    Antimicrobial, Antioxidant and Protective Effect of Methanol Extract of Trichilia emetica (Meliaceae) Stem and Root Bark against Free Radical-induced Oxidative Haemolysis

    Background: The objectives of this study are to investigate the antimicrobial, antioxidant and antiradical properties of methanol extract of the stem and root bark of Trichilia emetica on a model of free radical-induced membrane damage. Methodology: p-Iodonitrotetrazolium chloride colorometric assay was used to determined bacterial and yeast susceptibility to plant samples. The antioxidant activity was evaluated using DPPH radical, hydroxyl radical, nitric oxide scavenging and reducing power assays. The cellular oxidative stress was evaluated through haematolysis and haematoprotective assays. Results: The stem bark extract revealed a great antibacterial activity against 62.5% of bacteria strains tested with minimal inhibitory concentrations ranging from 32 to 64 µg/ml. The IC50 value the stem and root bark extracts were 9.68 ± 0.75 and 10.05 ± 0.86 μg/ml, 9.98 ± 0.52 and 200.49 ± 3.40 μg/ml, 45.92 ± 2.45 and 4081 ± 212 μg/ml, respectively for DPPH radical, hydroxyl radical and reducing ability assays. Both extracts exhibited concentration dependent haemolysis of sheep red blood cells (RBCs). Stem and root bark extracts (25 µg/ml) diminish respectively by 95.9 and 85.68% hydrogen peroxide-mediated cytotoxicity. Conclusion: Our data reveals that antibacterial and antioxidant activities of the methanol extract of Trichilia emetica stem bark were better than that of root bark

    Chemical analysis of the stem bark of Erythrina sigmoidea (Leguminoseae) yielded two known isoflavones, 6,8-diprenylgenisteine (3) and warangalone (4) as well as a new triterpenoid saponin designated sigmoiside E (1). Its structure was established by chemical and spectroscopic means as 16-O-β-D-galactopyranosyl maniladiol (1). Sigmoiside E exhibited antibacterial activity against gram-negative bacteria.     Assessment of Potential Herb-Drug Interactions among Nigerian Adults with Type-2 Diabetes.

    It is becoming increasingly evident that patients with diabetes do not rely only on prescription drugs for their disease management. The use of herbal medicines is one of the self-management practices adopted by these patients, often without the knowledge of their healthcare practitioners. This study assessed the potential for pharmacokinetic herb-drug interactions (HDIs) amongst Nigerian adult diabetic patients. This was done through a literature analysis of the pharmacokinetic profile of their herbal medicines and prescription drugs, based on information obtained from 112 patients with type-2 diabetes attending two secondary health care facilities in Nigeria. Fifty percent of the informants used herbal medicines alongside their prescription drugs. Worryingly, 60% of the patients taking herbal medicines did not know their identity, thus increasing the risk of unidentified HDIs. By comparing the pharmacokinetic profile of eight identified herbs taken by the patients for the management of diabetes against those of the prescription drugs, several scenarios of potential HDIs were identified and their clinical relevance is discussed. The lack of clinical predictors points toward cultural factors as the influence for herb use, making it more difficult to identify these patients and in turn monitor potential HDIs. In identifying these possible interactions, we have highlighted the need for healthcare professionals to promote a proactive monitoring of patients' use of herbal medicines

    Antagonistic fluorescent Pseudomonads: rhizobacteria with suppressive and plant growth promoting properties against Phytophthora colocasiae, the causal agent of taro leaf blight

    Taro leaf blight caused by Phytophthora colocasiae affects plant health and is a major threat to taro culture in Cameroon. Chemical fertilizers used often harm the ecosystem. Plant growth-promoting rhizobacteria (PGPR) are better alternatives that increase plant growth promotion and suppress phytopathogens. In the present study, a total of 67 fluorescent Pseudomonas spp. was characterized by 17.91, 5.97, and 4.47% populations of P. fluorescens, P. chlororaphis, and P. putida, respectively, among the most represented. More than 36% of bacteria showed antagonistic potential through the production of both diffusible and volatile compounds. Some of them (03) exhibited antagonistic activity in dual culture against P. colocasiae with a diameter greater than 13 mm. These rhizobacteria produced a significant amount of siderophore, IAA, SA, HCN, protease, lipases, and cellulases. For the pot experiment, treatment by Pseudomonas significantly increased the enzymatic activity involved in the resistance of taro, such as peroxidase (PO), polyphenol oxidase (PPO), and phenylalanine ammonia-lyase (PAL). The two antagonists also increased plant growth parameters of taro such as chlorophyll, plant height, shoot length, total leaf surface, fresh root biomass, and fresh leaf biomass. These findings showed that fluorescent Pseudomonas have an intriguing and undeniable potential in the fight against P. colocasiae, which could lead to the development of a biopesticide in the future

    An overview of anti-diabetic plants used in Gabon: Pharmacology and Toxicology

The aim of this review was to collect and present the scientific evidence for the use of medicinal plants that are in currect by Gabonese traditional healers to manage diabetes or hyperglycaemia based here on the pharmacological and toxicological profiles of plants with anti-diabetic activity. There are presented in order to promote their therapeutic value, ensure a safer use by population and provide some bases for further study on high potential plants reviewed. Materials and methods: Ethnobotanical studies were sourced using databases such as Online Wiley library, Pubmed, Google Scholar, PROTA, books and unpublished data including Ph.D. and Master thesis, African and Asian journals. Keywords including ‘Diabetes’ ‘Gabon’ ‘Toxicity’ ‘Constituents’ ‘hyperglycaemia’ were used. Results: A total of 69 plants currently used in Gabon with potential anti-diabetic activity have been identified in the literature, all of which have been used in in vivo or in vitro studies. Most of the plants have been studied in human or animal models for their ability to reduce blood glucose, stimulate insulin secretion or inhibit carbohydrates enzymes. Active substances have been identified in 12 out of 69 plants outlined in this review, these include Allium cepa and Tabernanthe iboga. Only eight plants have their active substances tested for anti-diabetic activity and are suitables for further investigation. Toxicological data is scarce and is dose-related to the functional parameters of major organs such as kidney and liver. Conclusion: An in-depth understanding on the pharmacology and toxicology of Gabonese anti-diabetic plants is lacking yet there is a great scope for new treatments.     The use of plants in the traditional management of diabetes in Nigeria: Pharmacological and toxicological considerations

    Ethnopharmacological relevance: The prevalence of diabetes is on a steady increase worldwide and it is now identified as one of the main threats to human health in the 21st century. In Nigeria, the use of herbal medicine alone or alongside prescription drugs for its management is quite common. We hereby carry out a review of medicinal plants traditionally used for diabetes management in Nigeria. Based on the available evidence on the species׳ pharmacology and safety, we highlight ways in which their therapeutic potential can be properly harnessed for possible integration into the country׳s healthcare system. Materials and methods: Ethnobotanical information was obtained from a literature search of electronic databases such as Google Scholar, Pubmed and Scopus up to 2013 for publications on medicinal plants used in diabetes management, in which the place of use and/or sample collection was identified as Nigeria. ‘Diabetes’ and ‘Nigeria’ were used as keywords for the primary searches; and then ‘Plant name – accepted or synonyms’, ‘Constituents’, ‘Drug interaction’ and/or ‘Toxicity’ for the secondary searches. Results: The hypoglycemic effect of over a hundred out of the 115 plants reviewed in this paper is backed by preclinical experimental evidence, either in vivo or in vitro. One-third of the plants have been studied for their mechanism of action, while isolation of the bioactive constituent(s) has been accomplished for twenty three plants. Some plants showed specific organ toxicity, mostly nephrotoxic or hepatotoxic, with direct effects on the levels of some liver function enzymes. Twenty eight plants have been identified as in vitro modulators of P-glycoprotein and/or one or more of the cytochrome P450 enzymes, while eleven plants altered the levels of phase 2 metabolic enzymes, chiefly glutathione, with the potential to alter the pharmacokinetics of co-administered drugs. Conclusion: This review, therefore, provides a useful resource to enable a thorough assessment of the profile of plants used in diabetes management so as to ensure a more rational use. By anticipating potential toxicities or possible herb–drug interactions, significant risks which would otherwise represent a burden on the country׳s healthcare system can be avoided

    The Helicobacter pylori Genome Project : insights into H. pylori population structure from analysis of a worldwide collection of complete genomes

    Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics

    Abstract Background Helicobacter pylori is a Gram negative bacterium that colonizes the stomach of approximately two-thirds of the human population and it is involved in the pathogenesis of gastroduodenal diseases. This study sought to determine potential risk factors associated with seroprevalence of H. pylori among dyspepsia patients in Cameroon for a better management of the disease. Methods The study was carried out from August to December 2014 at Laquintinie Hospital and District Hospital of Bonassama in Douala metropolis. 205 patients (127 women and 78 men; mean age, 53.79 ± 11.11 years; range, 35–75 years) were enrolled. Each subject gave a written consent. The study was approved by the local Ethical Committee of Medical Sciences. A structured questionnaire was used to collect information on sociodemographic parameters and predisposing risk factors for Helicobacter pylori infection. For each patient, body mass index (BMI) and direct inquiry about dyspeptic symptoms were done. Blood samples were tested for H. pylori antibodies, and ABO/Rhesus blood group antigen typing was performed. Results The overall prevalence was 64.39%. All patients with upper abdominal pains and frequent burping were H. pylori seropositive. We found that infection takes place early in childhood and adolescence, and reaches its peak at adulthood at 35 to 44 years. Sixty-two percent of women over 68 of men were infected. 80.39% of patients with family history of gastric cancer were seropositive, while 19.60% were seronegative (p = 0.001). Prevalence of 79.09 and 47.4% was recorded respectively for subjects with low, middle and high income levels (p = 0.001). H. pylori infection rate was 60.48% in blood group O patients compared with 70.37% in other blood groups (p = 0.203). 73% of infected subjects over 59% of uninfected ones currently take NSAIDs (p = 0.0509). Overcrowded households have a higher rate of 65.32% seropositivity in contrast with a lower rate of 33.33% from norm household (p = 0.197). 69.29% of obese and overweight patients versus 58.24% of subjects with normal weight were seropositive (P = 0.215). Conclusion The results of this study demonstrate that low income, family history of gastric cancer, clinical symptoms of nausea/vomiting and flatulence/bloating were risk factors of H. pylori infection in this population

    Context: Bryophyllum pinnatum (Lam.) Kurz (Crassulaceae) is a plant known for its antiulcer properties. Objective: This study evaluates the anti-Helicobacter pylori activity of Bryophyllum pinnutum methanol extract with a mouse model and its antioxidant properties. Materials and methods: Dried leaves of Bryophyllum pinnutum were extracted with methanol and ethyl acetate. Broth microdilution method was used to evaluate the anti-Helicobacter activity of extract samples in vitro. Swiss mice were inoculated with a suspension of Helicobacter pylori and divided into control group and four others that received 125, 250, 500 mg/kg of methanol extract or ciprofloxacin (500 mg/kg), respectively, for 7 days. Helicobacter pylori colonization and bacterial load of mouse stomach was assessed on day 1 and 7 post-treatment. The antioxidant activity of Bryophyllum pinnutum was evaluated through DPPH radical, hydroxyl radical and reducing power assay. Results: Methanol extract showed a significant anti-Helicobacter activity with MIC and MBC values of 32 and 256 μg/mL, respectively. Bryophyllum pinnatum and ciprofloxacin reduced H. pylori colonization of gastric tissue from 100% to 17%. Bryophyllum pinnatum extract (85.91 ± 52.91 CFU) and standard (25.74 ± 16.15 CFU) also reduced significantly (p < 0.05) bacterial load of gastric mucosa as compared to untreated infected mice (11883 ± 1831 CFU). DPPH radical, hydroxyl radical and reducing power assays showed IC50 values of 25.31 ± 0.34, 55.94 ± 0.68 and 11.18 ± 0.74 μg/mL, respectively. Discussion and conclusion: The data suggest that the methanol extract of Bryophyllum pinnatum could inhibit Helicobacter pylori growth, and may also acts as an antioxidant to protect gastric mucosa against reactive oxygen species

    Evaluation of antidiarrhoeal activity of the fruit-rind of Picralima nitida (Apocynaceae)

    The methanol (M) extract of the fruit-rind of Picralima nitida (PN) (Apocynaceae) was tested for its anti-diarrhoeal activity. Like loperamide (3 mg/kg body weight), a single oral dose of PN-M (375, 750 mg/kg body weight) produced a significant decrease in the frequency of defecation and severity of diarrhoea. To understand the mechanism of its anti-diarrhoeal activity, its effect was further evaluated on intestinal transit; castor oil-induced intestinal fluid accumulation (enteropooling) and electrolyte concentration in the small intestinal fluid. PN-M produced a decrease in intestinal transit (18.81-21.86%) as compared to castor oil treated animals. Unlike atropine, PN-M significantly inhibited castor oil-induced enteropooling. However it did not alter the electrolyte concentration in intestinal fluid as compared to castor oil treated rats.
