    The effect of carnitine on Arabidopsis development and recovery in salt stress conditions

    Carnitine exists in all living organisms where it plays diverse roles. In animals and yeast, it is implicated in lipid metabolism and is also associated with oxidative stress tolerance. In bacteria, it is a major player in osmotic stress tolerance. We investigate the carnitine function in plants and our present work shows that carnitine enhances the development and recovery of Arabidopsis thaliana seedlings subjected to salt stress. Biological data show that exogenous carnitine supplies improve the germination and survival rates of seedlings grown on salt-enriched medium, in a manner comparable to proline. Both compounds are shown to improve seedling survival under oxidative constraint meaning that they may act on salt stress through antioxidant properties. A transcriptome analysis of seedlings treated with exogenous carnitine reveals that it modulates the expression of genes involved in water stress and abscisic acid responses. Analyses of the abscisic acid mutants, aba1-1 and abi1-1, indicate that carnitine and proline may act through a modulation of the ABA pathway

    Molecular bacterial load assay, a culture-free biomarker for rapid and accurate quantification of sputum Mycobacterium tuberculosis bacillary load during treatment

    A molecular assay to quantify Mycobacterium tuberculosis is described. In vitro, 98% (n = 96) of sputum samples with a known number of bacilli (107 to 102 bacilli) could be enumerated within 0.5 log10. In comparison to culture, the molecular bacterial load (MBL) assay is unaffected by other microorganisms present in the sample, results are obtained more quickly (within 24 h) and are seldom inhibited (0.7% samples), and the MBL assay critically shows the same biphasic decline as observed longitudinally during treatment. As a biomarker of treatment response, the MBL assay responds rapidly, with a mean decline in bacterial load for 111 subjects of 0.99 log10 (95% confidence interval [95% CI], 0.81 to 1.17) after 3 days of chemotherapy. There was a significant association between the rate of bacterial decline during the same 3 days and bacilli ml−1 sputum at day 0 (linear regression, P = 0.0003) and a 3.62 increased odds ratio of relapse for every 1 log10 increase in pretreatment bacterial load (95% CI, 1.53 to 8.59)

    The role of skeletal micro-architecture in diagenesis and dating of Acropora palmata

    Past variations in global sea-level reflect continental ice volume, a crucial factor for understanding the Earth's climate system. The Caribbean coral Acropora palmata typically forms dense stands in very shallow water and therefore fossil samples mark past sea-level. Uranium-series methods are commonly used to establish a chronology for fossil coral reefs, but are compromised by post mortem diagenetic changes to coral skeleton. Current screening approaches are unable to identify all altered samples, whilst models that attempt to correct for 'open-system' behaviour are not applicable across all diagenetic scenarios. In order to better understand how U-series geochemistry varies spatially with respect to diagenetic textures, we examine these aspects in relation to skeletal micro-structure and intra-crystalline amino acids, comparing an unaltered modern coral with a fossil A. palmata colony containing zones of diagenetic alteration (secondary overgrowth of aragonite, calcite cement and dissolution features). We demonstrate that the process of skeletogenesis in A. palmata causes heterogeneity in porosity, which can account for the observed spatial distribution of diagenetic features; this in turn explains the spatially-systematic trends in U-series geochemistry and consequently, U-series age. We propose a scenario that emphasises the importance of through-flow of meteoric waters, invoking both U-loss and absorption of mobilised U and Th daughter isotopes. We recommend selective sampling of low porosity A. palmata skeleton to obtain the most reliable U-series ages. We demonstrate that intra-crystalline amino acid racemisation (AAR) can be applied as a relative dating tool in Pleistocene A. palmata samples that have suffered heavy dissolution and are therefore unsuitable for U-series analyses. Based on relatively high intra-crystalline concentrations and appropriate racemisation rates, glutamic acid and valine are most suited to dating mid-late Pleistocene A. palmata. Significantly, the best-preserved material in the fossil specimen yields a U-series age of 165 ± 8 ka, recording a paleo sea-level of -35 ± 7 msl during the MIS 6.5 interstadial on Barbados

    We show the distribution of SARS-CoV-2 genetic clades over time and between countries and outline potential genomic surveillance objectives. We applied three available genomic nomenclature systems for SARS-CoV-2 to all sequence data from the WHO European Region available during the COVID-19 pandemic until 10 July 2020. We highlight the importance of real-time sequencing and data dissemination in a pandemic situation. We provide a comparison of the nomenclatures and lay a foundation for future European genomic surveillance of SARS-CoV-2.Peer reviewe

    Multiplicity dependence of inclusive J/psi production at midrapidity in pp collisions at root s=13 TeV

    Measurements of the inclusive J/psi yield as a function of charged-particle pseudorapidity density dN(ch)/d eta in pp collisions at root s = 13 TeV with ALICE at the LHC are reported. The J/psi meson yield is measured at midrapidity (vertical bar y vertical bar <0.9) in the dielectron channel, for events selected based on the charged-particle multiplicity at midrapidity (vertical bar eta vertical bar <1) and at forward rapidity (-3.7 <eta <-1.7 and 2.8 <eta <5.1); both observables are normalized to their corresponding averages in minimum bias events. The increase of the normalized J/psi yield with normalized dN(ch)/d eta is significantly stronger than linear and dependent on the transverse momentum. The data are compared to theoretical predictions, which describe the observed trends well, albeit not always quantitatively. (C) 2020 European Organization for Nuclear Research.     Measurement of the azimuthal anisotropy of Y(1S) and Y(2S) mesons in PbPb collisions at √S^{S}NN = 5.02 TeV

    The second-order Fourier coefficients (υ2_{2}) characterizing the azimuthal distributions of ΄(1S) and ΄(2S) mesons produced in PbPb collisions at sNN\sqrt{s_{NN}} = 5.02 TeV are studied. The ΄mesons are reconstructed in their dimuon decay channel, as measured by the CMS detector. The collected data set corresponds to an integrated luminosity of 1.7 nb−1^{-1}. The scalar product method is used to extract the υ2_{2} coefficients of the azimuthal distributions. Results are reported for the rapidity range |y| < 2.4, in the transverse momentum interval 0 < pT_{T} < 50 GeV/c, and in three centrality ranges of 10–30%, 30–50% and 50–90%. In contrast to the J/ψ mesons, the measured υ2_{2} values for the ΄ mesons are found to be consistent with zero

    Measurement of prompt D0^{0} and D‟\overline{D}0^{0} meson azimuthal anisotropy and search for strong electric fields in PbPb collisions at root SNN\sqrt{S_{NN}} = 5.02 TeV

    The strong Coulomb field created in ultrarelativistic heavy ion collisions is expected to produce a rapiditydependent difference (Av2) in the second Fourier coefficient of the azimuthal distribution (elliptic flow, v2) between D0 (uc) and D0 (uc) mesons. Motivated by the search for evidence of this field, the CMS detector at the LHC is used to perform the first measurement of Av2. The rapidity-averaged value is found to be (Av2) = 0.001 ? 0.001 (stat)? 0.003 (syst) in PbPb collisions at ?sNN = 5.02 TeV. In addition, the influence of the collision geometry is explored by measuring the D0 and D0mesons v2 and triangular flow coefficient (v3) as functions of rapidity, transverse momentum (pT), and event centrality (a measure of the overlap of the two Pb nuclei). A clear centrality dependence of prompt D0 meson v2 values is observed, while the v3 is largely independent of centrality.
