    Notas sobre las algas marinas de la Reserva Internacional de la Biosfera Seaflower, Caribe Colombiano IX: nuevos registros de Lobophora declerckii, Taonia abbottiana y Verosphacela ebrachia para Colombia

    Background and Aims: During the Seaflower Scientific Expedition in 2019, extensive seaweed collection was carried out around Old Providence and Santa Catalina islands. Despite the intensive phycological studies realized during the past decade, new reports of macroalgae are still recorded for the Seaflower Biosphere Reserve. The objective of this work is to present three new records of brown algae for Colombia.Methods: The algal sampling was carried out in September 2019 during the Seaflower Scientific Expedition. The samples were collected around the Providencia and Santa Catalina islands in coral patches ecosystems, by SCUBA diving at depths between 3 and 16 m. The algae collected were preserved in a 4% formalin/seawater solution. The identification was carried out using optical and stereoscopic microscopes. All specimens were deposited in the herbarium JIW of the Biology Department of the Universidad Nacional de Colombia, Bogotá, Colombia. Key results: Three species of brown algae are newly reported for Colombia: Lobophora declerckii, Taonia abbottiana and Verosphacela ebrachia. Verosphacela has been reported, to date, only from the type locality in Florida and from Isla Mujeres, in Mexico. Due to its cryptic and small habit, it may be much more common than actually reported. Lobophora declerckii is apparently the most common species of Lobophora in the Archipelago. Taonia abbottiana may be much more common than reported to date, but it is easily confused with Stypopodium zonale. Conclusions: Despite the intense phycological research carried out in the past decade in the Archipelago of San Andrés, Old Providence and Santa Catalina, there is still much to be discovered.Antecedentes y Objetivos: Durante la Expedición Científica Seaflower realizada en 2019, se hizo una extensa recolecta de macroalgas alrededor de las islas de Providencia y Santa Catalina. No obstante el intenso trabajo ficológico de la última década, se siguen encontrando nuevos registros de macroalgas para la Reserva de la Biosfera Seaflower. El objetivo del presente trabajo es presentar tres nuevos registros de algas pardas para Colombia.Metodos: El muestreo de algas se realizó en septiembre de 2019 durante la Expedición Científica Seaflower. Las muestras fueron recolectadas alrededor de las islas Providencia y Santa Catalina en ecosistemas de parches de coral, por medio de buceo SCUBA a profundidades entre 3 y 16 m. Las algas recolectadas se conservaron en una solución de formalina/agua de mar al 4%. La identificación se realizó mediante microscopios óptico y estereoscopio. Todos los especímenes fueron depositados en el herbario JIW del Departamento de Biología de la Universidad Nacional de Colombia, en Bogotá, Colombia. Resultados clave: Tres especies de algas pardas son reportadas por primera vez para Colombia: Lobophora declerckii, Taonia abbottiana y Verosphacela ebrachia. Verosphacela ha sido reportada, hasta la fecha, solamente en la localidad tipo en Florida y en Isla Mujeres, en México. Debido a su hábito pequeño y críptico, puede ser mucho más común de lo actualmente reportado. Lobophora declerckii parece ser la especie más común de Lobophora en el Archipiélago. Taonia abbottiana puede ser mucho más común de lo reportado actualmente, pero es fácilmente confundida con Stypopodium zonale.     Reptile remains from Tiga (Tokanod), Loyalty Islands, New Caledonia

    Archaeological excavations on Tiga provide the first vouchered herpetological records for this small island between Lifou and Maré in the Loyalty Islands. Eighty-three skeletal elements from four sites yielded material assignable to skinks (Emoia loyaltiensis, Lioscincus nigrofasciolatus), geckos (Bavayia crass i-collis, B. sp., Gehyra georgpotthasti, Nactus pelagicus), and a boid snake (Candoia bihroni) all known from elsewhere in the Loyalties, as well as undetermined material consistent with these and other Loyalties lizards. Diagnostic features of geckos versus skinks for elements commonly recovered from archaeological sites and from owl pellets are discussed. Gehyra georgpotthasti has a limited distribution in the Loyalties and its occurrence on Tiga clarifies its range. The boid snake is the only reptile likely to have been harvested by human inhabitants of Tiga. The presence of gekkonid geckos in pre-European times is confirmed and contrasts with the situation of Grande Terre fossil sites, where only diplodactylid geckos have been recovered. Although it is anticipated that all species recovered from archaeological sites are still present on the island, a modern herpetofaunal survey is needed

    Procjena cito-/genotoksičnosti irinotekana u V79-stanicama primjenom komet-testa, mikronukleus-testa i testa kromosomskih aberacija

    Irinotecan is a topoisomerase I interactive agent, widely used in the treatment of metastatic colorectal cancer. The genotoxic effects of the maximum single dose (18 μg mL-1), recommended monotherapy dose (9 μg mL-1), and recommended combined therapy dose (4.5 μg mL-1) of irinotecan were studied on V79 cells using the comet assay, chromosome aberration assay, and micronucleus test. The cells were treated with irinotecan for 2 h or 24 h. The statistical signifi cance of the results was determined using the one-way ANOVA test and a nonparametric Mann Whitney U test. The comet assay did not show dose-dependent or time-dependent effects. The chromosome aberration analysis showed large DNA rearrangements, i.e., chromosome exchanges. Although the exposed cultures showed a signifi cant increase in micronucleated cells in respect to control, no dose-dependent relation was established among the treated cultures. Timedependent effect was also not observed.Irinotekan je citotoksični lijek koji inhibira enzim DNA-topoizomerazu I. U širokoj je primjeni u terapiji metastatskog karcinoma kolona i rektuma. U uvjetima in vitro primjenom komet-testa, analize kromosomskih aberacija i mikronukleus-testa na V79-stanicama istražili smo genotoksični učinak maksimalne pojedinačne doze (18 μg mL-1), preporučene monoterapijske doze (9 μg mL-1) i preporučene doze irinotekana za kombiniranu terapiju (4,5 μg mL-1). Kulture stanica bile su tretirane irinotekanom 2 h i 24 h. Statistička značajnost određivana je jednosmjernim ANOVA-testom i neparametrijskim Mann Whitneyevim U-testom. Komet-testom nije utvrđen učinak koncentracije i/ili vremena izloženosti. Analiza kromosomskih aberacija pokazala je prisutnost izmjena kromatida, tj. porast broja triradijusa i tetraradijusa. Iako je u kulturama stanica izloženi irinotekanu opažen značajan porast broja mikronukleusa u odnosu na kontrolu, nije uočena ovisnost o dozi lijeka ni o vremenu izloženosti u opisanim eksperimentalnim uvjetima. Dobiveni rezultati upućuju na genotoksičnost irinotekana za V79-stanice. Nijednom od primijenjenih metoda nije utvrđena ovisnost učinka irinotekana o vremenu ili dozi

    Natural Killer Cells from Patients with Recombinase-Activating Gene and Non-Homologous End Joining Gene Defects Comprise a Higher Frequency of CD56bright NKG2A+++ Cells, and Yet Display Increased Degranulation and Higher Perforin Content.

    Mutations of the recombinase Activating Genes 1 and 2 (RAG1, RAG2) in humans are associated with a broad range of phenotypes. For patients with severe clinical presentation, hematopoietic stem cell transplantation (HSCT) represents the only curative treatment, however high rates of graft failure and incomplete immune reconstitution have been observed, especially after unconditioned haploidentical transplantation. Studies in mice have shown that Rag-/- NK cells have a mature phenotype, reduced fitness and increased cytotoxicity. We aimed to analyze NK cell phenotype and function in patients with mutations in RAG and in non-homologous end joining (NHEJ) genes. Here we provide evidence that NK cells from these patients have an immature phenotype, with significant expansion of CD56bright CD16-/int CD57- cells, yet increased degranulation and high perforin content. Correlation was observed between in vitro recombinase activity of the mutant proteins, NK cell abnormalities, and in vivo clinical phenotype. Addition of serotherapy in the conditioning regimen, with the aim of depleting the autologous NK cell compartment, may be important to facilitate engraftment and immune reconstitution in patients with RAG and NHEJ defects treated by HSCT

    Objectives: The COVID-19 pandemic has resulted in widespread morbidity and mortality with the consequences expected to be felt for many years. Significant variation exists in the care even of similar patients with COVID-19, including treatment practices within and between institutions. Outcome measures vary among clinical trials on the same therapies. Understanding which therapies are of most value is not possible unless consensus can be reached on which outcomes are most important to measure. Furthermore, consensus on the most important outcomes may enable patients to monitor and track their care, and may help providers to improve the care they offer through quality improvement. To develop a standardised minimum set of outcomes for clinical care, the International Consortium for Health Outcomes Measurement (ICHOM) assembled a working group (WG) of 28 volunteers, including health professionals, patients and patient representatives. Design: A list of outcomes important to patients and professionals was generated from a systematic review of the published literature using the MEDLINE database, from review of outcomes being measured in ongoing clinical trials, from a survey distributed to patients and patient networks, and from previously published ICHOM standard sets in other disease areas. Using an online-modified Delphi process, the WG selected outcomes of greatest importance. Results: The outcomes considered by the WG to be most important were selected and categorised into five domains: (1) functional status and quality of life, (2) mental functioning, (3) social functioning, (4) clinical outcomes and (5) symptoms. The WG identified demographic and clinical variables for use as case-mix risk adjusters. These included baseline demographics, clinical factors and treatment-related factors. Conclusion: Implementation of these consensus recommendations could help institutions to monitor, compare and improve the quality and delivery of care to patients with COVID-19. Their consistent definition and collection could also broaden the implementation of more patient-centric clinical outcomes research.</p

    MPC with Friends and Foes

    Classical definitions for secure multiparty computation assume the existence of a single adversarial entity controlling the set of corrupted parties. Intuitively, the definition requires that the view of the adversary, corrupting tt parties, in a real-world execution can be simulated by an adversary in an ideal model, where parties interact only via a trusted-party. No restrictions, however, are imposed on the view of honest parties in the protocol, thus, if honest parties obtain information about the private inputs of other honest parties -- it is not counted as a violation of privacy. This is arguably undesirable in many situations that fall into the MPC framework. Nevertheless, there are secure protocols (e.g., the 2-round multiparty protocol of Ishai et al.~[CRYPTO 2010] tolerating a single corrupted party) that instruct the honest parties to reveal their private inputs to all other honest parties (once the malicious party is somehow identified). In this paper, we put forth a new security notion, which we call \textit{FaF-security}, extending the classical notion. In essence, (t,h)(t,h^*)-FaF-security requires the view of a subset of up to hh^* honest parties to also be simulatable in the ideal model (in addition to the view of the malicious adversary, corrupting up to tt parties). This property should still hold, even if the adversary leaks information to honest parties by sending them non-prescribed messages. We provide a thorough exploration of the new notion, investigating it in relation to a variety of existing security notions. We further investigate the feasibility of achieving FaF-security and show that every functionality can be computed with (computational) (t,h)(t,h^*)-FaF full-security, if and only if 2t+h<m2t+ h^*<m. Interestingly, the lower-bound result actually shows that even fair FaF-security is impossible in general when 2t+hm2t+ h^*\ge m (surprisingly, the view of the malicious attacker is not used as the trigger for the attack). We also investigate the optimal round complexity for (t,h)(t,h^*)-FaF-secure protocols and give evidence that the leakage of private inputs of honest parties in the protocol of Ishai et al.~[CRYPTO 2010] is inherent. Finally, we investigate the feasibility of statistical/perfect FaF-security, employing the viewpoint used by Fitzi et al.~[ASIACRYPT 1999] for \textit{mixed-adversaries}

    An Ontological Approach to Inform HMI Designs for Minimizing Driver Distractions with ADAS

    ADAS (Advanced Driver Assistance Systems) are in-vehicle systems designed to enhance driving safety and efficiency as well as comfort for drivers in the driving process. Recent studies have noticed that when Human Machine Interface (HMI) is not designed properly, an ADAS can cause distraction which would affect its usage and even lead to safety issues. Current understanding of these issues is limited to the context-dependent nature of such systems. This paper reports the development of a holistic conceptualisation of how drivers interact with ADAS and how such interaction could lead to potential distraction. This is done taking an ontological approach to contextualise the potential distraction, driving tasks and user interactions centred on the use of ADAS. Example scenarios are also given to demonstrate how the developed ontology can be used to deduce rules for identifying distraction from ADAS and informing future designs

    Guidelines for the use and interpretation of assays for monitoring autophagy (3rd edition)

    In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure fl ux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defi ned as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (inmost higher eukaryotes and some protists such as Dictyostelium ) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the fi eld understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation it is imperative to delete or knock down more than one autophagy-related gene. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways so not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field
