Effects of experimental design and its role in interpretation of results
A total of 256 weanling pigs (PIC TR4 Ă 1050, initially 13.8 lb and 21 d of age) were used in a 28-d growth trial to compare allotment methods of a completely randomized design (CRD) and a randomized complete block design (RCBD). Two treatments were used to compare these designs: a negative control with no antibiotic or growth promoter and a positive control with 35 g/ton of Denagard (Novartis Animal Health), 400 g/ton of chlortetracycline, and zinc from zinc oxide at 3,000 and 2,000 ppm in Phases 1 and 2, respectively. Experimental diets were fed in 2 phases: Phase 1 from d 0 to 14 and Phase 2 from d 14 to 28. Eight replications of each dietary treatment were used for each experimental design. The first statistical model examined dietary treatment,
experimental design, and the design Ă dietary treatment as fixed factors. With the exception of pens in the CRD having a trend for improved (P 0.11) for any responses variables, indicating that treatment means reacted similarly in each of the experimental designs. In both the CRD and the RCBD, pig weights were increased (P < 0.003) with supplementation of growth promoters on d 14 and 28. Variation of weight within pen remained the same in the CRD from d 0 to 28 at approximately 20% but increased from 3% on d 0 to 10% on d 28 for the RCBD. Dietary addition of growth promoters increased (P < 0.003) ADG and ADFI and improved F/G (P < 0.04) in both the CRD and RCBD from d 0 to 14, with lower P-values for the CRD than the RCBD. From d 14 to 28, the CRD detected an increase (P < 0.001) in ADG and ADFI with dietary addition of growth promoters, and the RCBD detected an increase (P < 0.001) only in ADFI. Over the entire 28-d trial, growth promoters increased (P < 0.001) ADG and ADFI and improved (P < 0.03) F/G in the CRD and increased (P < 0.02) ADG and ADFI in the RCBD. Lower standard errors for the difference were also estimated for ADG and F/G in the CRD than in the RCBD from d 0 to 28. The average corrected relative efficiency for each of the three periods was 2.08 for ADG, 5.05 for ADFI, and 0.80 for F/G. The gain and intake values suggest that the added variation explained by blocks in the RCBD was beneficial for achieving a more reduced estimate of Ï2error compared with analyzing that particular data set as a CRD. The variance ratios of the CRD to RCBD from d 0 to 28 depict the different responses well with ADG at 0.67, ADFI at 1.70, and F/G at 0.22. When these ratios were compared with an F-test, they were well below the upper critical limit of 4.60, suggesting that the CRD offered estimates for Ï2error similar to those of the RCBD. With the same estimate for Ï2error, the non-centrality parameter for each design would be similar, and therefore, the increase in degrees of freedom (DF) for the error term would lead to greater power to detect differences in the CRD. Additional studies are needed to verify these results and determine whether blocking is an efficient use of error DF
Secondary education reform in Lesotho and Zimbabwe and the needs of rural girls: Pronouncements, policy and practice
Analysis of the educational needs of rural girls in Lesotho and Zimbabwe suggests a number of shortcomings in the current form of secondary education, and ways in which it might be modified so as to serve this sizeable group of students better. Several of the shortcomings, notably in relation to curricular irrelevance and excessive focus on examinations, have long been recognised, including by politicians. Yet political pronouncements are seldom translated into policy, and even where policy is formulated, reforms are seldom implemented in schools. This paper makes use of interviews with educational decision-makers in the two southern African countries and a range of documentary sources to explore why, despite the considerable differences between the two contexts, much needed educational reforms have been implemented in neither
Big GABA II: Water-referenced edited MR spectroscopy at 25 research sites
Accurate and reliable quantification of brain metabolites measured in vivo using 1H magnetic resonance spectroscopy (MRS) is a topic of continued interest. Aside from differences in the basic approach to quantification, the quantification of metabolite data acquired at different sites and on different platforms poses an additional methodological challenge. In this study, spectrally edited Îł-aminobutyric acid (GABA) MRS data were analyzed and GABA levels were quantified relative to an internal tissue water reference. Data from 284 volunteers scanned across 25 research sites were collected using GABA+ (GABA + co-edited macromolecules (MM)) and MM-suppressed GABA editing. The unsuppressed water signal from the volume of interest was acquired for concentration referencing. Whole-brain T1-weighted structural images were acquired and segmented to determine gray matter, white matter and cerebrospinal fluid voxel tissue fractions. Water-referenced GABA measurements were fully corrected for tissue-dependent signal relaxation and water visibility effects. The cohort-wide coefficient of variation was 17% for the GABA + data and 29% for the MM-suppressed GABA data. The mean within-site coefficient of variation was 10% for the GABA + data and 19% for the MM-suppressed GABA data. Vendor differences contributed 53% to the total variance in the GABA + data, while the remaining variance was attributed to site- (11%) and participant-level (36%) effects. For the MM-suppressed data, 54% of the variance was attributed to site differences, while the remaining 46% was attributed to participant differences. Results from an exploratory analysis suggested that the vendor differences were related to the unsuppressed water signal acquisition. Discounting the observed vendor-specific effects, water-referenced GABA measurements exhibit similar levels of variance to creatine-referenced GABA measurements. It is concluded that quantification using internal tissue water referencing is a viable and reliable method for the quantification of in vivo GABA levels
Recruitment and selection strategies in optometric education towards addressing human resource disparities in sub-Saharan Africa
The dire need for eye care services and a dearth of human resources (HR) in sub-Saharan Africa motivated the setting up of new optometry programmes. However, to make a meaningful impact, geographical, gender, economic and educational disparities must additionally be addressed. A qualitative study utilizing purposive sampling to select academic leadership and students from optometry programmes in sub-Saharan Africa was conducted. Individual and focus group interviews produced data that were coded and analysed using a deductive thematic analysis approach. The themes that emerged as contributing to disparities in access through recruitment and selection were institutional barriers (student intake numbers, programme marketing, minimum entry requirements, absence of pre-medical programme) and socio-economic barriers (finance, poor secondary school education, lack of knowledge of optometry, geographic location of institutions, gender). To address equity, institutions should engage with communities, market via community radio stations, offer pre-medical and bridging programmes, partner with governments and private funders to offer loans and bursaries and affirm females and rural applicants in recruitment and selection. In conclusion, universities must be socially accountable in all facets of education including recruitment and selection
Cross-ancestry genome-wide association analysis of corneal thickness strengthens link between complex and Mendelian eye diseases
Central corneal thickness (CCT) is a highly heritable trait associated with complex eye diseases such as keratoconus and glaucoma. We perform a genome-wide association meta-analysis of CCT and identify 19 novel regions. In addition to adding support for known connective tissue-related pathways, pathway analyses uncover previously unreported gene sets. Remarkably, >20% of the CCT-loci are near or within Mendelian disorder genes. These included FBN1, ADAMTS2 and TGFB2 which associate with connective tissue disorders (Marfan, Ehlers-Danlos and Loeys-Dietz syndromes), and the LUM-DCN-KERA gene complex involved in myopia, corneal dystrophies and cornea plana. Using index CCT-increasing variants, we find a significant inverse correlation in effect sizes between CCT and keratoconus (r =-0.62, P = 5.30 Ă 10-5) but not between CCT and primary open-angle glaucoma (r =-0.17, P = 0.2). Our findings provide evidence for shared genetic influences between CCT and keratoconus, and implicate candidate genes acting in collagen and extracellular matrix regulation
Measurement of the View the tt production cross-section using eÎŒ events with b-tagged jets in pp collisions at âs = 13 TeV with the ATLAS detector
This paper describes a measurement of the inclusive top quark pair production cross-section (ÏttÂŻ) with a data sample of 3.2 fbâ1 of protonâproton collisions at a centre-of-mass energy of âs = 13 TeV, collected in 2015 by the ATLAS detector at the LHC. This measurement uses events with an opposite-charge electronâmuon pair in the final state. Jets containing b-quarks are tagged using an algorithm based on track impact parameters and reconstructed secondary vertices. The numbers of events with exactly one and exactly two b-tagged jets are counted and used to determine simultaneously ÏttÂŻ and the efficiency to reconstruct and b-tag a jet from a top quark decay, thereby minimising the associated systematic uncertainties. The cross-section is measured to be:
ÏttÂŻ = 818 ± 8 (stat) ± 27 (syst) ± 19 (lumi) ± 12 (beam) pb,
where the four uncertainties arise from data statistics, experimental and theoretical systematic effects, the integrated luminosity and the LHC beam energy, giving a total relative uncertainty of 4.4%. The result is consistent with theoretical QCD calculations at next-to-next-to-leading order. A fiducial measurement corresponding to the experimental acceptance of the leptons is also presented
The performance of the jet trigger for the ATLAS detector during 2011 data taking
The performance of the jet trigger for the ATLAS detector at the LHC during the 2011 data taking period is described. During 2011 the LHC provided protonâproton collisions with a centre-of-mass energy of 7 TeV and heavy ion collisions with a 2.76 TeV per nucleonânucleon collision energy. The ATLAS trigger is a three level system designed to reduce the rate of events from the 40 MHz nominal maximum bunch crossing rate to the approximate 400 Hz which can be written to offline storage. The ATLAS jet trigger is the primary means for the online selection of events containing jets. Events are accepted by the trigger if they contain one or more jets above some transverse energy threshold. During 2011 data taking the jet trigger was fully efficient for jets with transverse energy above 25 GeV for triggers seeded randomly at Level 1. For triggers which require a jet to be identified at each of the three trigger levels, full efficiency is reached for offline jets with transverse energy above 60 GeV. Jets reconstructed in the final trigger level and corresponding to offline jets with transverse energy greater than 60 GeV, are reconstructed with a resolution in transverse energy with respect to offline jets, of better than 4 % in the central region and better than 2.5 % in the forward direction
Search for dark matter produced in association with a hadronically decaying vector boson in pp collisions at sqrt (s) = 13 TeV with the ATLAS detector
A search is presented for dark matter produced in association with a hadronically decaying W or Z boson using 3.2 fbâ1 of pp collisions at View the MathML sources=13 TeV recorded by the ATLAS detector at the Large Hadron Collider. Events with a hadronic jet compatible with a W or Z boson and with large missing transverse momentum are analysed. The data are consistent with the Standard Model predictions and are interpreted in terms of both an effective field theory and a simplified model containing dark matter
