    Molecularly imprinted polymers for the detection of caffeine in water

    This work investigates the potential of Molecularly Imprinted Polymers (MIPs) for the detection of caffeine in water samples. Different polymerization techniques were explored; namely free-radical bulk, miniemulsion and precipitation polymerization. Building the adsorption isotherms allowed determining the rebinding capacities of the resulting polymers (binding constant and number of sites). Using the nanoparticles prepared by precipitation polymerization in acetonitrile, new analytical techniques were developed. The latex was analyzed directly by High Performance Liquid Chromatography allowing fast template quantification in water samples. In addition, the nanoparticles were coated on a 5MHz microbalance quartz crystal. The bi-Langmuir isotherm allowed computing the mass uptakes at the sensor\u27s surface and suggested that as little as 0.1 ng of caffeine could be detected

    Meiotic silencing in pigs: A case study in a translocated azoospermic boar

    International audienceCarriers of balanced constitutional reciprocal translocations usually present a normal phenotype, but often show reproductive disorders. For the first time in pigs, we analyzed the meiotic process of an autosome–autosome translocation associated with azoospermia. Meiotic process analysis revealed the presence of unpaired autosomal segments with histone ÎłH2AX accumulation sometimes associated with the XY body. Additionally, ÎłH2AX signals were observed on apparently synapsed autosomes other than the SSC1 or SSC15, as previously observed in Ataxia with oculomotor apraxia type 2 patients or knock-out mice for the Senataxin gene. Gene expression showed a downregulation of genes selected on chromosomes 1 and 15, but no upregulation of SSCX genes. We hypothesized that the total meiotic arrest observed in this boar might be due to the silencing of crucial autosomal genes by the mechanism referred to as meiotic silencing of unsynapsed chromatin (MSUC).Les porteurs de translocations rĂ©ciproques constitutionnelles Ă©quilibrĂ©es prĂ©sentent gĂ©nĂ©ralement un phĂ©notype normal, mais prĂ©sentent souvent des troubles de la reproduction. Pour la premiĂšre fois chez le porc, nous avons analysĂ© le processus mĂ©iotique d'une translocation autosome-autosome associĂ©e Ă  une azoospermie. L'analyse du processus mĂ©iotique a rĂ©vĂ©lĂ© la prĂ©sence de segments autosomiques non appariĂ©s avec une accumulation d'histone ÎłH2AX parfois associĂ©e Ă  la vĂ©sicule sexuelle. De plus, des signaux ÎłH2AX ont Ă©tĂ© observĂ©s sur des autosomes apparemment appariĂ©s autres que le SSC1 ou le SSC15, comme cela avait Ă©tĂ© prĂ©cĂ©demment observĂ© chez des patients atteints d'Ataxie avec apraxie oculomotrice de type 2 ou chez des souris knock-out pour le gĂšne de la senataxine. L'expression gĂ©nique a montrĂ© une rĂ©gulation nĂ©gative des gĂšnes sĂ©lectionnĂ©s sur les chromosomes 1 et 15, mais aucune rĂ©gulation positive des gĂšnes du SSCX. Nous avons Ă©mis l'hypothĂšse que l'arrĂȘt mĂ©iotique total observĂ© chez ce verrat pourrait ĂȘtre dĂ» Ă  la rĂ©pression de gĂšnes autosomiques cruciaux par le mĂ©canisme appelĂ© " meiotic silencing of unsynapsed chromatin" (MSUC)
