    Effect of Restricted Preen-Gland Access on Maternal Self Maintenance and Reproductive Investment in Mallards

    As egg production and offspring care are costly, females should invest resources adaptively into their eggs to optimize current offspring quality and their own lifetime reproductive success. Parasite infections can influence maternal investment decisions due to their multiple negative physiological effects. The act of preening--applying oils with anti-microbial properties to feathers--is thought to be a means by which birds combat pathogens and parasites, but little is known of how preening during the reproductive period (and its expected disease-protecting effects) influences maternal investment decisions at the level of the egg.Here, we experimentally prevented female mallards (Anas platyrhynchos) from accessing their preen gland during breeding and monitored female immunoresponsiveness (e.g., plasma lysozyme concentration) as well as some egg traits linked to offspring quality (e.g., egg mass, yolk carotenoid content, and albumen lysozyme levels). Females with no access to their preen gland showed an increase in plasma lysozyme level compared to control, normally preening females. In addition, preen-gland-restricted females laid significantly lighter eggs and deposited higher carotenoid concentrations in the yolk compared to control females. Albumen lysozyme activity did not differ significantly between eggs laid by females with or without preen gland access.Our results establish a new link between an important avian self-maintenance behaviour and aspects of maternal health and reproduction. We suggest that higher yolk carotenoid levels in eggs laid by preen-gland-restricted females may serve to boost health of offspring that would hatch in a comparatively microbe-rich environment

    Neonatal presentation of ventricular tachycardia and a Reye-like syndrome episode associated with disturbed mitochondrial energy metabolism

    BACKGROUND: Hyperammonemia, hypoglycemia, hepatopathy, and ventricular tachycardia are common presenting features of carnitine-acylcarnitine translocase deficiency (Mendelian Inheritance in Man database: *212138), a mitochondrial fatty acid oxidation disorder with a lethal prognosis. These features have not been identified as the presenting features of mitochondrial cytopathy in the neonatal period. CASE PRESENTATION: We describe an atypical presentation of mitochondrial cytopathy in a 2 day-old neonate. She presented with a Reye-like syndrome episode, premature ventricular contractions and ventricular tachycardia. Initial laboratory evaluation exhibited a large amount of 3-methylglutaconic acid on urine organic acid analysis, mild orotic aciduria and a nonspecific abnormal acylcarnitine profile. The evaluation for carnitine-acylcarnitine translocase deficiency and other fatty acid oxidation disorders was negative. The patient later developed a hypertrophic cardiomyopathy and continued to be affected by recurrent Reye-like syndrome episodes triggered by infections. A muscle biopsy exhibited signs of a mitochondrial cytopathy. During the course of her disease, her Reye-like syndrome episodes have subsided; however, cardiomyopathy has persisted along with fatigue and exercise intolerance. CONCLUSIONS: This case illustrates that, in the neonatal period, hyperammonemia and ventricular tachycardia may be the presenting features of a lethal carnitine-acylcarnitine translocase deficiency or of a mitochondrial cytopathy, associated with a milder clinical course. This association broadens the spectrum of presenting phenotypes observed in patients with disturbed mitochondrial energy metabolism. Also, the presence of 3-methylglutaconic aciduria suggests mitochondrial dysfunction and mild orotic aciduria could potentially be used as a marker of mitochondrial disease

    Guidelines for the use and interpretation of assays for monitoring autophagy (3rd edition)

    In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure fl ux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defi ned as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (inmost higher eukaryotes and some protists such as Dictyostelium ) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the fi eld understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation it is imperative to delete or knock down more than one autophagy-related gene. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways so not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field

    Orbital spectral variables, growth analysis and sugarcane yield Variáveis espectrais orbitais, indicadoras de desenvolvimento e produtividade da cana-de-açúcar

    Temporal analysis of crop development in commercial fields requires tools for large area monitoring, such as remote sensing. This paper describes the temporal evolution of sugar cane biophysical parameters such as total biomass (BMT), yield (TSS), leaf area index (LAI), and number of plants per linear meter (NPM) correlated to Landsat data. During the 2000 and 2001 cropping seasons, a commercial sugarcane field in Araras, São Paulo state, Brazil, planted with the SP80-1842 sugarcane variety in the 4th and 5th cuts, was monitored using nine Landsat images. Spectral data were correlated with agronomic data, obtained simultaneously to the imagery acquisition. Two methodologies were used to collect spectral data from the images: four pixels (2 &#215; 2) window and average of total pixels in the field. Linear and multiple regression analysis was used to study the spectral behavior of the plants and to correlate with agronomic variables (days after harvest-DAC, LAI, NPM, BMT and TSS). No difference was observed between the methodologies to collect spectral data. The best models to describe the spectral crop development in relation to DAC were the quadratic and cubic models. Ratio vegetation index and normalized difference vegetation index demonstrated correlation with DAC, band 3 (B3) was correlated with LAI, and NDVI was well correlated with TSS and BMT. The best fit curves to estimate TSS and BMT presented r² between 0.68 and 0.97, suggesting good potential in using orbital spectral data to monitor sugarcane fields.<br>Dados de satélites são tradicionalmente utilizados em monitoramento de culturas. O presente trabalho busca contribuir no entendimento da evolução temporal de indicadores de crescimento da cana-de-açúcar como a biomassa total (BMT), produtividade (TSS), índice de área foliar (LAI) e número de plantas por metro (NPM) por meio de dados orbitais dos satélites Landsat 5 e 7, e verificar o seu potencial para o monitoramento desta. Durante as safras 2000 e 2001, uma área comercial em Araras, SP, cultivada com a variedade SP80-1842 no 4º e 5º cortes, foi acompanhada por imagens, buscando-se correlacionar dados espectrais com dados agronômicos. Os dados espectrais foram coletados de duas formas: uma com janelas de quatro pixels e outra com dados médios do talhão (DMt). Regressão linear e múltipla foram usadas para a análise temporal das bandas 3 e 4 e de índices de vegetação. As correlações e ajuste de modelos entre os dados espectrais orbitais e as variáveis agronômicas não apresentaram diferenças estatísticas. Os modelos quadráticos e cúbicos melhor descreveram o desenvolvimento temporal das variáveis espectrais, em função dos dias após o corte e apresentaram significância com os índices de vegetação da razão e por diferença normalizada (NDVI). As correlações entre os dados espectrais médios do talhão e as variáveis agronômicas foram significativas para banda3 e LAI, e entre NDVI e TSS/BMT. Os dados médios do talhão (DMt), para primeira safra (1ªS), para a segunda safra (2ªS) e ambas juntas geraram regressões múltiplas, com coeficientes determinação (r²) variando de 0,68 a 0,97 para a TSS e a BMT, mostrando que os dados espectrais orbitais estudados podem ser empregados no monitoramento da cultura da cana-de-açúcar

    High contribution of Rhizaria (Radiolaria) to vertical export in the California Current Ecosystem revealed by DNA metabarcoding

    International audiencePassive sinking of particulate organic matter (POM) is the main mechanism through which the biological pump transports surface primary production to the ocean interior. However, the contribution and variability of different biological sources to vertical export is not fully understood. Here, we use DNA metabarcoding of the 18S rRNA gene and particle interceptor traps (PITs) to characterize the taxonomic composition of particles sinking out of the photic layer in the California Current Ecosystem (CCE), a productive system with high export potential. The PITs included formalin-fixed and ‘live’ traps to investigate eukaryotic communities involved in the export and remineralization of sinking particles. Sequences affiliated with Radiolaria dominated the eukaryotic assemblage in fixed traps (90%), with Dinophyta and Metazoa making minor contributions. The prominence of Radiolaria decreased drastically in live traps, possibly due to selective consumption by copepods, heterotrophic nanoflagellates, and phaeodarians that were heavily enriched in these traps. These patterns were consistent across the water masses surveyed extending from the coast to offshore, despite major differences in productivity and trophic structure of the epipelagic plankton community. Our findings identify Radiolaria as major actors in export fluxes in the CCE

    Life history shapes variation in egg composition in the blue tit Cyanistes caeruleus

    Maternal investment directly shapes early developmental conditions and therefore has long-term fitness consequences for the offspring. In oviparous species prenatal maternal investment is fixed at the time of laying. To ensure the best survival chances for most of their offspring, females must equip their eggs with the resources required to perform well under various circumstances, yet the actual mechanisms remain unknown. Here we describe the blue tit egg albumen and yolk proteomes and evaluate their potential to mediate maternal effects. We show that variation in egg composition (proteins, lipids, carotenoids) primarily depends on laying order and female age. Egg proteomic profiles are mainly driven by laying order, and investment in the egg proteome is functionally biased among eggs. Our results suggest that maternal effects on egg composition result from both passive and active (partly compensatory) mechanisms, and that variation in egg composition creates diverse biochemical environments for embryonic development
