    Assessing environmental awareness towards protection of the Alps: a case study

    The paper focuses on environmental awareness towards the protection of Alpine areas, and presents the results of a research aimed at assessing people\u2019s awareness in regards to certain topical environmental issues such as protected areas, ecological connectivity, and wildlife. The study area is the Julian Prealps Natural Park, in the region of Friuli Venezia Giulia (Northeast Italy). In order to obtain a measure of \u201cenvironmental awareness\u201d, the Rasch model was applied. It allowed us first to identify which items are most suitable for this. The results obtained show that people are quite aware of some issues, e.g. that wellbeing is related to greener areas and that they can change things to solve environmental problems. We also found that it is easier to agree with items relating to the environment in general than with more specific issues, probably due to a lack of knowledge. However, the provision of pertinent information, in our case on ecological connectivity, can increase the level of environmental awareness. The model also showed that the level varies according to certain sociodemographic characteristics, particularly gender and age. These results could serve as a starting point for planning effective information activities aimed at raising environmental awareness and, possibly, bolstering participation in initiatives for protecting Alpine areas

    The industrial districts have given rise to a variety of biocultural paths, but in the post-industrial era these are continuously threatened by the trend towards product standardisation. Despite this, local districts are able to survive, albeit in a weakened state, with two possible effects: A) the risk of fading; B) the desirability of resilience. The Biocultural fingerprint of the Prosecco District (placed 3rd in the Intesa San Paolo ranking of Italian districts for 2008-2011) reveals that its clusters of municipalities are characterised by agricultural and eno-gastronomic excellence in both rural and urban contexts. Meanwhile, its tourist attractiveness index, owing to its lack of apparently "useless components" such as nature parks and protected areas, and cultural heritage, is relatively low. It follows that the marketing-image strategies devised for immediate profit appear to be insufficient at coping with the competition and ensuring a sustained level of development in the long term

    Percezioni paesistico-ambientali nelle microfiliere di qualit\ue0 (Farmers and Landscape: Environmental Perceptions in Quality Micro-Chains)

    Among the phenomena of transition from useless to useful in the landscape-cultural mosaic is that of the farmers\u2019 changing perceptions of the environment and the creation of externalities to their micro-chain underlying business, or those of others. The objective of this study is to verify whether there is a connection between those perceptions and the management classes of their farms, in regards to both environmental aspects and to their participation in joint initiatives. From the statistical analysis of 30 FVG micro-chains, it was found that multi-active advanced farms perceive most of the improvements that are derived from their activities or the activities of others. Additionally, their willingness to engage in joint-initiatives is significant in certain aspects. Whereas amateur farmers are sceptical about their ability to positively affect the landscape and the environment, conventional farmers are much more confident that their own actions are more effective than those of others. In regards to joint initiatives, neither conventional nor amateur farmers are at all willing to engage in the collection and disposal of waste, whilst they do appear to be more positive in relation to territorial branding and, moderately, to retail stores

    Antifungal Activity and DNA Topoisomerase Inhibition of Hydrolysable Tannins from Punica granatum L

    Punica granatum L. (pomegranate) fruit is known to be an important source of bioactive phenolic compounds belonging to hydrolysable tannins. Pomegranate extracts have shown antifungal activity, but the compounds responsible for this activity and their mechanism/s of action have not been completely elucidated up to now. The aim of the present study was the investigation of the inhibition ability of a selection of pomegranate phenolic compounds (i.e., punicalagin, punicalin, ellagic acid, gallic acid) on both plant and human fungal pathogens. In addition, the biological target of punicalagin was identified here for the first time. The antifungal activity of pomegranate phenolics was evaluated by means of Agar Disk Diffusion Assay and minimum inhibitory concentration (MIC) evaluation. A chemoinformatic analysis predicted for the first time topoisomerases I and II as potential biological targets of punicalagin, and this prediction was confirmed by in vitro inhibition assays. Concerning phytopathogens, all the tested compounds were effective, often similarly to the fungicide imazalil at the label dose. Particularly, punicalagin showed the lowest MIC for Alternaria alternata and Botrytis cinerea, whereas punicalin was the most active compound in terms of growth control extent. As for human pathogens, punicalagin was the most active compound among the tested ones against Candida albicans reference strains, as well as against the clinically isolates. UHPLC coupled with HRMS indicated that C. albicans, similarly to the phytopathogen Coniella granati, is able to hydrolyze both punicalagin and punicalin as a response to the fungal attack. Punicalagin showed a strong inhibitory activity, with IC50 values of 9.0 and 4.6 µM against C. albicans topoisomerases I and II, respectively. Altogether, the results provide evidence that punicalagin is a valuable candidate to be further exploited as an antifungal agent in particular against human fungal infections

    Effects of cytokine blocking agents on hospital mortality in patients admitted to ICU with acute respiratory distress syndrome by SARS-CoV-2 infection: Retrospective cohort study

    Background: The use of cytokine-blocking agents has been proposed to modulate the inflammatory response in patients with COVID-19. Tocilizumab and anakinra were included in the local protocol as an optional treatment in critically ill patients with acute respiratory distress syndrome (ARDS) by SARS-CoV-2 infection. This cohort study evaluated the effects of therapy with cytokine blocking agents on in-hospital mortality in COVID-19 patients requiring mechanical ventilation and admitted to intensive care unit. Methods: The association between therapy with tocilizumab or anakinra and in-hospital mortality was assessed in consecutive adult COVID-19 patients admitted to our ICU with moderate to severe ARDS. The association was evaluated by comparing patients who received to those who did not receive tocilizumab or anakinra and by using different multivariable Cox models adjusted for variables related to poor outcome, for the propensity to be treated with tocilizumab or anakinra and after patient matching. Results: Sixty-six patients who received immunotherapy (49 tocilizumab, 17 anakinra) and 28 patients who did not receive immunotherapy were included. The in-hospital crude mortality was 30,3% in treated patients and 50% in non-treated (OR 0.77, 95% CI 0.56-1.05, p=0.069). The adjusted Cox model showed an association between therapy with immunotherapy and in-hospital mortality (HR 0.40, 95% CI 0.19-0.83, p=0.015). This protective effect was further confirmed in the analysis adjusted for propensity score, in the propensity-matched cohort and in the cohort of patients with invasive mechanical ventilation within 2 hours after ICU admission. Conclusions: Although important limitations, our study showed that cytokine-blocking agents seem to be safe and to improve survival in COVID-19 patients admitted to ICU with ARDS and the need for mechanical ventilation

    DNA methylation levels in candidate genes associated with chronological age in mammals are not conserved in a long-lived seabird

Most seabirds do not have any outward identifiers of their chronological age, so estimation of seabird population age structure generally requires expensive, long-term banding studies. We investigated the potential to use a molecular age biomarker to estimate age in short-tailed shearwaters (Ardenna tenuirostris). We quantified DNA methylation in several A. tenuirostris genes that have shown age-related methylation changes in mammals. In birds ranging from chicks to 21 years of age, bisulphite treated blood and feather DNA was sequenced and methylation levels analysed in 67 CpG sites in 13 target gene regions. From blood samples, five of the top relationships with age were identified in KCNC3 loci (CpG66: R2 = 0.325, p = 0.019). In feather samples ELOVL2 (CpG42: R2 = 0.285, p = 0.00048) and EDARADD (CpG46: R2 = 0.168, p = 0.0067) were also weakly correlated with age. However, the majority of markers had no clear association with age (of 131 comparisons only 12 had a p-value < 0.05) and statistical analysis using a penalised lasso approach did not produce an accurate ageing model. Our data indicate that some age-related signatures identified in orthologous mammalian genes are not conserved in the long-lived short tailed shearwater. Alternative molecular approaches will be required to identify a reliable biomarker of chronological age in these seabirds

    Pan-cancer analysis of whole genomes

    Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale(1-3). Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4-5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter(4); identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation(5,6); analyses timings and patterns of tumour evolution(7); describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity(8,9); and evaluates a range of more-specialized features of cancer genomes(8,10-18).Peer reviewe
