    Numerical semigroups, polyhedra, and posets III: minimal presentations and face dimension

    This paper is the third in a series of manuscripts that examines the combinatorics of the Kunz polyhedron PmP_m, whose positive integer points are in bijection with numerical semigroups (cofinite subsemigroups of (Z0,+)(\mathbb Z_{\ge 0}, +) whose smallest positive element is mm. The faces of PmP_m are indexed by a family of finite posets (called Kunz posets) obtained from the divisibility posets of the numerical semigroups lying on a given face. In this paper, we characterize to what extent the minimal presentation of a numerical semigroup can be recovered from its Kunz poset. In doing so, we prove that all numerical semigroups lying on the interior of a given face of PmP_m have identical minimal presentation cardinality, and provide a combinatorial method of obtaining the dimension of a face from its corresponding Kunz poset

    INTRODUCTION: The COVID-19 pandemic led to the reorganization of healthcare services and teleconsultation emerged as a solution to facilitate patient appointments. The aim of this study was to characterize, from a physician perspective, the teleconsultations carried out in the Portuguese National Health Service (SNS) during the first phase of the pandemic and to identify factors that influence the experience of the teleconsultation. MATERIAL AND METHODS: A cross-sectional analytical study was conducted based on an online survey, between July-September 2020, aimed at SNS doctors. Data on demographics, practice, attitudes and perceptions associated with the teleconsultation was collected. The adjusted prevalence ratio (aPR) was calculated to identify demographic factors and determinants of teleconsultation associated with satisfaction, use of video calls and greater motivation to carry out teleconsultations in the future. RESULTS: 2225 valid responses were obtained. Teleconsultation was carried out by 93.8% of participants in this period, 99.0% used the telephone as a form of communication and only 8.0% used a video call. A high degree of satisfaction with the teleconsultation was significantly associated with perceiving the teleconsultation as providing care with equivalent quality to a face-to-face consultation (aPR = 1.472) and being motivated to do teleconsultation after the pandemic (aPR = 4.081). Reporting clinical (aPR = 0.763) or technical difficulties (aPR = 0.666) was negatively associated with satisfaction). 70.4% of doctors would like to continue doing follow-up teleconsultations and 53.3% consider that video call technologies should always or often be used during teleconsultations. CONCLUSION: Teleconsultation seems to have potential to to become a common practice in the future. However, it is important to address clinical, technical, organizational, and legal questions and, above all, to ensure that it is a safe and valuable practice for patients.publishersversionepub_ahead_of_prin

    The Extracellular Vesicles of the Helminth Pathogen, Fasciola hepatica: Biogenesis Pathways and Cargo Molecules Involved in Parasite Pathogenesis

    Extracellular vesicles (EVs) released by parasites have important roles in establishing and maintaining infection. Analysis of the soluble and vesicular secretions of adult Fasciola hepatica has established a definitive characterization of the total secretome of this zoonotic parasite. Fasciola secretes at least two subpopulations of EVs that differ according to size, cargo molecules and site of release from the parasite. The larger EVs are released from the specialized cells that line the parasite gastrodermus and contain the zymogen of the 37 kDa cathepsin L peptidase that performs a digestive function. The smaller exosome-like vesicle population originate from multivesicular bodies within the tegumental syncytium and carry many previously described immunomodulatory molecules that could be delivered into host cells. By integrating our proteomics data with recently available transcriptomic data sets we have detailed the pathways involved with EV biogenesis in F. hepatica and propose that the small exosome biogenesis occurs via ESCRT-dependent MVB formation in the tegumental syncytium before being shed from the apical plasma membrane. Furthermore, we found that the molecular "machinery" required for EV biogenesis is constitutively expressed across the intramammalian development stages of the parasite. By contrast, the cargo molecules packaged within the EVs are developmentally regulated, most likely to facilitate the parasites migration through host tissue and to counteract host immune attack

    <div><p>Helminth parasites secrete extracellular vesicles (EVs) that can be internalised by host immune cells resulting in modulation of host immunity. While the molecular cargo of EVs have been characterised in many parasites, little is known about the surface-exposed molecules that participate in ligand-receptor interactions with the host cell surface to initiate vesicle docking and subsequent internalisation. Using a membrane-impermeable biotin reagent to capture proteins displayed on the outer membrane surface of two EV sub-populations (termed 15k and 120k EVs) released by adult <i>F</i>. <i>hepatica</i>, we describe 380 surface proteins including an array of virulence factors, membrane transport proteins and molecules involved in EV biogenesis/trafficking. Proteomics and immunohistochemical analysis show that the 120k EVs have an endosomal origin and may be released from the parasite via the protonephridial (excretory) system whilst the larger 15k EVs are released from the gastrodermal epithelial cells that line the fluke gut. A parallel lectin microarray strategy was used to profile the topology of major surface oligosaccharides of intact fluorogenically-labelled EVs as they would be displayed to the host. Lectin profiles corresponding to glycoconjugates exposed on the surface of the 15 K and 120K EV sub-populations are practically identical but are distinct from those of the parasite surface tegument, although all are predominated by high mannose sugars. We found that while the <i>F</i>. <i>hepatica</i> EVs were resistant to <i>exo</i>- and <i>endo</i>-glycosidases, the glyco-amidase PNGase F drastically remodelled the surface oligosaccharides and blocked the uptake of EVs by host macrophages. In contrast, pre-treatment with antibodies obtained from infected hosts, or purified antibodies raised against the extracellular domains of specific EV surface proteins (DM9-containing protein, CD63 receptor and myoferlin), significantly enhanced their cellular internalisation. This work highlights the diversity of EV biogenesis and trafficking pathways used by <i>F</i>. <i>hepatica</i> and sheds light on the molecular interaction between parasite EVs and host cells.</p></div

    The impact of city-wide deployment of Wolbachia-carrying mosquitoes on arboviral disease incidence in Medellín and Bello, Colombia: study protocol for an interrupted time-series analysis and a test-negative design study.

    Background: Dengue, chikungunya and Zika are viral infections transmitted by Aedes aegypti mosquitoes, and present major public health challenges in tropical regions. Traditional vector control methods have been ineffective at halting disease transmission. The World Mosquito Program has developed a novel approach to arbovirus control using Ae. aegypti stably transfected with the Wolbachia bacterium, which have significantly reduced ability to transmit dengue, Zika and chikungunya in laboratory experiments. Field releases in eight countries have demonstrated Wolbachia establishment in local Ae. aegypti populations. Methods: We describe a pragmatic approach to measuring the epidemiological impact of city-wide Wolbachia deployments in Bello and Medellín, Colombia. First, an interrupted time-series analysis will compare the incidence of dengue, chikungunya and Zika case notifications before and after Wolbachia releases, across the two municipalities. Second, a prospective case-control study using a test-negative design will be conducted in one quadrant of Medellín. Three of the six contiguous release zones in the case-control area were allocated to receive the first Wolbachia deployments in the city and three to be treated last, approximating a parallel two-arm trial for the >12-month period during which Wolbachia exposure remains discordant. Allocation, although non-random, aimed to maximise balance between arms in historical dengue incidence and demographics. Arboviral disease cases and arbovirus-negative controls will be enrolled concurrently from febrile patients presenting to primary care, with case/control status classified retrospectively following laboratory diagnostic testing. Intervention effect is estimated from an aggregate odds ratio comparing Wolbachia-exposure odds among test-positive cases versus test-negative controls. Discussion: The study findings will add to an accumulating body of evidence from global field sites on the efficacy of the Wolbachia method in reducing arboviral disease incidence, and can inform decisions on wider public health implementation of this intervention in the Americas and beyond.     Redshift distributions of galaxies in the Dark Energy Survey Science Verification shear catalogue and implications for weak lensing

    We present photometric redshift estimates for galaxies used in the weak lensing analysis of the Dark Energy Survey Science Verification (DES SV) data. Four model- or machine learning-based photometric redshift methods—ANNZ2, BPZ calibrated against BCC-Ufig simulations, SKYNET, and TPZ—are analyzed. For training, calibration, and testing of these methods, we construct a catalogue of spectroscopically confirmed galaxies matched against DES SV data. The performance of the methods is evaluated against the matched spectroscopic catalogue, focusing on metrics relevant for weak lensing analyses, with additional validation against COSMOS photo-z’s. From the galaxies in the DES SV shear catalogue, which have mean redshift 0.72 0.01 over the range 0.3 < z < 1.3, we construct three tomographic bins with means of z ¼ f0.45; 0.67; 1.00g. These bins each have systematic uncertainties δz ≲ 0.05 in the mean of the fiducial SKYNET photo-z nðzÞ. We propagate the errors in the redshift distributions through to their impact on cosmological parameters estimated with cosmic shear, and find that they cause shifts in the value of σ8 of approximately 3%. This shift is within the one sigma statistical errors on σ8 for the DES SV shear catalogue. We further study the potential impact of systematic differences on the critical surface density, Σcrit, finding levels of bias safely less than the statistical power of DES SV data. We recommend a final Gaussian prior for the photo-z bias in the mean of nðzÞ of width 0.05 for each of the three tomographic bins, and show that this is a sufficient bias model for the corresponding cosmology analysis
