    Effect of variety and planting density on weed dynamics and yield performance of transplant Aman rice

    The experiment was conducted at the Agronomy Field Laboratory, Bangladesh Agricultural University, Mymensingh during Aman season from June to November 2016 to find out the effect of variety and planting density on weed dynamics and yield performance of transplant Aman rice. The experiment consisted of four varieties viz. Binadhan-7, BR25, BRRI dhan56 and BRRI dhan62 and four planting density viz. 25 cm × 15 cm, 25 cm × 10 cm, 20 cm × 15 cm and 20 cm × 10 cm. The experiment was laid out in a randomized complete block design with three replications. Variety exerted significant effect on weed density and dry weight at different days after transplanting (DAT). The lowest weed density and dry weight were observed in BR25 and the highest ones were observed in Binadhan-7. Weed population was not significantly affected by planting density while weed dry weight was significantly affected and closer spacing produce the lowest weed dry weight. Yield and yield contributing characters of transplant Aman rice were significantly influenced by variety and planting density. BR25 showed produce the highest plant height (157.9 cm), panicle length (24.94 cm), grains panicle(103.10), sterile spikelets panicle−1 (29.36), grain yield (4.30 t ha−1) and straw yield (8.99 t ha−1) while BRRI dhan62 the highest number of total tillers hill−1 (14.75), effective tillers hill−1 (11.62), and non-effective tillers hill−1 (3.10). The highest 1000-grain weight (25.21g) was found in the variety Binadhan-7and the highest harvest index (53.50%) was obtained from BRRI dhan56. In case of planting density, the highest value of plant height (124.27 cm), total tillers hill−1 (13.53), effective tillers hill−1 (11.20), non-effective tillers hill−1 (2.32), panicle length (22.59 cm), grain yield (4.17 t ha−1) and straw yield (5.75 t ha−1) were obtained from 25 cm × 15 cm spacing. On the other hand, the highest number of grains panicle−1 (84.23) and harvest index (45.18 %) were obtained from 25 cm × 10 cm spacing, heaviest 1000-grain weight (23.83 g) from 20 cm × 15 cm spacing. Experimental results indicated that BR25 grown under 25 cm × 15 cm planting density appeared to be the best in order to get maximum grain yield as well as reducing weed infestation

    ‘B-M Model’ for Farmers’ Knowledge Management in Increasing Rice Production

    Quantifying knowledge on agriculture can have many benefits to stakeholders. While many knowledge-based systems exist in modern days for farmers’ decision support, specific models are lacking on how knowledge traits can impact on agricultural production systems. This study employed modelling technique, supported by field data, to provide a clear understanding and quantifying how knowledge management in production practices can contribute to rice productivity in the environmentally stressed southwest Bangladesh. This research accounted for ‘Boro’ rice as the target crop and ‘BRRI dhan28’ as the test variety. The ‘B-M Model’ was developed following the principle and procedure from published literature, ‘brainstorming’ and data from field surveys. Three knowledge management traits (KMT) were defined and quantified as the inputs of the model. Those are: self-experience and observation (SEO), extension advisory services (EAS) and accessed information sources (AIS). The yield influencing process (YIP), the intermediate state variable of the model, was deduced by accounting for the two dominant agronomic practices, seedling age for transplanting and triple superphosphate (TSP) application. ‘Knowledge drives farmers’ practice change which in turn influences yield’ was composed as the theoretical framework of the ‘B-M Model’. The model performed strongly against an independently collected field data set. Across the 180 farmers’ data, the average relative rice yield (RRY) predicted by the model (0.705) and observed in the field (0.716) was close (root mean squared deviation (RMSD) = 0.018). The difference between predicted and observed RRY was not statistically different (LSD = 0.03), indicating the model fully captured the field data. A regression of predicted and observed RRY explained 96% variance in observation, further proving the model’s strength in estimating RRY in a wider range of farmers’ rice yield. In a normative analysis, the practicality and usefulness of the model to stakeholders were simulated for the understanding of how much achievable yield could be expected by changing farmers’ knowledge pool (the sum of three KMT) on rice production practices, and at what combination(s) of KMT to be considered at strategic hierarchy to materialize a targeted achievable yield. To the best of the knowledge, a model quantifying rice yield in relation to knowledge management trait does not exist in literature. Upon successful testing under diverse yield scenarios using multiple and sophisticated statistical tools that enhanced the credibility of the model, it is concluded that the model has the potential to be used for identifying quantitative pathways of farmers’ knowledge acquisition for practice change leading to improved productivity of rice in the southwest region of Bangladesh.</jats:p

    Voltage Gated Calcium Channels Negatively Regulate Protective Immunity to Mycobacterium tuberculosis

    Mycobacterium tuberculosis modulates levels and activity of key intracellular second messengers to evade protective immune responses. Calcium release from voltage gated calcium channels (VGCC) regulates immune responses to pathogens. In this study, we investigated the roles of VGCC in regulating protective immunity to mycobacteria in vitro and in vivo. Inhibiting L-type or R-type VGCC in dendritic cells (DCs) either using antibodies or by siRNA increased calcium influx in an inositol 1,4,5-phosphate and calcium release calcium activated channel dependent mechanism that resulted in increased expression of genes favoring pro-inflammatory responses. Further, VGCC-blocked DCs activated T cells that in turn mediated killing of M. tuberculosis inside macrophages. Likewise, inhibiting VGCC in infected macrophages and PBMCs induced calcium influx, upregulated the expression of pro-inflammatory genes and resulted in enhanced killing of intracellular M. tuberculosis. Importantly, compared to healthy controls, PBMCs of tuberculosis patients expressed higher levels of both VGCC, which were significantly reduced following chemotherapy. Finally, blocking VGCC in vivo in M. tuberculosis infected mice using specific antibodies increased intracellular calcium and significantly reduced bacterial loads. These results indicate that L-type and R-type VGCC play a negative role in M. tuberculosis infection by regulating calcium mobilization in cells that determine protective immunity

    Laparoscopy in management of appendicitis in high-, middle-, and low-income countries: a multicenter, prospective, cohort study.

    BACKGROUND: Appendicitis is the most common abdominal surgical emergency worldwide. Differences between high- and low-income settings in the availability of laparoscopic appendectomy, alternative management choices, and outcomes are poorly described. The aim was to identify variation in surgical management and outcomes of appendicitis within low-, middle-, and high-Human Development Index (HDI) countries worldwide. METHODS: This is a multicenter, international prospective cohort study. Consecutive sampling of patients undergoing emergency appendectomy over 6 months was conducted. Follow-up lasted 30 days. RESULTS: 4546 patients from 52 countries underwent appendectomy (2499 high-, 1540 middle-, and 507 low-HDI groups). Surgical site infection (SSI) rates were higher in low-HDI (OR 2.57, 95% CI 1.33-4.99, p = 0.005) but not middle-HDI countries (OR 1.38, 95% CI 0.76-2.52, p = 0.291), compared with high-HDI countries after adjustment. A laparoscopic approach was common in high-HDI countries (1693/2499, 67.7%), but infrequent in low-HDI (41/507, 8.1%) and middle-HDI (132/1540, 8.6%) groups. After accounting for case-mix, laparoscopy was still associated with fewer overall complications (OR 0.55, 95% CI 0.42-0.71, p < 0.001) and SSIs (OR 0.22, 95% CI 0.14-0.33, p < 0.001). In propensity-score matched groups within low-/middle-HDI countries, laparoscopy was still associated with fewer overall complications (OR 0.23 95% CI 0.11-0.44) and SSI (OR 0.21 95% CI 0.09-0.45). CONCLUSION: A laparoscopic approach is associated with better outcomes and availability appears to differ by country HDI. Despite the profound clinical, operational, and financial barriers to its widespread introduction, laparoscopy could significantly improve outcomes for patients in low-resource environments.     Pooled analysis of WHO Surgical Safety Checklist use and mortality after emergency laparotomy

    Background The World Health Organization (WHO) Surgical Safety Checklist has fostered safe practice for 10 years, yet its place in emergency surgery has not been assessed on a global scale. The aim of this study was to evaluate reported checklist use in emergency settings and examine the relationship with perioperative mortality in patients who had emergency laparotomy. Methods In two multinational cohort studies, adults undergoing emergency laparotomy were compared with those having elective gastrointestinal surgery. Relationships between reported checklist use and mortality were determined using multivariable logistic regression and bootstrapped simulation. Results Of 12 296 patients included from 76 countries, 4843 underwent emergency laparotomy. After adjusting for patient and disease factors, checklist use before emergency laparotomy was more common in countries with a high Human Development Index (HDI) (2455 of 2741, 89.6 per cent) compared with that in countries with a middle (753 of 1242, 60.6 per cent; odds ratio (OR) 0.17, 95 per cent c.i. 0.14 to 0.21, P <0001) or low (363 of 860, 422 per cent; OR 008, 007 to 010, P <0.001) HDI. Checklist use was less common in elective surgery than for emergency laparotomy in high-HDI countries (risk difference -94 (95 per cent c.i. -11.9 to -6.9) per cent; P <0001), but the relationship was reversed in low-HDI countries (+121 (+7.0 to +173) per cent; P <0001). In multivariable models, checklist use was associated with a lower 30-day perioperative mortality (OR 0.60, 0.50 to 073; P <0.001). The greatest absolute benefit was seen for emergency surgery in low- and middle-HDI countries. Conclusion Checklist use in emergency laparotomy was associated with a significantly lower perioperative mortality rate. Checklist use in low-HDI countries was half that in high-HDI countries.Peer reviewe

    Global variation in anastomosis and end colostomy formation following left-sided colorectal resection

    Background End colostomy rates following colorectal resection vary across institutions in high-income settings, being influenced by patient, disease, surgeon and system factors. This study aimed to assess global variation in end colostomy rates after left-sided colorectal resection. Methods This study comprised an analysis of GlobalSurg-1 and -2 international, prospective, observational cohort studies (2014, 2016), including consecutive adult patients undergoing elective or emergency left-sided colorectal resection within discrete 2-week windows. Countries were grouped into high-, middle- and low-income tertiles according to the United Nations Human Development Index (HDI). Factors associated with colostomy formation versus primary anastomosis were explored using a multilevel, multivariable logistic regression model. Results In total, 1635 patients from 242 hospitals in 57 countries undergoing left-sided colorectal resection were included: 113 (6·9 per cent) from low-HDI, 254 (15·5 per cent) from middle-HDI and 1268 (77·6 per cent) from high-HDI countries. There was a higher proportion of patients with perforated disease (57·5, 40·9 and 35·4 per cent; P < 0·001) and subsequent use of end colostomy (52·2, 24·8 and 18·9 per cent; P < 0·001) in low- compared with middle- and high-HDI settings. The association with colostomy use in low-HDI settings persisted (odds ratio (OR) 3·20, 95 per cent c.i. 1·35 to 7·57; P = 0·008) after risk adjustment for malignant disease (OR 2·34, 1·65 to 3·32; P < 0·001), emergency surgery (OR 4·08, 2·73 to 6·10; P < 0·001), time to operation at least 48 h (OR 1·99, 1·28 to 3·09; P = 0·002) and disease perforation (OR 4·00, 2·81 to 5·69; P < 0·001). Conclusion Global differences existed in the proportion of patients receiving end stomas after left-sided colorectal resection based on income, which went beyond case mix alone

    Quantitative PCR analysis of abundance of airborne propagules of Leptosphaeria species in air samples from different regions of Poland

    When airborne propagules of Leptosphaeria maculans and L. biglobosa were collected in Poland at three ecologically different sites from 1 September to 30 November in 2004 to 2008, using a Hirst-type seven-day volumetric spore trap, there were fluctuations in timing of ascospore release and diverse ratios between airborne propagules of both species depending on season, field location and weather conditions. The detection was done using the microscope as well as quantitative PCR with species-specific primers targeted against fragments of β-tubulin genes and quantified with a dual-labelled fluorescent probe approach. This detection chemistry is described for the first time for L. maculans and L. biglobosa. Its advantage over the previous ITS-based SYBR-Green chemistry resides in improved sensitivity and the virtual absence of false positives in the detection of these fungi. There were significant, positive correlations between data obtained using visual assessment of ascospore numbers and DNA concentrations that were measured by qPCR. Climatic differences between the oilseed rape growing regions could have significantly affected the biological processes of pseudothecial maturation and ascospore development of the pathogens. The data suggest that regular rain events of intermediate intensity recorded in the Maritime region favoured the maturation of the pathogen more than the drier weather recorded in the Silesia or Pomerania regions. It was observed that the number of rainy days was of greater importance than the cumulative rainfall to obtain the generative sporulation of the pathogen. Accurate detection of airborne inoculum of pathogenic Leptosphaeria spp. facilitates improved targeting of disease management decisions for oilseed rape protection against phoma stem canker and stem necrosis diseases.Peer reviewe
