    Microplastics: Nacogdoches County’s Hidden Threat

    Microplastics and their threat to marine environments are well known, but studies in freshwater environments are few and far between. Microplastics can be created by the breaking down of plastics or in the process of making plastics. They affect biological lifeforms in a multitude of ways, water temperature, permeability of the sediment. Pollution in Texas waterways have motivated groups like the Environmental Protection Agency, Trash Free Texas, and Environment Texas to get involved. The objectives of this study are two analyze Bonita and La Nana Bayou for microplastic presence and to educate and create awareness for Nacogdoches County on microplastics. This study uses the Assessment of Microplastic in Great Plains: Comparing densities in water and benthic sediment across Kansas as a guiding reference. Samples were filtered through a 300 μm mesh sieve, rinsed and fixed with 15mL of 0.1N KOH for 14 days. Microplastics were found in each sample. Data was analyzed using ANOVA Single Factor Test to confirm the hypothesis that the sample sites will not equal one another. Future studies should include more samples and experience

    Earthworms and mesofauna from an isolated, alkaline chemical waste site in Northwest England

    Post-industrial sites across Europe may have developed over periods in excess of a century, often leading to poor nutrient soils with noxious constituents and extreme pH values. One such site, Nob End (Bolton, UK), a toxic tip created in the 1800s from a sodium carbonate factory, represents an “island of alkalinity in an acidic sea” where the weathering process (from pH 12) has provided a suitable environment for a rich alkali-loving flora and hence, deserving its designation as a Site of Special Scientific Interest. Despite their importance, the belowground communities have not been investigated and for this reason, in this study, we explored how soil macro- and mesofauna communities respond to extreme pH values in a system that has also experienced recent changes in management practices. As expected, earthworms, mites, insects and woodlice numbers were significantly higher and the community diversity enriched at the (now pH 8) alkaline sites, whereas in areas where acidic boiler waste was historically deposited, enchytraeids, collembolans and dipteran larvae populations dominated the soil communities. Surprisingly, site management (cutting back of scrub) in the alkaline soil areas had a significant positive effect on soil macro-fauna by promoting numbers and biomass, but severely reduced the microarthropod populations. A transect investigation across an increasing pH gradient (from 4.5 to 8.0) was mirrored by a rise in earthworm numbers and species richness. Earthworms were further investigated surrounding the site, seeking potential sources of colonisation, with the majority of species at Nob End also present in adjacent non-industrially-influenced areas. This work demonstrates that soil fauna can ultimately colonise extreme edaphic conditions and these extreme environments have not prompted the development of specific faunal communities. As management of above-ground communities significantly influenced soil invertebrate communities, this could represent an important restoration practice to improve soil structure and fertility at this polluted site

    TU Tau B: The Peculiar 'Eclipse' of a possible proto-Barium Giant

    TU Tau (= HD 38218 = HIP 27135) is a binary system consisting of a C-N carbon star primary and an A-type secondary. We report on new photometry and spectroscopy which tracked the recent disappearance of the A-star secondary. The dimming of the A-star was gradual and irregular, with one or more brief brightenings, implying the presence of nonhomogeneities in the carbon star outflow. We also present evidence that the A-star is actively accreting s-process enriched material from the carbon star and suggest that it will therefore eventually evolve into a Barium giant. This is an important system as well because the A-type star can serve as a probe of the outer atmosphere of the carbon star.Comment: 9 pages, 9 figures, 4 tables, a number of amateur observatories made significant contributions to this research.     There and back again: historical perspective and future directions for Vaccinium breeding and research studies

    The genus Vaccinium L. (Ericaceae) contains a wide diversity of culturally and economically important berry crop species. Consumer demand and scientific research in blueberry (Vaccinium spp.) and cranberry (Vaccinium macrocarpon) have increased worldwide over the crops' relatively short domestication history (~100 years). Other species, including bilberry (Vaccinium myrtillus), lingonberry (Vaccinium vitis-idaea), and ohelo berry (Vaccinium reticulatum) are largely still harvested from the wild but with crop improvement efforts underway. Here, we present a review article on these Vaccinium berry crops on topics that span taxonomy to genetics and genomics to breeding. We highlight the accomplishments made thus far for each of these crops, along their journey from the wild, and propose research areas and questions that will require investments by the community over the coming decades to guide future crop improvement efforts. New tools and resources are needed to underpin the development of superior cultivars that are not only more resilient to various environmental stresses and higher yielding, but also produce fruit that continue to meet a variety of consumer preferences, including fruit quality and health related trait

    Type 2 Diabetes Variants Disrupt Function of SLC16A11 through Two Distinct Mechanisms

    Type 2 diabetes (T2D) affects Latinos at twice the rate seen in populations of European descent. We recently identified a risk haplotype spanning SLC16A11 that explains ∼20% of the increased T2D prevalence in Mexico. Here, through genetic fine-mapping, we define a set of tightly linked variants likely to contain the causal allele(s). We show that variants on the T2D-associated haplotype have two distinct effects: (1) decreasing SLC16A11 expression in liver and (2) disrupting a key interaction with basigin, thereby reducing cell-surface localization. Both independent mechanisms reduce SLC16A11 function and suggest SLC16A11 is the causal gene at this locus. To gain insight into how SLC16A11 disruption impacts T2D risk, we demonstrate that SLC16A11 is a proton-coupled monocarboxylate transporter and that genetic perturbation of SLC16A11 induces changes in fatty acid and lipid metabolism that are associated with increased T2D risk. Our findings suggest that increasing SLC16A11 function could be therapeutically beneficial for T2D.     Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Internalised Values and Fairness Perception: Ethics in Knowledge Management

    This chapter argues for ethical consideration in knowledge management (KM). It explores the effect that internalised values and fairness perception have on individuals’ participation in KM practices. Knowledge is power, and organisations seek to manage knowledge through KM practices. For knowledge to be processed, individual employees—the source of all knowledge—need to be willing to participate in KM practices. As knowledge is power and a key constituent part of knowledge is ethics, individuals’ internalised values and fairness perception affect knowledge-processing. Where an organisation claims ownership over knowledge, an individual may perceive being treated unfairly, which may obstruct knowledge-processing. Through adopting ethical KM practices, individual needs are respected, enabling knowledge-processing. Implications point towards an ethical agenda in KM theory and practice

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival
