    Mass Azithromycin Distribution and Community Microbiome: A Cluster-Randomized Trial.

    BackgroundMass distributions of oral azithromycin have long been used to eliminate trachoma, and they are now being proposed to reduce childhood mortality. The observed benefit appears to be augmented with each additional treatment, suggesting a possible community-level effect. Here, we assess whether 2 biannual mass treatments of preschool children affect the community's gut microbiome at 6 months after the last distribution.MethodsIn this cluster-randomized controlled trial, children aged 1-60 months in the Dossa region of Niger were randomized at the village level to receive a single dose of azithromycin or placebo every 6 months. Fecal samples were collected 6 months after the second treatment for metagenomic deep sequencing. The prespecified primary outcome was the Euclidean PERMANOVA of the gut microbiome, or effectively the distance between the genus-level centroid at the community level, with the secondary outcome being the Simpson's α diversity.ResultsIn the azithromycin arm, the gut microbial structures were significantly different than in the placebo arm (Euclidean PERMANOVA, P < .001). Further, the diversity of the gut microbiome in the azithromycin arm was significantly lower than in the placebo arm (inverse Simpson's index, P = .005).ConclusionsTwo mass azithromycin administrations, 6 months apart, in preschool children led to long-term alterations of the gut microbiome structure and community diversity. Here, long-term microbial alterations in the community did not imply disease but were associated with an improvement in childhood mortality.Clinical trials registrationNCT02048007

    Cratering and penetration experiments in teflon targets at velocities from 1 to 7 km/s

    Approximately 20 sq m of protective thermal blankets, largely composed of Teflon, were retrieved from the Long Duration Exposure Facility after the spacecraft spent approximately 5.7 years in space. Examination of these blankets revealed that they contained thousands of hypervelocity impact features ranging from micron-sized craters to penetration holes several millimeters in diameter. We conducted impact experiments to reproduce such features and to understand the relationships between projectile size and the resulting crater or penetration hole diameter over a wide range of impact velocities. Such relationships are needed to derive the size and mass frequency distribution and flux of natural and man-made particles in low-earth orbit. Powder propellant and light-gas guns were used to launch soda-lime glass spheres into pure Teflon targets at velocities ranging from 1 to 7 km/s. Target thickness varied over more than three orders of magnitude from finite halfspace targets to very thin films. Cratering and penetration of massive Teflon targets is dominated by brittle failure and the development of extensive spall zones at the target's front and, if penetrated, the target's rear side. Mass removal by spallation at the back side of Teflon targets may be so severe that the absolute penetration hole diameter can become larger than that of a standard crater. The crater diameter in infinite halfspace Teflon targets increases, at otherwise constant impact conditions, with encounter velocity by a factor of V (exp 0.44). In contrast, the penetration hole size in very thin foils is essentially unaffected by impact velocity. Penetrations at target thicknesses intermediate to these extremes will scale with variable exponents of V. Our experimental matrix is sufficiently systematic and complete, up to 7 km/s, to make reasonable recommendations for velocity-scaling of Teflon craters and penetrations. We specifically suggest that cratering behavior and associated equations apply to all impacts in which the shock-pulse duration of the projectile is shorter than that assigned a unique projectile size, provided an impact velocity is known or assumed. This calibration seems superior to the traditional ballistic-limit approach

    Oecophylla smaragdina ants provide pest control in Australian cacao

    Ecosystem service-driven methods of agricultural production that make optimal use of an ecosystem’s natural services are often effective in smallholder crops, leading increases in habitat heterogeneity at large spatial scales that encourage beneficial species. Pollination services in cacao farms can be enhanced through efforts to conserve pollinator habitats, such as adding cacao fruit husks underneath trees to provide habitat and food resources for midge pollinators. However, it remains unclear how this habitat manipulation affects pest densities or biological control. Here, we evaluated the effects of the predaceous ant Oecophylla smaragdina on three major pests of Australian cacao (Helopeltis spp., Amblypelta spp. and Rhyparida nitida) in an Australian cacao farm, in the presence and absence of cacao fruit husks. We also evaluated the effect of O. smaragdina on their potentially plant-damaging, mutualistic mealybug (Pseudococcidae) partners in the presence and absence of cacao fruit husks. O. smaragdina reduced the combined densities of Helopeltis spp. and Amblypelta spp., regardless of cacao fruit husk addition. The ants also reduced the densities of R. nitida in the absence of cacao fruit husks, but R. nitida densities were naturally low in the experimental plots that received the addition of cacao fruit husks, regardless of ant presence. In contrast, ants generally increased the densities of mealybugs. These data suggest that O. smaragdina ants provide pest control services for three major insect pests in Australian cacao farms. Furthermore, this control may be compatible with habitat manipulations (i.e., the addition of cacao fruit husks) to improve pollinator habitat
