Design of a Robot Prototype with Rocker Geometry-Bogie

In this research a Rocker-Bogie type suspension geometry is designed and validated based on the analysis of the degrees of freedom, for its future implementation within the space exploration industry sector, having as main purpose the collection and transportation of garbage and waste to strategic sectors within the facilities, avoiding obstacles of different levels, although it could also be applied in the industrial, agricultural and mining areas, where a robust and stable system is required. We start by establishing a safety factor, determined through coefficients that involve safety and economy, then we calculate the dimensions and forces to be supported by the structural elements known as Rocker and Bogie, which are responsible for transmitting the support to the chassis as well as providing support to the connections for the axle components and motors for the wheels. The design and calculation of these elements, depends on the robot prototype being able to move through irregular fields avoiding obstacles such as: surface anomalies, potholes, rocks and curbs. For the validation of the prototype design, a static analysis of the structural elements (Rocker and Bogie) is performed by means of the Von Misses parameter, obtaining a high level of safety before rupture. Subsequently, the simulation of the prototype is executed based on a track created in the software, analyzing parameters of speed, acceleration and displacements in the axes X, Y. Finally, the results obtained from the simulation are compared based mainly on height and maximum length between front and rear axles, being crucial the maximum permissible angle of inclination

Maintenance of ancient road and railway metallic bridges has become a major concern for governmental agencies in the past few decades. Indeed, since the construction of these structures, between the end of the 19th century and the beginning of the 20th century, traffic conditions have evolved, both in weight and frequency. In the purpose to assess the remaining life of old metallic bridges, some critical structural details have been identified and associated to S-N curves in order to be used in damage estimation (using Palmgren-Miner’s rule for cumulative damage, for example). These constructional details are described by design rules of several European and North American standards, such as the Eurocode 3, BS 5400 and AASHTO standards. The particularity of ancient bridges is that hot riveted assemblies, commonly used for their construction, are not represented in most construction standards. Further experiences on the matter by numerous research teams have suggested detail category C71 from the Eurocode 3 as appropriate. In this paper, experimental data from double shear assemblies manufactures from three different metallic ancient bridges is used to identify, through a statistical analysis, the S-N curves that best fit this constructional detail. Portuguese and French puddled iron bridges were considered. The authors of this paper thank the National Society of French Railways and the SciTech - Science and Technology for Competitive and Sustainable Industries, R&D project NORTE-01-0145-FEDER-000022 cofinanced by Programa Operacional Regional do Norte ("NORTE2020"), through Fundo Europeu de Desenvolvimento Regional (FEDER) for their collaboration and support during this research works.

An Influenza A/H1N1/2009 Hemagglutinin Vaccine Produced in Escherichia coli

The A/H1N1/2009 influenza pandemic made evident the need for faster and higher-yield methods for the production of influenza vaccines. Platforms based on virus culture in mammalian or insect cells are currently under investigation. Alternatively, expression of fragments of the hemagglutinin (HA) protein in prokaryotic systems can potentially be the most efficacious strategy for the manufacture of large quantities of influenza vaccine in a short period of time. Despite experimental evidence on the immunogenic potential of HA protein constructs expressed in bacteria, it is still generally accepted that glycosylation should be a requirement for vaccine efficacy. We expressed the globular HA receptor binding domain, referred to here as HA(63-286)-RBD, of the influenza A/H1N1/2009 virus in Escherichia coli using a simple, robust and scalable process. The recombinant protein was refolded and purified from the insoluble fraction of the cellular lysate as a single species. Recombinant HA(63-286)-RBD appears to be properly folded, as shown by analytical ultracentrifugation and bio-recognition assays. It binds specifically to serum antibodies from influenza A/H1N1/2009 patients and was found to be immunogenic, to be capable of triggering the production of neutralizing antibodies, and to have protective activity in the ferret model.Projections based on our production/purification data indicate that this strategy could yield up to half a billion doses of vaccine per month in a medium-scale pharmaceutical production facility equipped for bacterial culture. Also, our findings demonstrate that glycosylation is not a mandatory requirement for influenza vaccine efficacy

    States Of Discontent

Latin America’s recent inclusionary turn centers on changing relationships between the popular sectors and the state. Yet the new inclusion unfolds in a region in which most states are weak and prone to severe pathologies, such as corruption, inefficiency, and particularism. The first part of the chapter outlines an argument, developed at more length elsewhere, regarding how “state crises” helped drive the consolidation of three distinct party system trajectories among the eight South American countries where the Left would eventually win power. The second part of the chapter argues that these trajectories differed in three ways that likely conditioned how the concomitant inclusionary Left turn unfolded in each case: the institutionalization of left-wing parties, the occurrence of state transformation via constitutional reform, and the level of state capacity. The discussion helps highlight the central role of the state and its pathologies in both driving alternative paths of political development and in conditioning the politics of inclusion. By putting the emphasis on the state and its pathologies, we can better consider not just the sources of sociopolitical exclusion but also the limits of sociopolitical inclusion

Sperm DNA fragmentation: A new guideline for clinicians

    Get PDF
Sperm DNA integrity is crucial for fertilization and development of healthy offspring. The spermatozoon undergoes extensive molecular remodeling of its nucleus during later phases of spermatogenesis, which imparts compaction and protects the genetic content. Testicular (defective maturation and abortive apoptosis) and post-testicular (oxidative stress) mechanisms are implicated in the etiology of sperm DNA fragmentation (SDF), which affects both natural and assisted reproduction. Several clinical and environmental factors are known to negatively impact sperm DNA integrity. An increasing number of reports emphasizes the direct relationship between sperm DNA damage and male infertility. Currently, several assays are available to assess sperm DNA damage, however, routine assessment of SDF in clinical practice is not recommended by professional organizations

Numerous metallic bridges can be found in the French railway heritage. Since a majority of these structures was built during the second half of the 19th century, they have been in service for over a hundred years and need an adapted maintenance policy. These doctoral thesis, done in collaboration with SNCF Réseau, improves the understanding of the mechanical behaviour of hot riveted assemblies under monotonic and cyclic loadings. An experimental strategy was proposed and carried out in order to identify the failure mechanisms of riveted assemblies. First, the studied material, a puddled iron, was obtained from recovered elements from the bridge over the Adour river in Bayonne (1864-2013). Mechanical testing and characterisation was performed under monotonic and cyclic loadings. Puddled iron’s brittle, anisotropic and scattered behaviour was observed and analysed. In-house software applications were developed in order to study this kind of material. Additionally, hot riveted specimens (constructional details) were designed, manufactured and tested under monotonic and cyclic loadings. A precise working (and failure) scenario of the double shear mono-rivet assembly was identified. The influence of the material’s behaviour, stress concentrators, shearing of the rivet and clamping force of the rivet were studied. The results obtained during these research works, compared to the ones found in academic literature as well as construction norms for metallic structures, showed the importance and complementarity of a multi-scale approach (material behaviour and local study of assemblies) in order to understand and assess the behaviour of ancient riveted structures.
