Carbon source feeding strategies for recombinant protein expression in Pichia pastoris and Pichia methanolica
Pichia pastoris and Pichia methanolica have been used as expression systems for the production of recombinant protein. The main problems of the production are the slow hierarchic consumption of ethanol and acetate which cause toxicity problems due to methanol accumulation when this surpasses 0.5 gl-1. In some cases, the laboratory scale cultures does not show the methanol accumulation problems because the cells are usually washed before changing the depleted carbon source culture media, by a fresh one, supplemented with methanol; a strategy that is clearly inapplicable in the industrial productions. Other authors use to feed the methanol before the depletion of glycerol or Dglucose, but this practice does not guaranty the ethanol and acetate fast consumption; leading to methanol accumulation and toxicity problems. It was concluded that pre-induction stage strategy should be studied in detail and that it is very important to start the induction stage with a concentration of biomass as great as possible. On the other side, it is essential that there should be a monitoring of ethanol and acetate until reaching non-toxic methanol stable-concentration and these conditions should be maintained till the end of the process
Review of magnetic nanostructures grown by focused electron beam induced deposition (FEBID)
We review the current status of the use of focused electron beam induced deposition (FEBID) for the growth of magnetic nanostructures. This technique relies on the local dissociation of a precursor gas by means of an electron beam. The most promising results have been obtained using the Co₂(CO)₈ precursor, where the Co content in the grown nanodeposited material can be tailored up to more than 95%. Functional behaviour of these Co nanodeposits has been observed in applications such as arrays of magnetic dots for information storage and catalytic growth, magnetic tips for scanning probe microscopes, nano-Hall sensors for bead detection, nano-actuated magnetomechanical systems and nanowires for domain-wall manipulation. The review also covers interesting results observed in Fe-based and alloyed nanodeposits. Nanoscale chemical and structural study of Co-based FEBID structures by STEM-EELS and HRTEM
Nanolithography techniques in a scanning electron microscope/focused ion beam are very attractive tools for a number of synthetic processes, including the fabrication of ferromagnetic nano-objects, with potential applications in magnetic storage or magnetic sensing. One of the most versatile techniques is the focused electron beam induced deposition, an efficient method for the production of magnetic structures highly resolved at the nanometric scale. In this work, this method has been applied to the controlled growth of magnetic nanostructures using Co₂(CO)₈. The chemical and structural properties of these deposits have been studied by electron energy loss spectroscopy and high-resolution transmission electron microscopy at the nanometric scale. The effectiveness of a health promotion with group intervention by clinical trial. Study protocol
<p>Abstract</p> <p>Background</p> <p>The promotion of health and the interventions in community health continue to be one of the pending subjects of our health system. The most prevalent health problems (cardiovascular diseases, cancer, diabetes...) are for the most part related to life habits. We propose a holistic and integral approach as the best option for tackling behavior and its determinants. The research team has elaborated the necessary educational material to realize group teaching, which we call "Health Workshops". The goal of the present study is to evaluate the effectiveness of these Health Workshops in the following terms: Health Related Quality of Life (HRQOL), incorporate and maintain a balanced diet, do physical activity regularly, maintain risk factors such as tension, weight, cholesterol within normal limits and diminish cardiovascular risk.</p> <p>Methods/Design</p> <p>Controlled and random clinical testing, comparing a group of persons who have participated in the Health Workshops with a control group of similar characteristics who have not participated in the Health Workshops.</p> <p>Field of study: the research is being done in Health Centers of the city of Barcelona, Spain.</p> <p>Population studied: The group is composed of 108 persons that are actually doing the Health Workshops, and 108 that are not and form the control group. They are assigned at random to one group or the other.</p> <p>Data Analysis: With Student's t-distribution test to compare the differences between numerical variables or their non parametric equivalent if the variable does not comply with the criteria of normality. (Kolmogorov-Smirnof test). Chi-square test to compare the differences between categorical variables and the Logistic Regression Model to analyze different meaningful variables by dichotomous analysis related to the intervention.</p> <p>Discussion</p> <p>The Health Workshop proposed in the present study constitutes an innovative approach in health promotion, placing the emphasis on the person's self responsibility for his/her own health.</p> <p>The rhythm of a weekly session during 8 weeks with recommended activities to put into practice, as well as the support of the group is an opportunity to incorporate healthy habits and make a commitment to self-care. Saturation of an Intra-Gene Pool Linkage Map: Towards a Unified Consensus Linkage Map for Fine Mapping and Synteny Analysis in Common Bean
Map-based cloning and fine mapping to find genes of interest and marker assisted selection (MAS) requires good genetic maps with reproducible markers. In this study, we saturated the linkage map of the intra-gene pool population of common bean DOR364×BAT477 (DB) by evaluating 2,706 molecular markers including SSR, SNP, and gene-based markers. On average the polymorphism rate was 7.7% due to the narrow genetic base between the parents. The DB linkage map consisted of 291 markers with a total map length of 1,788 cM. A consensus map was built using the core mapping populations derived from inter-gene pool crosses: DOR364×G19833 (DG) and BAT93×JALO EEP558 (BJ). The consensus map consisted of a total of 1,010 markers mapped, with a total map length of 2,041 cM across 11 linkage groups. On average, each linkage group on the consensus map contained 91 markers of which 83% were single copy markers. Finally, a synteny analysis was carried out using our highly saturated consensus maps compared with the soybean pseudo-chromosome assembly. A total of 772 marker sequences were compared with the soybean genome. A total of 44 syntenic blocks were identified. The linkage group Pv6 presented the most diverse pattern of synteny with seven syntenic blocks, and Pv9 showed the most consistent relations with soybean with just two syntenic blocks. Additionally, a co-linear analysis using common bean transcript map information against soybean coding sequences (CDS) revealed the relationship with 787 soybean genes. The common bean consensus map has allowed us to map a larger number of markers, to obtain a more complete coverage of the common bean genome. Our results, combined with synteny relationships provide tools to increase marker density in selected genomic regions to identify closely linked polymorphic markers for indirect selection, fine mapping or for positional cloning
Measurement of the Bottom-Strange Meson Mixing Phase in the Full CDF Data Set
We report a measurement of the bottom-strange meson mixing phase \beta_s
using the time evolution of B0_s -> J/\psi (->\mu+\mu-) \phi (-> K+ K-) decays
in which the quark-flavor content of the bottom-strange meson is identified at
production. This measurement uses the full data set of proton-antiproton
collisions at sqrt(s)= 1.96 TeV collected by the Collider Detector experiment
at the Fermilab Tevatron, corresponding to 9.6 fb-1 of integrated luminosity.
We report confidence regions in the two-dimensional space of \beta_s and the
B0_s decay-width difference \Delta\Gamma_s, and measure \beta_s in [-\pi/2,
-1.51] U [-0.06, 0.30] U [1.26, \pi/2] at the 68% confidence level, in
agreement with the standard model expectation. Assuming the standard model
value of \beta_s, we also determine \Delta\Gamma_s = 0.068 +- 0.026 (stat) +-
0.009 (syst) ps-1 and the mean B0_s lifetime, \tau_s = 1.528 +- 0.019 (stat) +-
0.009 (syst) ps, which are consistent and competitive with determinations by

The macroecology of chemical communication in lizards: do climatic factors drive the evolution of signalling glands?
Chemical communication plays a pivotal role in shaping sexual and ecological interactions among animals. In lizards, fundamental mechanisms of sexual selection such as female mate choice have rarely been shown to be influenced by quantitative phenotypic traits (e.g., ornaments), while chemical signals have been found to potentially influence multiple forms of sexual and social interactions, including mate choice and territoriality. Chemical signals in lizards are secreted by glands primarily located on the edge of the cloacae (precloacal glands, PG) and thighs (femoral glands), and whose interspecific and interclade number ranges from 0 to >100. However, elucidating the factors underlying the evolution of such remarkable variation remains an elusive endeavour. Competing hypotheses suggest a dominant role for phylogenetic conservatism (i.e., species within clades share similar numbers of glands) or for natural selection (i.e., their adaptive diversification results in deviating numbers of glands from ancestors). Using the prolific Liolaemus lizard radiation from South America (where precloacal glands vary from 0-14), we present one of the largest-scale tests of both hypotheses to date. Based on climatic and phylogenetic modelling, we show a clear role for both phylogenetic inertia and adaptation underlying gland variation: (i) solar radiation, net primary productivity, topographic heterogeneity and precipitation range have a significant effect on number of PG variation, (ii) humid and cold environments tend to concentrate species with a higher number of glands, (iii) there is a strong phylogenetic signal that tends to conserve the number of PG within clades. Collectively, our study confirms that the inertia of niche conservatism can be broken down by the need of species facing different selection regimes to adjust their glands to suit the demands of their specific environments
ASSESSMENT OF THE PRESENCE OF Toxocara EGGS IN SOILS OF AN ARID AREA IN CENTRAL-WESTERN ARGENTINA
With the aim of studying the contamination of soils with eggs of Toxocara spp. in an arid area in the central-western region of Argentina, 76 soil samples were collected from 18 towns belonging to six provinces of central-western Argentina. They were processed by the centrifugal flotation method. No eggs of Toxocara spp. were found. It can be concluded that the negative results are directly related to the characteristics of the environment and climate present in the studied area. The finding of eggs in soils depends on several factors: the presence of canine or feline feces, the hygienic behavior of pet owners, the presence of stray animals without veterinary supervision, the weather and environmental conditions, and laboratory techniques used; and all these circumstances must be considered when comparing the results found in different geographical regions. In order to accurately define the importance of public spaces in the transmission of infection to humans, it is important to consider the role of backyards or green spaces around housing in small towns, where the population is not used to walking pets in public spaces, and in such cases a significant fraction of the population may acquire the infection within households
Movilidad urbana y pobreza: Aprendizajes de Medellín y Soacha, Colombia
Este libro es un análisis de la experiencia de Medellín, la segunda ciudad de Colombia, en intentar reducir la pobreza e integrar en el tejido urbano barrios marginalizados y estigmatizados durante muchos años por la pobreza y la violencia. En particular, examina el impacto de dos cables aéreos que conectan densos barrios en colinas empinadas con el resto de la ciudad, y un programa de mejoramiento urbano asociado a estos. También contrasta la experiencia exitosa de Medellín con la de Soacha, un municipio adyacente a Bogotá, la capital colombiana, donde se ha propuesto un cable aéreo como medio de conexión de dos barrios en colina con una vía arteria. El contraste entre un municipio bien gestionado y abundante en recursos como Medellín con un municipio denso, homogéneamente pobre e institucionalmente débil como Soacha ofrece aprendizajes muy valiosos para otras ciudades en América Latina y otras regiones del mundo. La mayor parte de los capítulos se apoyan en un proyecto de investigación de dos años coordinado por la Development Planning Unit, University College London (UCL), en asocio con la Universidad Nacional de Colombia (sede Medellín) y la Universidad de los Andes de Bogotá. Además de estudios de caso detallados de Medellín y Soacha, el libro agrupa también casos en América Latina en donde se han construido o se han propuesto cables aéreos en barrios de bajos ingresos, como Caracas y Rio de Janeiro
Urban Mobility and Poverty: Lessons from Medellin and Soacha, Colombia
This book examines the experience of Medellín, Colombia's second largest city, in seeking to reduce poverty and integrate large marginalised areas, marked by years of severe poverty and violence, into the urban fabric. It pays particular attention to the impact of two aerial cable-cars connecting high density hilly neighbourhoods with the rest of the city, and an associated urban upgrading programme. It also contrasts Medellín’s successful experience with that of Soacha, a municipality adjacent to Bogotá, Colombia’s capital city, where an aerial cable-car has been proposed as a means of linking two low-income hilly neighbourhoods with a main arterial road. The contrast between a well-resourced, well-managed municipality like Medellín with a dense and homogenously poor and institutionally weak municipality like Soacha offers valuable lessons to other cities in Latin America and elsewhere. Contributions draw from a two-year research project coordinated by the Development Planning Unit, University College London (UCL), in conjunction with Universidad Nacional de Colombia (Medellín campus) and Universidad de los Andes in Bogotá. In addition to the detailed case studies of Medellín and Soacha, the book also brings together cases in Latin America where aerial cable-cars have either been built or proposed in low-income neighbourhoods, including Caracas and Rio de Janeiro
