    Nanoantennas Inversely Designed to Couple Free Space and a Metal–Insulator–Metal Waveguide

    The metal–insulator–metal (MIM) waveguide, which can directly couple free space photons, acts as an important interface between conventional optics and subwavelength photoelectrons. The reason for the difficulty of this optical coupling is the mismatch between the large wave vector of the MIM plasmon mode and photons. With the increase in the wave vector, there is an increase in the field and Ohmic losses of the metal layer, and the strength of the MIM mode decreases accordingly. To solve those problems, this paper reports on inversely designed nanoantennas that can couple the free space and MIM waveguide and efficiently excite the MIM plasmon modes at multiple wavelengths and under oblique angles. This was achieved by implementing an inverse design procedure using a topology optimization approach. Simulation analysis shows that the coupling efficiency is enhanced 9.47-fold by the nanoantenna at the incident wavelength of 1338 nm. The topology optimization problem of the nanoantennas was analyzed by using a continuous adjoint method. The nanoantennas can be inversely designed with decreased dependence on the wavelength and oblique angle of the incident waves. A nanostructured interface on the subwavelength scale can be configured in order to control the refraction of a photonic wave, where the periodic unit of the interface is composed of two inversely designed nanoantennas that are decoupled and connected by an MIM waveguide

    Silica nanoparticles enhance autophagic activity, disturb endothelial cell homeostasis and impair angiogenesis

    BACKGROUND: Given that the effects of ultrafine fractions (<0.1 μm) on ischemic heart diseases (IHD) and other cardiovascular diseases are gaining attention, this study is aimed to explore the influence of silica nanoparticles (SiNPs)-induced autophagy on endothelial cell homeostasis and angiogenesis. METHODS AND RESULTS: Ultrastructural changes of autophagy were observed in both vascular endothelial cells and pericytes in the heart of ICR mice by TEM. Autophagic activity and impaired angiogenesis were further confirmed by the immunohistochemistry staining of LC3 and VEGFR2. In addition, the immunohistochemistry results showed that SiNPs had an inhibitory effect on ICAM-1 and VCAM-1, but no obvious effect on E-selectin in vivo. The disruption of F-actin cytoskeleton occurred as an initial event in SiNPs-treated endothelial cells. The depolarized mitochondria, autophagic vacuole accumulation, LC3-I/LC3-II conversion, and the down-regulation of cellular adhesion molecule expression were all involved in the disruption of endothelial cell homeostasis in vitro. Western blot analysis indicated that the VEGFR2/PI3K/Akt/mTOR and VEGFR2/MAPK/Erk1/2/mTOR signaling pathway was involved in the cardiovascular toxicity triggered by SiNPs. Moreover, there was a crosstalk between the VEGFR2-mediated autophagy signaling and angiogenesis signaling pathways. CONCLUSIONS: In summary, the results demonstrate that SiNPs induce autophagic activity in endothelial cells and pericytes, subsequently disturb the endothelial cell homeostasis and impair angiogenesis. The VEGFR2-mediated autophagy pathway may play a critical role in maintaining endothelium and vascular homeostasis. Our findings may provide experimental evidence and explanation for cardiovascular diseases triggered by nano-sized particles.     Toxic Effects of Silica Nanoparticles on Zebrafish Embryos and Larvae

    Silica nanoparticles (SiNPs) have been widely used in biomedical and biotechnological applications. Environmental exposure to nanomaterials is inevitable as they become part of our daily life. Therefore, it is necessary to investigate the possible toxic effects of SiNPs exposure. In this study, zebrafish embryos were treated with SiNPs (25, 50, 100, 200 μg/mL) during 4-96 hours post fertilization (hpf). Mortality, hatching rate, malformation and whole-embryo cellular death were detected. We also measured the larval behavior to analyze whether SiNPs had adverse effects on larvae locomotor activity. The results showed that as the exposure dosages increasing, the hatching rate of zebrafish embryos was decreased while the mortality and cell death were increased. Exposure to SiNPs caused embryonic malformations, including pericardial edema, yolk sac edema, tail and head malformation. The larval behavior testing showed that the total swimming distance was decreased in a dose-dependent manner. The lower dose (25 and 50 μg/mL SiNPs) produced substantial hyperactivity while the higher doses (100 and 200 μg/mL SiNPs) elicited remarkably hypoactivity in dark periods. In summary, our data indicated that SiNPs caused embryonic developmental toxicity, resulted in persistent effects on larval behavior. © 2013 Duan et al.published_or_final_versio

    Assessing the relationship between levator palpebrae superioris and thyroid-associated ophthalmopathy using the Dixon-T2WI sequence

    BackgroundThe current clinical practice lacks sufficient objective indicators for evaluating thyroid-associated ophthalmopathy (TAO). This study aims to quantitatively assess TAO by evaluating levator palpebrae superioris (LPS) using Dixon-T2WI.MethodsThe retrospective study included 231 eyes (119 patients) in the TAO group and 78 eyes (39 volunteers) in the normal group. Dixon-T2WI provided data on maximum thickness of LPS (LPS_T) and signal intensity ratio (LPS_SIR) between the muscle and ipsilateral brain white matter. TAO diagnosis and assessment of its activity and severity were quantitatively determined using LPS_T and LPS_SIR.ResultsIn the TAO group, LPS_T and LPS_SIR were higher than those in the normal group (p &lt; 2.2e-16). The upper lid retraction (ULR) ≥ 2 mm group exhibited higher LPS_T and LPS_SIR compared to the ULR &lt; 2 mm and normal groups. Optimal diagnostic performance was achieved with an AUC of 0.91 for LPS_T (cutoff: 1.505 mm) and 0.81 for LPS_SIR (cutoff: 1.170). LPS_T (p = 2.8e-07) and LPS_SIR (p = 3.9e-12) in the active phase were higher than in the inactive phase. LPS_T and LPS_SIR showed differences among the mild, moderate-to-severe, and sight-threatening groups (p &lt; 0.05). ROC showed an AUC of 0.70 for LPS_T (cutoff: 2.095 mm) in judging the active phase, and 0.78 for LPS_SIR (cutoff: 1.129). For judging the moderate-to-severe and above, AUC was 0.76 for LPS_T (cutoff: 2.095 mm) and 0.78 for LPS_SIR (cutoff: 1.197).ConclusionThe maximum thickness and SIR of LPS provide imaging indicators for assisting in the diagnosis and quantitative evaluation of TAO

    Mapping QTLs for the tissue culture performance of rice mature embryo using indica-japonica recombinant inbred lines

    Abstract The tissue culture performance is the determinant factor of genetic transformation in indica rice (Oryza sativa L.). Quantitative trait loci (QTLs) associated with the tissue culture performance of mature embryo were detected by 202 genetic markers and 190 recombinant inbred lines (RILs) derived from the cross between two rice varieties, 93-11 (indica) as female parent and Nipponbare (japonica) as male parent. A composite interval mapping (CIM) was used to identify the QTLs for the tissue culture performance. The tissue culture performance of rice mature embryo were evaluated by six parameters, embryogenic calli induction rate (ECR), callus subculture capability (CSC), plantlet regeneration rate (RR), average number of regenerated shoots per callus (NRS), green plantlet regeneration rate (GRR) and average number of regenerated green shoots per callus (NRGS). Nipponbare has better tissue culture response than 93-11 because all indicators were higher in Nipponbare except CSC. Four QTLs for ECR, five QTLs for CSC, three QTLs for RR, two QTLs for NRS, four QTLs for GRR and three QTLs for NRGS were detected. These putative QTLs associated with tissue culture performance were distributed on eight rice chromosomes. These results demonstrated the possibilities of improving the tissue culture performance of indica rice by marker assisted selection (MAS) with those desirable alleles of japonica rice.     A RG-II type polysaccharide purified from Aconitum coreanum and their anti-inflammatory activity

    Korean mondshood root polysaccharides (KMPS) isolated from the root of Aconitum coreanum (Lévl.) Rapaics have shown anti-inflammatory activity, which is strongly influenced by their chemical structures and chain conformations. However, the mechanisms of the anti-inflammatory effect by these polysaccharides have yet to be elucidated. A RG-II polysaccharide (KMPS-2E, Mw 84.8 kDa) was isolated from KMPS and its chemical structure was characterized by FT-IR and NMR spectroscopy, gas chromatography–mass spectrometry and high-performance liquid chromatography. The backbone of KMPS-2E consisted of units of [→6) -β-D-Galp (1→3)-β-L-Rhap-(1→4)-β-D-GalpA-(1→3)-β-D-Galp-(1→] with the side chain →5)-β-D-Arap (1→3, 5)-β-D-Arap (1→ attached to the backbone through O-4 of (1→3,4)-L-Rhap. T-β-D-Galp is attached to the backbone through O-6 of (1→3,6)-β-D-Galp residues and T-β-D-Ara is connected to the end group of each chain. The anti-inflammatory effects of KMPS-2E and the underlying mechanisms using lipopolysaccharide (LPS) - stimulated RAW 264.7 macrophages and carrageenan-induced hind paw edema were investigated. KMPS-2E (50, 100 and 200 µg/mL) inhibits iNOS, TLR4, phospho-NF-κB–p65 expression, phosphor-IKK, phosphor-IκB-α expression as well as the degradation of IκB-α and the gene expression of inflammatory cytokines (TNF-α, IL-1β, iNOS and IL-6) mediated by the NF-κB signal pathways in macrophages. KMPS-2E also inhibited LPS-induced activation of NF-κB as assayed by electrophorectic mobility shift assay (EMSA) in a dose-dependent manner and it reduced NF-κB DNA binding affinity by 62.1% at 200µg/mL. In rats, KMPS-2E (200 mg/kg) can significantly inhibit carrageenan-induced paw edema as ibuprofen (200 mg/kg) within 3 h after a single oral dose. The results indicate that KMPS-2E is a promising herb-derived drug against acute inflammation

    Large expert-curated database for benchmarking document similarity detection in biomedical literature search

    Document recommendation systems for locating relevant literature have mostly relied on methods developed a decade ago. This is largely due to the lack of a large offline gold-standard benchmark of relevant documents that cover a variety of research fields such that newly developed literature search techniques can be compared, improved and translated into practice. To overcome this bottleneck, we have established the RElevant LIterature SearcH consortium consisting of more than 1500 scientists from 84 countries, who have collectively annotated the relevance of over 180 000 PubMed-listed articles with regard to their respective seed (input) article/s. The majority of annotations were contributed by highly experienced, original authors of the seed articles. The collected data cover 76% of all unique PubMed Medical Subject Headings descriptors. No systematic biases were observed across different experience levels, research fields or time spent on annotations. More importantly, annotations of the same document pairs contributed by different scientists were highly concordant. We further show that the three representative baseline methods used to generate recommended articles for evaluation (Okapi Best Matching 25, Term Frequency-Inverse Document Frequency and PubMed Related Articles) had similar overall performances. Additionally, we found that these methods each tend to produce distinct collections of recommended articles, suggesting that a hybrid method may be required to completely capture all relevant articles. The established database server located at https://relishdb.ict.griffith.edu.au is freely available for the downloading of annotation data and the blind testing of new methods. We expect that this benchmark will be useful for stimulating the development of new powerful techniques for title and title/abstract-based search engines for relevant articles in biomedical research.Peer reviewe

    The Effects of the Relationships Between Object Features on Multiple-Identity Tracking

    Simultaneous tracking of multiple moving objects is essential in tasks such as traffic control, automobile driving, and scene surveillance. Recently, an increasing number of studies have focused on the roles of object identity and location binding in unique target tracking tasks, but contradictory results have been reported. In the present study, we introduced for the first time the Stroop stimuli of Chinese characters to the multiple-identity tracking paradigm, taking advantage of the ease to control the overall size, familiarity, and visual complexity of the Chinese characters. The results showed that when the observers were asked to track unique objects that bear two distinct features, the feature conflict disrupted the tracking performance, even when the given object has a distinctive identity. Our data revealed that the internal disaccord of semantic and physical features on an object can disturb the identity-location binding process in a multiple-identity tracking task, but does not affect the location information significantly
