    Magnolia officinalis L. bark extract and respiratory diseases: From traditional Chinese medicine to western medicine via network target

    The understanding of the use of Magnolia officinalis L. (Magnoliaceae) as a possible dietary supplement for supporting the treatment of airway pathologies might be of clinical interest. Two commercially available bark extracts (M. officinalis extract [MOE]) were characterized by quantitation in honokiol and magnolol content by means of high-performance liquid chromatography with UV detection. MOE effects, as well as those of the reference compounds per se, on some targets connected to airway pathologies (antibacterial- and lung and trachea relaxing- activities) were investigated. Results showed that MOE possessed interesting antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa, and Streptococcus pneumoniae. This was accompanied by a spasmolytic and antispasmodic activity, possibly owing to its ability to concurrently modulate different targets such as H-1-, beta(2)- and muscarinic receptors and l-type calcium channels involved in bronchodilation. All these effects were directly related to the MOE content in honokiol and magnolol. In conclusion, the properties of MOE highlighted here strongly encourage its application as dietary supplement in the treatment of airway diseases

    CLARO-CMOS, an ASIC for single photon counting with Ma-PMTs, MCPs and SiPMs

    An ASIC named CLARO-CMOS was designed for fast photon counting with MaPMTs, MCPs and SiPMs. The prototype was realized in a .35 ÎŒm CMOS technology and has four channels, each with a fast amplifier and a discriminator. The main features of the design are the high speed of operation and the low power dissipation, below 1 mW per channel. This paper focuses on the use of the CLARO for SiPM readout. The ASIC was tested with several SiPMs of various sizes, connected to the input of the chip both directly and through a coaxial cable about one meter long. In the latter case the ASIC is still fully functional although the speed of response is affected by the cable capacitance. The threshold could be set just above the single photoelectron level, and with 1 ×1 mm 2 SiPMs the discrete photoelectron peaks could be well resolve

    THE USE OF NUDGES FOR HAND HYGIENE AS A COMMUNITY MITIGATION STRATEGY IN THE FACE OF THE COVID-19 PANDEMIC

    Em um cenĂĄrio pandĂȘmico da COVID-19, comportamentos simples, como higienizar as mĂŁos, podem ter especial relevĂąncia na prevenção do contĂĄgio e no retardamento da progressĂŁo dos casos. Muitas das intervençÔes para o aumento da frequĂȘncia da higienização sĂŁo baseadas em campanhas de conscientização, seja com mensagens de apelos racionais ou emocionais. Contudo, essas intervençÔes tĂȘm demonstrado grandes limitaçÔes em seus resultados. Uma compreensĂŁo de que comportamentos sĂŁo influenciados por fatores ambientais pode ajudar a mudar a estratĂ©gia. O uso de nudges pode ser uma intervenção de baixo custo, acessĂ­vel, simples e nĂŁo constrangedora capaz de aumentar o comportamento de higienização. O presente artigo descreve a operacionalização de uma intervenção em uma escola rural primĂĄria de Bangladesh e explora outras possĂ­veis tecnologias comportamentais como estratĂ©gias mitigatĂłrias comunitĂĄrias. Palavras-chave: Comportamento; Nudges; Higienização das mĂŁos, COVID-19.In a pandemic scenario of COVID-19, simple behaviors, such as hand hygiene, can have special relevance in preventing contagion and delaying the progression of cases. Many interventions to increase hygiene are based on awareness campaigns, whether with messages of rational or emotional appeals. However, this intervention has shown great limitations in its results. An understanding that behaviors are influenced by environmental factors can help to change the strategy. The use of nudges can be a low-cost, accessible, simple, and non-embarrassing intervention capable of increasing hygiene behavior. This article describes the operationalization of an intervention in a rural primary school in Bangladesh and explores other possible behavioral technologies such as community mitigation strategies.Keywords: Behavior; Nudge; Hand hygiene, COVID-19

A partir disso, o site “Ativação em Casa” Ă© baseado em princĂ­pios da Ativação Comportamental que objetiva auxiliar as pessoas na construção de rotinas proativas, produtivas e prazerosas, aumentando as chances de cooperarem com as medidas sanitĂĄrias ao ficarem em casa.Palavras-chave: Ativação Comportamental; Quarentena; Pandemia; AnĂĄlise do Comportamento.  Quarantine and social isolation are prophylactic health measures to delay the advance of pandemics such as COVID-19. However, these measures can negatively impact people’s mental health, making it difficult for them to adhere to the recommendations of health agencies.     Italian real life experience with ibrutinib: Results of a large observational study on 77 relapsed/refractory mantle cell lymphoma

    Although sometimes presenting as an indolent lymphoma, mantle cell lymphoma (MCL) is an aggressive disease, hardly curable with standard chemo-immunotherapy. Current approaches have greatly improved patients' outcomes, nevertheless the disease is still characterized by high relapse rates. Before approval by EMA, Italian patients with relapsed/refractory MCL were granted ibrutinib early access through a Named Patient Program (NPP). An observational, retrospective, multicenter study was conducted. Seventyseven heavily pretreated patients were enrolled. At the end of therapy there were 14 complete responses and 14 partial responses, leading to an overall response rate of 36.4%. At 40 months overall survival was 37.8% and progression free survival was 30%; disease free survival was 78.6% at 4 years: 11/14 patients are in continuous complete response with a median of 36 months of follow up. Hematological toxicities were manageable, and main extra-hematological toxicities were diarrhea (9.4%) and lung infections (9.0%). Overall, 4 (5.2%) atrial fibrillations and 3 (3.9%) hemorrhagic syndromes occurred. In conclusions, thrombocytopenia, diarrhea and lung infections are the relevant adverse events to be clinically focused on; regarding effectiveness, ibrutinib is confirmed to be a valid option for refractory/relapsed MCL also in a clinical setting mimicking the real world

    Mitochondrial Pathway Mediates the Antileukemic Effects of Hemidesmus Indicus, a Promising Botanical Drug

    Although cancers are characterized by the deregulation of multiple signalling pathways, most current anticancer therapies involve the modulation of a single target. Because of the enormous biological diversity of cancer, strategic combination of agents targeted against the most critical of those alterations is needed. Due to their complex nature, plant products interact with numerous targets and influence several biochemical and molecular cascades. The interest in further development of botanical drugs has been increasing steadily and the FDA recently approved the first new botanical prescription drug. The present study is designed to explore the potential antileukemic properties of Hemidesmus indicus with a view to contributing to further development of botanical drugs. Hemidesmus was submitted to an extensive in vitro preclinical evaluation.A variety of cellular assays and flow cytometry, as well as a phytochemical screening, were performed on different leukemic cell lines. We have demonstrated that Hemidesmus modulated many components of intracellular signaling pathways involved in cell viability and proliferation and altered the protein expression, eventually leading to tumor cell death, mediated by a loss of mitochondrial transmembrane potential and increased Bax/Bcl-2 ratio. ADP, adenine nucleotide translocator and mitochondrial permeability transition pore inhibitors did not reverse Hemidesmus-induced mitochondrial depolarization. Hemidesmus induced a significant [Ca(2+)](i) raise through the mobilization of intracellular Ca(2+) stores. Moreover, Hemidesmus significantly enhanced the antitumor activity of three commonly used chemotherapeutic drugs (methotrexate, 6-thioguanine, cytarabine). A clinically relevant observation is that its cytotoxic activity was also recorded in primary cells from acute myeloid leukemic patients.These results indicate the molecular basis of the antileukemic effects of Hemidesmus and identify the mitochondrial pathways and [Ca(2+)](i) as crucial actors in its anticancer activity. On these bases, we conclude that Hemidesmus can represent a valuable tool in the anticancer pharmacology, and should be considered for further investigations

    A Genetic Animal Model of Alcoholism for Screening Medications to Treat Addiction

    The purpose of this review is to present up-to-date pharmacological, genetic, and behavioral findings from the alcohol-preferring P rat and summarize similar past work. Behaviorally, the focus will be on how the P rat meets criteria put forth for a valid animal model of alcoholism with a highlight on its use as an animal model of polysubstance abuse, including alcohol, nicotine, and psychostimulants. Pharmacologically and genetically, the focus will be on the neurotransmitter and neuropeptide systems that have received the most attention: cholinergic, dopaminergic, GABAergic, glutamatergic, serotonergic, noradrenergic, corticotrophin releasing hormone, opioid, and neuropeptide Y. Herein, we sought to place the P rat's behavioral and neurochemical phenotypes, and to some extent its genotype, in the context of the clinical literature. After reviewing the findings thus far, this chapter discusses future directions for expanding the use of this genetic animal model of alcoholism to identify molecular targets for treating drug addiction in general

    The production of tt‟ , W+bb‟ and W+cc‟ is studied in the forward region of proton–proton collisions collected at a centre-of-mass energy of 8 TeV by the LHCb experiment, corresponding to an integrated luminosity of 1.98±0.02 fb−1 . The W bosons are reconstructed in the decays W→ℓΜ , where ℓ denotes muon or electron, while the b and c quarks are reconstructed as jets.     Multidifferential study of identified charged hadron distributions in ZZ-tagged jets in proton-proton collisions at s=\sqrt{s}=13 TeV

    Jet fragmentation functions are measured for the first time in proton-proton collisions for charged pions, kaons, and protons within jets recoiling against a ZZ boson. The charged-hadron distributions are studied longitudinally and transversely to the jet direction for jets with transverse momentum 20 <pT<100< p_{\textrm{T}} < 100 GeV and in the pseudorapidity range 2.5<η<42.5 < \eta < 4. The data sample was collected with the LHCb experiment at a center-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 1.64 fb−1^{-1}. Triple differential distributions as a function of the hadron longitudinal momentum fraction, hadron transverse momentum, and jet transverse momentum are also measured for the first time. This helps constrain transverse-momentum-dependent fragmentation functions.
