    Cell cycle independent role of cyclin D3 in host restriction of influenza virus infection

    To identify new host factors that modulate the replication of influenza A virus, we performed a yeast two-hybrid screen using the cytoplasmic tail of matrix protein 2 from the highly pathogenic H5N1 strain. The screen revealed a high-score interaction with cyclin D3, a key regulator of cell cycle early G1 phase. M2-cyclin D3 interaction was validated through GST pull-down and recapitulated in influenza A/WSN/33-infected cells. Knockdown of Ccnd3 by small interfering RNA significantly enhanced virus progeny titers in cell culture supernatants. Interestingly, the increase in virus production was due to cyclin D3 deficiency per se, and not merely a consequence of cell cycle deregulation. A combined knockdown of Ccnd3 and Rb1, which rescued cell cycle progression into the S phase, failed to normalize virus production. Infection by IAV triggered redistribution of cyclin D3 from the nucleus to the cytoplasm followed by its proteasomal degradation. When over-expressed in HEK 293T cells cyclin D3 impaired binding of M2 with M1, which is essential for proper assembly of progeny virions, lending further support to its role as a putative restriction factor. Our study describes the identification and characterization of cyclin D3 as a novel interactor of influenza A virus M2 protein. We hypothesize that competitive inhibition of M1-M2 interaction by cyclin D3 impairs infectious virion formation and results in attenuated virus production.     THE RELATION BETWEEN METACOGNITION AND INTELLIGENCE IN ADOLESCENTS

    The primary purpose of this study was to investigate the relation between metacognitive abilities and intelligence. The secondary intent of the study was to examine the validity of methods used to assess metacognition. Subjects consisted of 38 grade nine students, whose IQs ranged between 80-137, measured using a short form of the WISC-R. Selected subjects successfully completed three sets of tasks on a problem solving battery and were interviewed concerning their awareness of possible strategies to be used in solving the tasks. The interviews were videotaped and subsequently scored for metacognitive knowledge, utilizing both a quantitative and a qualitative methodology. Two university students viewed the videotapes and independently checked off on a quantitative scoring sheet, strategy factors which subjects clearly related in the interviews. The quantitative metacognitive scores for the three sets of problems consisted of the number of relevant strategies checked off on the score sheet. A qualitative measure of metacognition was obtained by rating the interviews using a Q-sort technique, employing a rank-ordering procedure of piles. The Q-sort was done by two educational psychology graduate students knowledgeable of metacognition. Data were analyzed with subjects\u27 IQs serving as the independent variable and the quantitative and qualitative scores being the dependent variables. Two multivariate multiple regressions were done; the first to study the relation between the quantitative measure of metacognition and the IQ scores, and the second to look at the relation between the qualitative measure of metacognition and the IQ scores. A Pearson correlational analysis was done between corresponding pairs of quantitative and qualitative metacognitive scores to assess their relation. The results indicated there was no statistically significant relation between IQ and either of the metacognitive measures. An overall significant moderate correlation between the two metacognitive measures appears to indicate they are measuring similar, although not identical aspects of metacognition. Based on this study, it seems that metacognition can be measured in an objective, quantifiable manner

    Expression de Zap-70 dans les lymphocytes périphériques de sang normal et de Leucémie Lymphoïde Chronique dans un modèle unicellulaire

    Zap-70 est une protéine majeure de la signalisation intracellulaire dans les lymphocytes T et NK. Zap-70 est exprimée à tous les stades de maturation dans les lymphocytes B murins, tandis que chez l homme Zap-70 est détectée dans certaines hémopathies lymphoïdes B, dans les précurseurs B normaux, et les lymphocytes B activés.L expression de Zap-70 dans les lymphocytes B normaux, matures, périphériques chez l homme est un sujet de controverse.Pour répondre à cette question, nous avons mis au point une RT-PCR qualitative pour détecter Zap-70 dans les lymphocytes B périphériques à l échelon unicellulaire. Nos résultats sont en faveur d une absence d expression de Zap-70 dans les lymphocytes B périphériques matures. Nous mettons en évidence une hétérogénéité d expression de Zap-70 dans les lymphocytes B de LLC, ainsi que dans les lymphocytes T et NK périphériques à l échelon unicellulaire.PARIS6-Bibl.Pitié-Salpêtrie (751132101) / SudocSudocFranceF
