    A global climatology of total columnar water vapour from SSM/I and MERIS

    A global time series of total columnar water vapour from combined data of the Medium Resolution Imaging Spectrometer (MERIS) onboard ESA's Environmental Satellite (ENVISAT) and the Special Sensor Microwave/Imager (SSM/I) onboard the satellite series of the US Defense Meteorological Satellite Program (DMSP) is presented. The unique data set, generated in the framework of the ESA Data User Element (DUE) GlobVapour project, combines atmospheric water vapour observations over land and ocean, derived from measurements in the near- infrared and the microwave range, respectively. Daily composites and monthly means of total columnar water vapour are available as global maps on rectangular latitude–longitude grids with a spatial resolution of 0.05° × 0.05° over land and 0.5° × 0.5° over ocean for the years 2003 to 2008. The data are stored in NetCDF files and is fully compliant with the NetCDF Climate Forecast convention. Through the combination of high-quality microwave observations and near-infrared observations over ocean and land surfaces, respectively, the data set provides global coverage. The combination of both products is carried out such that the individual properties of the microwave and near-infrared products, in particular their uncertainties, are not modified by the merging process and are therefore well defined. Due to the global coverage and the provided uncertainty estimates this data set is potentially of high value for climate research. The SSM/I-MERIS TCWV data set is freely available via the GlobVapour project web page (www.globvapour.info) with associated doi:10.5676/DFE/WV_COMB/FP. In this paper, the details of the data set generation, i.e. the satellite data used, the retrieval techniques and merging approaches, are presented. The derived level 3 products are compared to global radiosonde data from the GCOS upper air network (GUAN), showing a high agreement with a root-mean-square deviation of roughly 4.4 kg m−2 and a small wet bias well below 1 kg m−2. Furthermore, the data set is shown to be free of seasonal biases. The consistency of the MERIS and SSM/I retrievals is demonstrated by applying the MERIS retrieval to sun glint areas over ocean

    An Ecohydrological Perspective on Drought-induced Forest Mortality

    Regional‐scale drought‐induced forest mortality events are projected to become more frequent under future climates due to changes in rainfall patterns. The occurrence of these mortality events is driven by exogenous factors such as frequency and severity of drought and endogenous factors such as tree water and carbon use strategies. To explore the link between these exogenous and endogenous factors underlying forest mortality, a stochastic ecohydrological framework that accounts for random arrival and length of droughts as well as responses of tree water and carbon balance to soil water deficit is proposed. The main dynamics of this system are characterized with respect to the spectrum of anisohydric‐isohydric stomatal control strategies. Using results from a controlled drought experiment, a maximum tolerable drought length at the point where carbon starvation and hydraulic failure occur simultaneously is predicted, supporting the notion of coordinated hydraulic function and metabolism. We find qualitative agreement between the model predictions and observed regional‐scale canopy dieback across a precipitation gradient during the 2002–2003 southwestern United States drought. Both the model and data suggest a rapid increase of mortality frequency below a precipitation threshold. The model also provides estimates of mortality frequency for given plant drought strategies and climate regimes. The proposed ecohydrological approach can be expanded to estimate the effect of anticipated climate change on drought‐induced forest mortality and associated consequences for the water and carbon balances

    The twilight of the Liberal Social Contract? On the Reception of Rawlsian Political Liberalism

    This chapter discusses the Rawlsian project of public reason, or public justification-based 'political' liberalism, and its reception. After a brief philosophical rather than philological reconstruction of the project, the chapter revolves around a distinction between idealist and realist responses to it. Focusing on political liberalism’s critical reception illuminates an overarching question: was Rawls’s revival of a contractualist approach to liberal legitimacy a fruitful move for liberalism and/or the social contract tradition? The last section contains a largely negative answer to that question. Nonetheless the chapter's conclusion shows that the research programme of political liberalism provided and continues to provide illuminating insights into the limitations of liberal contractualism, especially under conditions of persistent and radical diversity. The programme is, however, less receptive to challenges to do with the relative decline of the power of modern states

    Similarities and differences in the autonomic control of airway and urinary bladder smooth muscle

    The airways and the urinary bladder are both hollow organs serving very different functions, i.e. air flow and urine storage, respectively. While the autonomic nervous system seems to play only a minor if any role in the physiological regulation of airway tone during normal breathing, it is important in the physiological regulation of bladder smooth muscle contraction and relaxation. While both tissues share a greater expression of M2 than of M3 muscarinic receptors, smooth muscle contraction in both is largely mediated by the smaller M3 population apparently involving phospholipase C activation to only a minor if any extent. While smooth muscle in both tissues can be relaxed by β-adrenoceptor stimulation, this primarily involves β2-adrenoceptors in human airways and β3-adrenoceptors in human bladder. Despite activation of adenylyl cyclase by either subtype, cyclic adenosine monophosphate plays only a minor role in bladder relaxation by β-agonists; an important but not exclusive function is known in airway relaxation. While airway β2-adrenoceptors are sensitive to agonist-induced desensitization, β3-adrenoceptors are generally considered to exhibit much less if any sensitivity to desensitization. Gene polymorphisms exist in the genes of both β2- and β3-adrenoceptors. Despite being not fully conclusive, the available data suggest some role of β2-adrenoceptor polymorphisms in airway function and its treatment by receptor agonists, whereas the available data on β3-adrenoceptor polymorphisms and bladder function are too limited to allow robust interpretation. We conclude that the distinct functions of airways and urinary bladder are reflected in a differential regulation by the autonomic nervous system. Studying these differences may be informative for a better understanding of each tissue

    Tandem E2F Binding Sites in the Promoter of the p107 Cell Cycle Regulator Control p107 Expression and Its Cellular Functions

    The retinoblastoma tumor suppressor (Rb) is a potent and ubiquitously expressed cell cycle regulator, but patients with a germline Rb mutation develop a very specific tumor spectrum. This surprising observation raises the possibility that mechanisms that compensate for loss of Rb function are present or activated in many cell types. In particular, p107, a protein related to Rb, has been shown to functionally overlap for loss of Rb in several cellular contexts. To investigate the mechanisms underlying this functional redundancy between Rb and p107 in vivo, we used gene targeting in embryonic stem cells to engineer point mutations in two consensus E2F binding sites in the endogenous p107 promoter. Analysis of normal and mutant cells by gene expression and chromatin immunoprecipitation assays showed that members of the Rb and E2F families directly bound these two sites. Furthermore, we found that these two E2F sites controlled both the repression of p107 in quiescent cells and also its activation in cycling cells, as well as in Rb mutant cells. Cell cycle assays further indicated that activation of p107 transcription during S phase through the two E2F binding sites was critical for controlled cell cycle progression, uncovering a specific role for p107 to slow proliferation in mammalian cells. Direct transcriptional repression of p107 by Rb and E2F family members provides a molecular mechanism for a critical negative feedback loop during cell cycle progression and tumorigenesis. These experiments also suggest novel therapeutic strategies to increase the p107 levels in tumor cells

    Efficacy of Budesonide Orodispersible Tablets as Induction Therapy for Eosinophilic Esophagitis in a Randomized Placebo-Controlled Trial.

    BACKGROUND & AIMS: Swallowed topical-acting corticosteroids are recommended as first-line therapy for eosinophilic esophagitis (EoE). Asthma medications not optimized for esophageal delivery are sometimes effective, although given off-label. We performed a randomized, placebo-controlled trial to assess the effectiveness and tolerability of a budesonide orodispersible tablet (BOT), which allows the drug to be delivered to the esophagus in adults with active EoE. METHODS: We performed a double-blind, parallel study of 88 adults with active EoE in Europe. Patients were randomly assigned to groups that received BOT (1 mg twice daily; n = 59) or placebo (n = 29) for 6 weeks. The primary end point was complete remission, based on clinical and histologic factors, including dysphagia and odynophagia severity ≤2 on a scale of 0-10 on each of the 7 days before the end of the double-blind phase and a peak eosinophil count <5 eosinophils/high power field. Patients who did not achieve complete remission at the end of the 6-week double-blind phase were offered 6 weeks of open-label treatment with BOT (1 mg twice daily). RESULTS: At 6 weeks, 58% of patients given BOT were in complete remission compared with no patients given placebo (P < .0001). The secondary end point of histologic remission was achieved by 93% of patients given BOT vs no patients given placebo (P < .0001). After 12 weeks, 85% of patients had achieved remission. Six-week and 12-week BOT administration were safe and well tolerated; 5% of patients who received BOT developed symptomatic, mild candida, which was easily treated with an oral antifungal agent. CONCLUSIONS: In a randomized trial of adults with active EoE, we found that budesonide oral tablets were significantly more effective than placebo in inducing clinical and histologic remission.
