    Cloning and characterization of 5 '-upstream region of human phospholipase C-beta 2 gene

    5 ' -upstream region of the phospholipase C-beta2 gene, 810 bp, was cloned and characterized. S1 nuclease mapping and primer extension analyses revealed that a single transcriptional start site locates at 284 nucleotides upstream from the beginning of translation. The 5 ' -upstream region lacks both TATA motif and typical initiator sequence, but retains CC-rich segment. Two putative regulatory regions, a negative region (-636/-588) and a positive region (-98/-13) were identified in the upstream region of PLC-beta2 gene. We suggest that the transcription of PLC-beta2 may be regulated by binding of regulatory proteins to the negative and/or positive regulatory regions located in the upstream of the geneopen

    Magnetic Properties of Dilute Alloys: Equations for Magnetization and its Structural Fluctuations

    The dilute Heisenberg ferromagnet is studied taking into account fluctuations of magnetization caused by disorder. A self-consistent system of equations for magnetization and its mean quadratic fluctuations is derived within the configurationally averaged two-time temperature Green's function method. This system of equations is analised at low concentration of non-magnetic impurities. Mean relative quadratic fluctuations of magnetization are revealed to be proportional to the square of concentration of impurities.Comment: 16 pages, LaTe

    On the complexity of the Saccharomyces bayanus taxon: hybridization and potential hybrid speciation

    Although the genus Saccharomyces has been thoroughly studied, some species in the genus has not yet been accurately resolved; an example is S. bayanus, a taxon that includes genetically diverse lineages of pure and hybrid strains. This diversity makes the assignation and classification of strains belonging to this species unclear and controversial. They have been subdivided by some authors into two varieties (bayanus and uvarum), which have been raised to the species level by others. In this work, we evaluate the complexity of 46 different strains included in the S. bayanus taxon by means of PCR-RFLP analysis and by sequencing of 34 gene regions and one mitochondrial gene. Using the sequence data, and based on the S. bayanus var. bayanus reference strain NBRC 1948, a hypothetical pure S. bayanus was reconstructed for these genes that showed alleles with similarity values lower than 97% with the S. bayanus var. uvarum strain CBS 7001, and of 99¿100% with the non S. cerevisiae portion in S. pastorianus Weihenstephan 34/70 and with the new species S. eubayanus. Among the S. bayanus strains under study, different levels of homozygosity, hybridization and introgression were found; however, no pure S. bayanus var. bayanus strain was identified. These S. bayanus hybrids can be classified into two types: homozygous (type I) and heterozygous hybrids (type II), indicating that they have been originated by different hybridization processes. Therefore, a putative evolutionary scenario involving two different hybridization events between a S. bayanus var. uvarum and unknown European S. eubayanus-like strains can be postulated to explain the genomic diversity observed in our S. bayanus var. bayanus strains

    Penelitian ini mempelajari tentang karakteristik kuat tekan dan karakteristik pembakaran briket daun cengkeh dan jerami padi. Pembriketan dilakukan dengan menggunakan mesin pres hidrolik dengan tekanan pembriketan sebesar 450 kg/cm2, dengan bahan pengikat dan tanpa bahan pengikat. Bahan pengikat yang digunakan adalah lem kanji dengan kadar 5 %. Briket berbentuk silinder dengan diameter sekitar 3 cm dan tinggi 5 cm. Variasi parameter pembriketan yang digunakan adalah ukuran butir 20, 40 dan 80 mesh, kadar air 15 %, 20 % dan 25 %, serta suhu pembriketan sebesar 60 oC, 80 oC, 100 oC dan 120 oC. Uji pembakaran dilakukan dalam tungku berbentuk tabung horisontal berdiameter dalam 170 mm. Variasi perameter uji pembakaran yang digunakan adalah kecepatan aliran udara sebesar 0,6 m/s; 0,8 m/s; 1,0 m/s dan 1,2 m/s serta variasi ukuran butir sebesar 20, 40, dan 80 mesh. Suhu pembriketan berpengaruh signifikan terhadap peningkatan kuat tekan briket. Dari hasil uji pembakaran dapat ditentukan besarnya laju pembakaran, profil suhu pembakaran, nilai energi aktivasi (E ), konstanta Arrhenius (A), dan emisi CO. Dari semua percobaan, kadar emisi CO puncak lebih dari 400 ppm. Kata kunci: kuat tekan, daun cengkeh, jerami, bahan pengikat, ukuran butir, suhu pembriketan, kadar air, laju pembakaran, energi aktivasi, emisi CO.

    The S phase checkpoint promotes the Smc5/6 complex dependent SUMOylation of Pol2, the catalytic subunit of DNA polymerase ε

    Replication fork stalling and accumulation of single-stranded DNA trigger the S phase checkpoint, a signalling cascade that, in budding yeast, leads to the activation of the Rad53 kinase. Rad53 is essential in maintaining cell viability, but its targets of regulation are still partially unknown. Here we show that Rad53 drives the hyper-SUMOylation of Pol2, the catalytic subunit of DNA polymerase ε, principally following replication forks stalling induced by nucleotide depletion. Pol2 is the main target of SUMOylation within the replisome and its modification requires the SUMO-ligase Mms21, a subunit of the Smc5/6 complex. Moreover, the Smc5/6 complex co-purifies with Pol ε, independently of other replisome components. Finally, we map Pol2 SUMOylation to a single site within the N-terminal catalytic domain and identify a SUMO-interacting motif at the C-terminus of Pol2.     Molecular psychiatry of zebrafish

    Due to their well-characterized neural development and high genetic homology to mammals, zebrafish (Danio rerio) have emerged as a powerful model organism in the field of biological psychiatry. Here, we discuss the molecular psychiatry of zebrafish, and its implications for translational neuroscience research and modeling central nervous system (CNS) disorders. In particular, we outline recent genetic and technological developments allowing for in vivo examinations, high-throughput screening and whole-brain analyses in larval and adult zebrafish. We also summarize the application of these molecular techniques to the understanding of neuropsychiatric disease, outlining the potential of zebrafish for modeling complex brain disorders, including attention-deficit/hyperactivity disorder (ADHD), aggression, post-traumatic stress and substance abuse. Critically evaluating the advantages and limitations of larval and adult fish tests, we suggest that zebrafish models become a rapidly emerging new field in modern molecular psychiatry research

    Charged-particle distributions at low transverse momentum in √s=13 13 TeV pp interactions measured with the ATLAS detector at the LHC

    Measurements of distributions of charged particles produced in proton–proton collisions with a centre-of-mass energy of 13 TeV are presented. The data were recorded by the ATLAS detector at the LHC and correspond to an integrated luminosity of 151 μb −1 μb−1 . The particles are required to have a transverse momentum greater than 100 MeV and an absolute pseudorapidity less than 2.5. The charged-particle multiplicity, its dependence on transverse momentum and pseudorapidity and the dependence of the mean transverse momentum on multiplicity are measured in events containing at least two charged particles satisfying the above kinematic criteria. The results are corrected for detector effects and compared to the predictions from several Monte Carlo event generators

    Search for supersymmetry at √s = 13 TeV in final states with jets and two same-sign leptons or three leptons with the ATLAS detector

    A search for strongly produced supersymmetric particles is conducted using signatures involving multiple energetic jets and either two isolated leptons (e or μμ ) with the same electric charge or at least three isolated leptons. The search also utilises b-tagged jets, missing transverse momentum and other observables to extend its sensitivity. The analysis uses a data sample of proton–proton collisions at s√=13s=13 TeV recorded with the ATLAS detector at the Large Hadron Collider in 2015 corresponding to a total integrated luminosity of 3.2 fb −1−1. No significant excess over the Standard Model expectation is observed. The results are interpreted in several simplified supersymmetric models and extend the exclusion limits from previous searches. In the context of exclusive production and simplified decay modes, gluino masses are excluded at 95%95% confidence level up to 1.1–1.3 TeV for light neutralinos (depending on the decay channel), and bottom squark masses are also excluded up to 540 GeV. In the former scenarios, neutralino masses are also excluded up to 550–850 GeV for gluino masses around 1 TeV

    Measurement of the inelastic proton-proton cross section at √s=13 TeV with the ATLAS detector at the LHC

    This Letter presents a measurement of the inelastic proton-proton cross section using 60  μb −1 of pp collisions at a center-of-mass energy √s of 13 TeV with the ATLAS detector at the LHC. Inelastic interactions are selected using rings of plastic scintillators in the forward region (2.0710 −6 , where M X is the larger invariant mass of the two hadronic systems separated by the largest rapidity gap in the event. In this ξ range the scintillators are highly efficient. For diffractive events this corresponds to cases where at least one proton dissociates to a system with M X >13  GeV . The measured cross section is compared with a range of theoretical predictions. When extrapolated to the full phase space, a cross section of 78.1±2.9  mb is measured, consistent with the inelastic cross section increasing with center-of-mass energy
