A Method for Predicting the Effects of Specimen Geometry and Loading Condition on Fatigue Strength
Specimen geometry and loading condition usually have a great influence on the fatigue strength of metallic materials, which is an important issue in evaluating the reliability of component parts. In this paper, a rotating bending fatigue test is performed at first on an hourglass specimen and a notch specimen of a high strength titanium alloy. Experimental results indicate that, in terms of local stress, the notch specimen endures higher fatigue strength in comparison with the hourglass specimen due to its relatively smaller control volume. Then, a probabilistic control volume method is proposed for correlating the effects of specimen geometry and loading condition on the fatigue strength based on Weibull distribution and the concept of control volume. A simple formula is obtained for the fatigue strength in relation to control volumes, in which the parameter is the shape parameter of Weibull distribution of fatigue strength. The predicted results are in good agreement with the present experimental data for high strength titanium alloy and the data for the high strength steel and the full scale EA4T axle in the literature
Effects of Thioglycolic Acid on Parthenogenetic Activation of Xenopus Oocytes
BACKGROUND: Existing in Permanent-wave solutions (PWS), thioglycolic acid (TGA) is widely used in hairdressing industry for its contribution to hair styling. However, the toxicity of TGA, especially its reproductive toxicity, gradually calls the attention of more and more researchers. METHOD: In this work, xenopus oocytes were pretreated with different concentration of TGA, and then activated by calcium ionophore A23187. During culture, the oocytes activation rates were taken note at different time after adding calcium ionophore A23187. At the end of the culture period, the nuclear status was detected under confocal microscope. In addition, some other samples were collected for Western-Blotting analysis. RESULT: TGA significantly inhibited the oocytes activation rate and pronuclear formation. It may be resulted from the inhibition of the degradation of p-ERK1, Mos and CyclinB2. CONCLUSION: TGA inhibits in vitro parthenogenetic activation of xenopus oocytes with inhibited the degradation of proteins involved in mitogenic-activated protein kinase (MAPK) and maturation-promoting factor (MPF) pathways
Electronic Origin of High Temperature Superconductivity in Single-Layer FeSe Superconductor
The latest discovery of high temperature superconductivity signature in
single-layer FeSe is significant because it is possible to break the
superconducting critical temperature ceiling (maximum Tc~55 K) that has been
stagnant since the discovery of Fe-based superconductivity in 2008. It also
blows the superconductivity community by surprise because such a high Tc is
unexpected in FeSe system with the bulk FeSe exhibiting a Tc at only 8 K at
ambient pressure which can be enhanced to 38 K under high pressure. The Tc is
still unusually high even considering the newly-discovered intercalated FeSe
system A_xFe_{2-y}Se_2 (A=K, Cs, Rb and Tl) with a Tc at 32 K at ambient
pressure and possible Tc near 48 K under high pressure. Particularly
interesting is that such a high temperature superconductivity occurs in a
single-layer FeSe system that is considered as a key building block of the
Fe-based superconductors. Understanding the origin of high temperature
superconductivity in such a strictly two-dimensional FeSe system is crucial to
understanding the superconductivity mechanism in Fe-based superconductors in
particular, and providing key insights on how to achieve high temperature
superconductivity in general. Here we report distinct electronic structure
associated with the single-layer FeSe superconductor. Its Fermi surface
topology is different from other Fe-based superconductors; it consists only of
electron pockets near the zone corner without indication of any Fermi surface
around the zone center. Our observation of large and nearly isotropic
superconducting gap in this strictly two-dimensional system rules out existence
of node in the superconducting gap. These results have provided an unambiguous
case that such a unique electronic structure is favorable for realizing high
temperature superconductivity
Functional ectodomain of the hemagglutinin-neuraminidase protein is expressed in transgenic tobacco cells as a candidate vaccine against Newcastle disease virus.
Recently, the use of plants for the production of recombinant proteins has been well demonstrated with promising outcomes. In this study, an efficient Nicotiana tabacum L. cv. Bright Yellow 2 (BY-2) cells system expressing the ectodomain of hemagglutinin-neuraminidase (eHN) protein from Newcastle disease virus (NDV) strain AF2240 was established. Transgenic tobacco BY-2 cell cultures expressing the immunogenic eHN protein were generated and the translation efficiency of eHN protein was enhanced using the 5âČ-untranslated region of Nicotiana tabacum alcohol dehydrogenase gene (NtADH 5âČ-UTR) under the control of strong cauliflower mosaic virus (CaMV 35S) promoter. Transgenic lines verified by real-time PCR showed high level of eHN mRNA transcripts and immunoblotting confirmed the presence of 66 kD eHN protein. The eHN protein was stably produced in an average of 0.2â0.4 % total soluble protein. Green fluorescent protein-tagged eHN protein was expressed and localized at the cytosol of BY-2 cell. All mice receiving purified eHN protein from transgenic tobacco BY-2 cells produced specific anti-NDV antibodies. We concluded that plant made eHN elicit immune response and can serve as candidate vaccine against NDV
Monsoon versus Uplift in Southwestern ChinaâLate Pliocene Climate in Yuanmou Basin, Yunnan
Yuanmou Basin of Yunnan, SW China, is a famous locality with hominids, hominoids, mammals and plant fossils. Based on the published megaflora and palynoflora data from Yuanmou Basin, the climate of Late Pliocene is reconstructed using the Coexistence Approach. The results indicate a warm and humid subtropical climate with a mean annual temperature of ca. 16â17°C and a mean annual precipitation of ca. 1500â1600 mm in the Late Pliocene rather than a dry, hot climate today, which may be due to the local tectonic change and gradual intensification of India monsoon. The comparison of Late Pliocene climate in Eryuan, Yangyi, Longling, and Yuanmou Basin of Yunnan Province suggests that the mean annual temperatures generally show a latitudinal gradient and fit well with their geographic position, while the mean annual precipitations seem to be related to the different geometries of the valleys under the same monsoon system
Structural Characterization of Mesoporous Silica Nanofibers Synthesized Within Porous Alumina Membranes
Mesoporous silica nanofibers were synthesized within the pores of the anodic aluminum oxide template using a simple solâgel method. Transmission electron microscopy investigation indicated that the concentration of the structure-directing agent (EO20PO70EO20) had a significant impact on the mesostructure of mesoporous silica nanofibers. Samples with alignment of nanochannels along the axis of mesoporous silica nanofibers could be formed under the P123 concentration of 0.15 mg/mL. When the P123 concentration increased to 0.3 mg/mL, samples with a circular lamellar mesostructure could be obtained. The mechanism for the effect of the P123 concentration on the mesostructure of mesoporous silica nanofibres was proposed and discussed
Soluble Fas ligand released by colon adenocarcinoma cells induces host lymphocyte apoptosis: an active mode of immune evasion in colon cancer
Expression of membrane-bound Fas ligand (mFasL) on colon cancer cells serves as a potential mechanism to inhibit host immune function by inducing apoptosis of host lymphocytes. Membrane-bound FasL can be cleaved and released as a soluble mediator (sFasL), which may spread the apoptosis induction effect. Our study examined whether colon adenocarcinoma cells release sFasL, and induce apoptosis of host lymphocytes without direct cellâcell contact. In 12 consecutive patients with colon adenocarcinoma mFasL was identified in the tumours, sFasL was measured in the sera and apoptosis identified in tumour-infiltrating and peripheral blood lymphocytes. To analyse the function of sFasL, colon cancer cells were primarily cultured; sFasL was isolated from supernatants, measured, incubated with Fas-bearing Jurkat cells, and the resulting apoptosis was analysed. Serum levels of sFasL were significantly elevated in all colon cancer patients with mFasL expression in tumour tissues (n = 8). In these patients, the number of apoptotic lymphocytes was significantly increased within tumour and peripheral blood. Furthermore, sFasL was present in the corresponding supernatants and induced apoptosis of Jurkat cells in a dose-dependent manner. These findings suggest that mFasL-positive colon cancer cells release sFasL, and thus may induce apoptosis of host lymphocytes as a potential mechanism for immune evasion. © 2001 Cancer Research Campaignhttp://www.bjcancer.co
Measurement of the Bottom-Strange Meson Mixing Phase in the Full CDF Data Set
We report a measurement of the bottom-strange meson mixing phase \beta_s
using the time evolution of B0_s -> J/\psi (->\mu+\mu-) \phi (-> K+ K-) decays
in which the quark-flavor content of the bottom-strange meson is identified at
production. This measurement uses the full data set of proton-antiproton
collisions at sqrt(s)= 1.96 TeV collected by the Collider Detector experiment
at the Fermilab Tevatron, corresponding to 9.6 fb-1 of integrated luminosity.
We report confidence regions in the two-dimensional space of \beta_s and the
B0_s decay-width difference \Delta\Gamma_s, and measure \beta_s in [-\pi/2,
-1.51] U [-0.06, 0.30] U [1.26, \pi/2] at the 68% confidence level, in
agreement with the standard model expectation. Assuming the standard model
value of \beta_s, we also determine \Delta\Gamma_s = 0.068 +- 0.026 (stat) +-
0.009 (syst) ps-1 and the mean B0_s lifetime, \tau_s = 1.528 +- 0.019 (stat) +-
0.009 (syst) ps, which are consistent and competitive with determinations by
other experiments.Comment: 8 pages, 2 figures, Phys. Rev. Lett 109, 171802 (2012
