    Using Sentinel-2 and canopy height models to derive a landscape-level biomass map covering multiple vegetation types

    Vegetation biomass is a globally important climate-relevant terrestrial carbon pool and also drives local hydrological systems via evapotranspiration. Vegetation biomass of individual vegetation types has been successfully estimated from active and passive remote sensing data. However, for many tasks, landscape-level biomass maps across several vegetation types are more suitable than biomass maps of individual vegetation types. For example, the validation of ecohydrological models and carbon budgeting typically requires spatially continuous biomass estimates, independent from vegetation type. Studies that derive biomass estimates across multiple vegetation or land-cover types to merge them into a single landscape-level biomass map are still scarce, and corresponding workflows must be developed. Here, we present a workflow to derive biomass estimates on landscape-level for a large watershed in central Chile. Our workflow has three steps: First, we combine field plotbased biomass estimates with spectral and structural information collected from Sentinel-2, TanDEM-X and airborne LiDAR data to map grassland, shrubland, native forests and pine plantation biomass using random forest regressions with an automatic feature selection. Second, we predict all models to the entire landscape. Third, we derive a land-cover map including the four considered vegetation types. We then use this land-cover map to assign the correct vegetation type-specific biomass estimate to each pixel according to one of the four considered vegetation types. Using a single repeatable workflow, we obtained biomass predictions comparable to earlier studies focusing on only one of the four vegetation types (Spearman correlation between 0.80 and 0.84; normalized-RMSE below 16 % for all vegetation types). For all woody vegetation types, height metrics were amongst the selected predictors, while for grasslands, only Sentinel-2 bands were selected. The land-cover was also mapped with high accuracy (OA = 83.1 %). The final landscape-level biomass map spatially agrees well with the known biomass distribution patterns in the watershed. Progressing from vegetation-type specific maps towards landscape-level biomass maps is an essential step towards integrating remote-sensing based biomass estimates into models for water and carbon management

Essa arquiteturaconfigurou a prática em sala de aula e foi realizada com 120 alunos doensino básico em uma escola de Porto Alegre. Sua aplicação junto aos alunosresultou em: (1) dedicação intensiva que ultrapassou o tempo da sala de aula;(2) interesse ampliado por criar disponibilidade para a aprendizagem; (3)aprendizagem maximizada.     Evaluation of the DREAM Technique for a High-Throughput Deorphanization of Chemosensory Receptors in Drosophila

    In the vinegar fly Drosophila melanogaster, the majority of olfactory receptors mediating the detection of volatile chemicals found in their natural habitat have been functionally characterized (deorphanized) in vivo. In this process, receptors have been assigned ligands leading to either excitation or inhibition in the olfactory sensory neuron where they are expressed. In other, non-drosophilid insect species, scientists have not yet been able to compile datasets about ligand–receptor interactions anywhere near as extensive as in the model organism D. melanogaster, as genetic tools necessary for receptor deorphanization are still missing. Recently, it was discovered that exposure to artificially high concentrations of odorants leads to reliable alterations in mRNA levels of interacting odorant receptors in mammals. Analyzing receptor expression after odorant exposure can, therefore, help to identify ligand–receptor interactions in vivo without the need for other genetic tools. Transfer of the same methodology from mice to a small number of receptors in D. melanogaster resulted in a similar trend, indicating that odorant exposure induced alterations in mRNA levels are generally applicable for deorphanization of interacting chemosensory receptors. Here, we evaluated the potential of the DREAM (Deorphanization of receptors based on expression alterations in mRNA levels) technique for high-throughput deorphanization of chemosensory receptors in insect species using D. melanogaster as a model. We confirmed that in some cases the exposure of a chemosensory receptor to high concentration of its best ligand leads to measureable alterations in mRNA levels. However, unlike in mammals, we found several cases where either confirmed ligands did not induce alterations in mRNA levels of the corresponding chemosensory receptors, or where gene transcript-levels were altered even though there is no evidence for a ligand–receptor interaction. Hence, there are severe limitations to the suitability of the DREAM technique for deorphanization as a general tool to characterize olfactory receptors in insects

Estetrabalho relata o desenvolvimento e a validação de um software de simulaçãolaboratorial denominado Cromatografia Ascendente em Papel de Aminoácidos usando aferramenta o Adobe® Flash® CS3. A organização e a coleta do material multimídiaocorreram no semestre 2008/2. As imagens mais representativas foram inseridas noobjeto de aprendizagem A primeira aplicação deste objeto foi realizada em 2009/1, naBioquímica I (Farmácia-UFRGS). Com base nesta experiência, uma nova versão foidesenvolvida a qual foi utilizada pelos alunos da mesma disciplina em 2009/2. Apósuma aula expositiva de 50 minutos (teórico-prática), os alunos dos dois semestres foramdivididos em dois grupos. O grupo I respondeu a um questionário sobre conceitosbásicos de cromatografia e depois utilizou o software. O grupo II foi submetido aoprotocolo inverso. Os grupos também avaliaram os aspectos técnicos daanimação/simulação e a atividade realizada. Associando os resultados das duasaplicações (2009/1 e 2009/2), o objeto de aprendizagem em questão pode serconsiderado válido como suporte para o ensino prático de Bioquímica básica

    Anemia prevalence in women of reproductive age in low- and middle-income countries between 2000 and 2018

    Anemia is a globally widespread condition in women and is associated with reduced economic productivity and increased mortality worldwide. Here we map annual 2000–2018 geospatial estimates of anemia prevalence in women of reproductive age (15–49 years) across 82 low- and middle-income countries (LMICs), stratify anemia by severity and aggregate results to policy-relevant administrative and national levels. Additionally, we provide subnational disparity analyses to provide a comprehensive overview of anemia prevalence inequalities within these countries and predict progress toward the World Health Organization’s Global Nutrition Target (WHO GNT) to reduce anemia by half by 2030. Our results demonstrate widespread moderate improvements in overall anemia prevalence but identify only three LMICs with a high probability of achieving the WHO GNT by 2030 at a national scale, and no LMIC is expected to achieve the target in all their subnational administrative units. Our maps show where large within-country disparities occur, as well as areas likely to fall short of the WHO GNT, offering precision public health tools so that adequate resource allocation and subsequent interventions can be targeted to the most vulnerable populations. **Please note that there are multiple authors for this article therefore only the name of the first 30 including Federation University Australia affiliate “Muhammad Aziz Rahman” is provided in this record*

    Removal of Uracil by Uracil DNA Glycosylase Limits Pemetrexed Cytotoxicity: Overriding the Limit with Methoxyamine to Inhibit Base Excision Repair

    Uracil DNA glycosylase (UDG) specifically removes uracil bases from DNA, and its repair activity determines the sensitivity of the cell to anticancer agents that are capable of introducing uracil into DNA. In the present study, the participation of UDG in the response to pemetrexed-induced incorporation of uracil into DNA was studied using isogenic human tumor cell lines with or without UDG (UDG+/+/UDG−/−). UDG−/− cells were very sensitive to pemetrexed. Cell killing by pemetrexed was associated with genomic uracil accumulation, stalled DNA replication, and catastrophic DNA strand breaks. By contrast, UDG+/+ cells were \u3e10 times more resistant to pemetrexed due to the rapid removal of uracil from DNA by UDG and subsequent repair of the resultant AP sites (abasic sites) via the base excision repair (BER). The resistance to pemetrexed in UDG+/+ cells could be reversed by the addition of methoxyamine (MX), which binds to AP sites and interrupts BER pathway. Furthermore, MX-bound AP sites induced cell death was related to their cytotoxic effect of dual inactivation of UDG and topoisomerase IIα, two genes that are highly expressed in lung cancer cells in comparison with normal cells. Thus, targeting BER-based therapy exhibits more selective cytotoxicity on cancer cells through a synthetic lethal mechanism

    Deployment of the C-band radar Poldirad on Barbados during EUREC4A

    The German polarimetric C-band weather radar Poldirad (Polarization Diversity Radar) was deployed for the international field campaign EUREC4A (Elucidating the role of clouds-circulation coupling in climate) on the island of Barbados where it was operated from February until August 2020. Focus of the installation was monitoring clouds and precipitation in the trade wind region east of Barbados. Different scanning modes were used with a temporal sequence of 5 min and a maximum range of 375 km. In addition to built-in quality control performed by the radar signal processor, it was found that the copoloar correlation coefficient rhoHV can be used to remove contamination of radar products by sea clutter. Radar images were available in real time for all campaign participants and aboard research aircraft. Examples of mesoscale precipitation patterns, rain rate accumulation, diurnal cycle, and vertical distribution are given to show the potential of the radar measurements for further studies on the life cycle of precipitating shallow cumulus clouds and other related aspects.
