L-Fucose-containing arabinogalactan-protein in radish leaves.
The carbohydrate moieties of arabinogalactan-proteins (AGPs) have β-(1 → 3)-galactan backbones to which side chains of (1 → 6)-linked β-Gal residues are attached through O-6. Some of these side chains are further substituted with other sugars. We investigated the structure of L-Fuc-containing oligosaccharides released from the carbohydrate moieties of a radish leaf AGP by digestion with α-L-arabinofuranosidase, followed by exo-β-(1 → 3)-galactanase. We detected a series of neutral β-(1 → 6)-galactooligosaccharides branching variously at O-3 of the Gal residues, together with corresponding acidic derivatives terminating in 4-O-methyl-GlcA (4-Me-GlcA) or GlcA at the non-reducing terminals. In neutral oligosaccharides with degree of polymerization (dp) mainly higher than 10, L-Fuc groups were attached through L-Ara residues as the sequence, α-L-Fucp-(1 → 2)-α-L-Araf-(1 →. This sequence was verified by isolation of the pentasaccharide α-L-Fuc-(1 → 2)-α-L-Araf-(1 → 3)-β-Gal-(1 → 6)-β-Gal-(1 → 6)-Gal upon digestion of the higher oligosaccharides with endo-β-(1 → 6)-galactanase. By contrast, in lower polymerized (predominantly dp 4) acidic oligosaccharides, L-Fuc groups were attached directly at the non-reducing terminals through α-(1 → 2)-linkages, resulting in the release of the tetrasaccharides, α-L-Fucp-(1 → 2)-β-GlcA-(1 → 6)-β-Gal-(1 → 6)-Gal and α-L-Fucp-(1 → 2)-β-4-Me-GlcA-(1 → 6)-β-Gal-(1 → 6)-Gal. In long acidic oligosaccharides with dp mainly higher than 13, L-Fuc groups localized on branches were attached to the uronic acids directly and/or L-Ara residues as in the neutral oligosaccharides.The authors would like to thank Prof. M. Impact of the 2011 Tohoku Earthquake on the Use of Tidal Flats: A Case Study in Inner Tokyo Bay
Coastal areas have recently been revaluated to understand the relationship between ecosystem services and humans. However, devastation from the 2011 earthquake and tsunami in Japan might have limited the use of ecosystem services. Thus, we investigate the effects of the earthquake and tsunami on the use of tidal flats. We used questionnaires to evaluate changes in the number and anxiety of users for each type of use of the Sanbanze tidal flat in the innermost part of Tokyo Bay. We also conducted a field survey to verify the trends. We found that clamming and walking or jogging were the most common uses in the surveyed area. However, clamming activity decreased after the earthquake as indicated by field observation (73% decrease in spring long holiday season) and questionnaires (64%). Other clamming ground in Tokyo Bay also showed a similar decrease (62%). Many visitors were anxious about the damaged infrastructure. Compared to other types of use such as swimming or total tourism in the tourism statistics, the recovery phase was slower for clamming using the data of other locations. Considering these evidence, the innermost part of Tokyo Bay can be classified as an earthquake-devastated area, from the view point of ecosystem services. Age-dependent motor dysfunction due to neuron-specific disruption of stress-activated protein kinase MKK7.
c-Jun N-terminal kinase (JNK) is a member of the mitogen-activated protein kinase family and controls various physiological processes including apoptosis. A specific upstream activator of JNKs is the mitogen-activated protein kinase kinase 7 (MKK7). It has been reported that MKK7-JNK signaling plays an important regulatory role in neural development, however, post-developmental functions in the nervous system have not been elucidated. In this study, we generated neuron-specific Mkk7 knockout mice (MKK7 cKO), which impaired constitutive activation of JNK in the nervous system. MKK7 cKO mice displayed impaired circadian behavioral rhythms and decreased locomotor activity. MKK7 cKO mice at 8 months showed motor dysfunctions such as weakness of hind-limb and gait abnormality in an age-dependent manner. Axonal degeneration in the spinal cord and muscle atrophy were also observed, along with accumulation of the axonal transport proteins JNK-interacting protein 1 and amyloid beta precursor protein in the brains and spinal cords of MKK7 cKO mice. Thus, the MKK7-JNK signaling pathway plays important roles in regulating circadian rhythms and neuronal maintenance in the adult nervous system
Quality of life of children with neurodevelopmental disorders and their parents during the COVID-19 pandemic : a 1-year follow-up study
This study aimed to reveal changes in the quality of life (QOL) of children with neurodevelopmental disorders and their parents, and the interaction between their QOL and parental mental state during the coronavirus 2019 (COVID-19) pandemic. Eighty-nine school-aged children and parents participated in surveys in May 2020 (T1) and May 2021 (T2). The parents completed questionnaires that assessed their QOL, depression, parenting stress, and living conditions. Children's temporary mood status was evaluated using the self-reported visual analog scale (VAS). Children's QOL and VAS at T2 were higher than their QOL at T1. Parents' QOL at T2 was lower than their QOL at T1. Severe parental depression at T1 had a synergistic effect on severe parenting stress and severe depressive state at T2. Additionally, children's high QOL at T1 had a synergistic effect on low parenting stress and children's high QOL at T2. Furthermore, children's low VAS scores and parents' low QOL at T2 were associated with deterioration of family economic status. Children and parents' QOL changed during the prolonged COVID-19 pandemic. Improvement in children's QOL was influenced by reduced maternal depressive symptoms. Public support for parental mental health is important to avoid decreasing QOL.Peer reviewe
Phosphorylation of the Leukemic Oncoprotein EVI1 on Serine 196 Modulates DNA Binding, Transcriptional Repression and Transforming Ability
The EVI1 (ecotropic viral integration site 1) gene at 3q26 codes for a transcriptional regulator with an essential role in haematopoiesis. Overexpression of EVI1 in acute myeloid leukaemia (AML) is frequently associated with 3q26 rearrangements and confers extremely poor prognosis. EVI1 mediates transcriptional regulation, signalling, and epigenetic modifications by interacting with DNA, proteins and protein complexes. To explore to what extent protein phosphorylation impacts on EVI1 functions, we analysed endogenous EVI1 protein from a high EVI1 expressing Fanconi anaemia (FA) derived AML cell line. Mass spectrometric analysis of immunoprecipitated EVI1 revealed phosphorylation at serine 196 (S196) in the sixth zinc finger of the N-terminal zinc finger domain. Mutated EVI1 with an aspartate substitution at serine 196 (S196D), which mimics serine phosphorylation of this site, exhibited reduced DNA-binding and transcriptional repression from a gene promotor selectively targeted by the N-terminal zinc finger domain. Forced expression of the S196D mutant significantly reduced EVI1 mediated transformation of Rat1 fibroblasts. While EVI1-mediated serial replating of murine haematopoietic progenitors was maintained by EVI1-S196D, this was associated with significantly higher Evi1-trancript levels compared with WT-EVI1 or EVI1-S196A, mimicking S196 non-phosphorylated EVI1. These data suggest that EVI1 function is modulated by phosphorylation of the first zinc finger domain
Effect of Plateau Carbon-Ion Beams on Lipid Molecules
The effect of the plateau region carbon ion (C-ion) beams (13 keV/µm) on lipid molecules was investigated by 2,2,6-trimethyl-4-(4-nitrobenzo[1,2,5]oxadiazol-7-ylamino)-6-pentylpiperidine-1-oxyl (NBD-Pen), a highly selective fluorescence probe for lipid radicals with high sensitivity. An increase in the fluorescence intensity due to lipid radical adducts of NBD-Pen was observed for the isopropyl myristate (IPM) and acetonitrile (MeCN) solutions of NBD-Pen after the plateau region C-ion irradiation, while such an increase was not observed for a methanol (MeOH) solution of NBD-Pen. A slight increase in the fluorescence intensity was observed after 30 Gy plateau region C-ion irradiation to phosphate buffer solutions containing unsaturated fatty acids, such as linoleic acid (LA) and alpha-linoleic acid (ALA), and NBD-Pen
The quality of life of children with neurodevelopmental disorders and their parents during the Coronavirus disease 19 emergency in Japan
This study aimed to reveal how the COVID-19 stay-at-home period has affected the quality of life (QOL) of children with neurodevelopmental disorders and their parents and to identify possible factors that enabled them to maintain their QOL. We enrolled 136 school-aged children (intellectual quotient >= 50) and their parents and administered QOL questionnaires to assess the maladaptive behavior of the children; depression, anxiety, and stress of the parents; and activities of their daily lives. The relationship between their QOL and clinical features was examined. The decrease in QOL of children and parents was associated with the mother's limited job flexibility. Decreased QOL was also associated with changes in the sleep rhythms of the children. Maladaptive behaviors in children were associated with parental stress. However, maintained QOL of some families who faced these same conditions of job stress and sleep disorders was associated with less parental stress, less parental depression and anxiety, and milder maladaptive behavior in children. Both mothers with limited job flexibility and changes in the sleep rhythm of children were associated with reduced QOL of children and their parents. Low parental stress was associated with decreased maladaptive behavior in children and with maintained QOL of the family.Peer reviewe
