely, and mechanisms for its effects investigated

    Pan-cancer analysis of whole genomes

    Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale(1-3). Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4-5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter(4); identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation(5,6); analyses timings and patterns of tumour evolution(7); describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity(8,9); and evaluates a range of more-specialized features of cancer genomes(8,10-18).Peer reviewe

    Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples

    Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content.
