    The last glacial maximum Balearic Abyssal Plain megabed revisited

    Megabeds are thick sedimentary layers extending over thousands square kilometres in deep sea basins and are thought to result from large slope failures triggered by major external events. Such deposits have been found in at least three areas of the Mediterranean Sea. Although their discovery dates back to the early 1980s, many questions remain, concerning their initiation, source area, extent, and the nature of their emplacement. One of the largest previously documented megabeds was emplaced during the Last Glacial Maximum across the Balearic Abyssal Plain with a thickness of 8-10 m in water depths of up to 2800 m. New 3.5 kHz sub-bottom profiles and sediment cores provide greater constraint on the lateral variability of the megabed and allow to map it beyond previous estimates, with a revised areal extent up to 90,000-100,000 km2. Megabed terminations show gradual pinch-out to the West and an abrupt eastward termination against the Sardinia steep margin. The megabed presents both in seismic profiles and in sediment cores a tripartite subdivision likely corresponding to changes in flow regimes across the basin with a central area of sandy facies and erosional base oriented NNE-SSW allowing renewed discussions about sources and trigger of the megabed

    New catechol-containing chemical entities have been investigated as matrix metalloproteinase inhibitors as well as antioxidant molecules. The combination of the two properties could represent a useful feature due to the potential application in all the pathological processes characterized by increased proteolytic activity and radical oxygen species (ROS) production, such as inflammation and photoaging. A series of catechol-based molecules were synthesized and tested for both proteolytic and oxidative inhibitory activity, and the detailed binding mode was assessed by crystal structure determination of the complex between a catechol derivative and the matrix metalloproteinase-8.     SMUG1 is able to excise uracil from immunoglobulin genes: insight into mutation versus repair

    Mammals harbour multiple enzymes capable of excising uracil from DNA, although their distinct physiological roles remain uncertain. One of them (UNG) plays a critical role in antibody gene diversification, as UNG deficiency alone is sufficient to perturb the process. Here, we show this unique requirement for UNG does not reflect the fact that other glycosylases are unable to access the U:G lesion. SMUG1, if overexpressed, can partially substitute for UNG to assist antibody diversification as judged by its effect on somatic hypermutation patterns (in both DT40 B cells and mice) as well as a restoration of isotype switching in SMUG-transgenic msh2(−/−)ung(−/−) mice. However, SMUG1 plays little natural role in antibody diversification because (i) it is diminishingly expressed during B-cell activation and (ii) even if overexpressed, SMUG1 more appears to favour conventional repair of the uracil lesion than assist diversification. The distinction between UNG and overexpressed SMUG1 regarding the balance between antibody diversification and non-mutagenic repair of the U:G lesion could reflect the association of UNG (but not SMUG1) with sites of DNA replication
