    Preparation and emulsifying properties of trace elements fortified gum arabic

    Gum arabic was enriched with trace elements (Zn2+, Fe3+, Fe2+) by ion exchange against ZnCl2, FeCl3 and FeCl2. Trace elements content, molecular parameters and emulsifying properties of the gum arabic rich in trace elements (GARTE) were characterized by flame atomic absorption spectrometry (FAAS), gel permeation chromatography-multi angle laser light scattering (GPC-MALLS), interfacial rheometer, laser particle analyzer and zeta potentiometry. With trace elements, molecular weight and arabinogalactan protein (AGP) content of gum arabic have increased probably due to the high surface energy leading to the aggregation of protein. GARTE has good emulsion stability performance with increasing molecular weight and AGP content compared to the control gum arabic. GARTE can be applied as a natural functional ingredient for trace element fortification, where the ferric ions and zinc ions are chelated by the self-assembled polymer host

    Smad Ubiquitination Regulatory Factor 1 (Smurf1) Promotes Thyroid Cancer Cell Proliferation and Migration via Ubiquitin-Dependent Degradation of Kisspeptin-1

    Background/Aims: Thyroid cancer is the most common malignancy in human endocrine system. Smad ubiquitination regulatory factor 1 (Smurf1) is an E3 ubiquitin-protein ligase in ubiquitin-proteasome pathway (UPP) system. This study aimed to investigate the effects of Smurf1 on thyroid cancer proliferation and metastasis, as well as underlying potential mechanism. Methods: The expression levels of Smurf1 in thyroid tumor tissues and thyroid cancer cells were detected by western blotting and qRT-PCR. Then, the effects of up-regulation or down-regulation of Smurf1 on thyroid cancer cell viability, migration, invasion, proliferation and apoptosis were measured using trypan blue exclusion assay, two-chamber migration (invasion) assay, cell colony formation assay and Guava Nexin assay, respectively. The ubiquitination of kisspeptin-1 (KISS-1) was assessed by protein ubiquitination assay. Finally, the effects of KISS-1 overexpression on activity of nuclear factor-kappa B (NF-κB) signaling pathway, as well as thyroid cancer cell viability, migration, invasion, proliferation and apoptosis were also detected, respectively. Results: Smurf1 was highly expressed in thyroid tumor tissues and thyroid cancer cells. Up-regulation of Smurf1 promoted the viability, migration, invasion and proliferation of thyroid cancer cells. Knockdown of Smurf1 had opposite effects. Moreover, smurf1 promoted the ubiquitination of KISS-1. Overexpression of KISS-1 inactivated NF-κB pathway, suppressed thyroid cancer cell viability, migration, invasion and proliferation, and induced cell apoptosis. Conclusion: Up-regulation of Smurf1 exerted important roles in thyroid cancer formation and development by promoting thyroid cancer proliferation and metastasis. The ubiquitin-dependent degradation of KISS-1 induced by Smurf1 and the activation of NF-κB signaling pathway might be involved in this process. Smurf1 could be an effective therapy target and biomarker for thyroid cancer treatment

    Large expert-curated database for benchmarking document similarity detection in biomedical literature search

    Document recommendation systems for locating relevant literature have mostly relied on methods developed a decade ago. This is largely due to the lack of a large offline gold-standard benchmark of relevant documents that cover a variety of research fields such that newly developed literature search techniques can be compared, improved and translated into practice. To overcome this bottleneck, we have established the RElevant LIterature SearcH consortium consisting of more than 1500 scientists from 84 countries, who have collectively annotated the relevance of over 180 000 PubMed-listed articles with regard to their respective seed (input) article/s. The majority of annotations were contributed by highly experienced, original authors of the seed articles. The collected data cover 76% of all unique PubMed Medical Subject Headings descriptors. No systematic biases were observed across different experience levels, research fields or time spent on annotations. More importantly, annotations of the same document pairs contributed by different scientists were highly concordant. We further show that the three representative baseline methods used to generate recommended articles for evaluation (Okapi Best Matching 25, Term Frequency-Inverse Document Frequency and PubMed Related Articles) had similar overall performances. Additionally, we found that these methods each tend to produce distinct collections of recommended articles, suggesting that a hybrid method may be required to completely capture all relevant articles. The established database server located at https://relishdb.ict.griffith.edu.au is freely available for the downloading of annotation data and the blind testing of new methods. We expect that this benchmark will be useful for stimulating the development of new powerful techniques for title and title/abstract-based search engines for relevant articles in biomedical research.Peer reviewe

    Coherent Perfect Absorption Laser Points in One-Dimensional Anti-Parity–Time-Symmetric Photonic Crystals

    We investigate the coherent perfect absorption laser points (CPA-LPs) in anti-parity–time-symmetric photonic crystals. CPA-LPs, which correspond to the poles of reflection and transmission, can be found in the parameter space composed of gain–loss factor and angular frequency. Discrete exceptional points (EPs) split as the gain–loss factor increases. The CPA-LPs sandwiched between the EPs are proved to be defective modes. The localization of light field and the bulk effect of gain/loss in materials induce a sharp change in phase of the reflection coefficient near the CPA-LPs. Consequently, a large spatial Goos–Hänchen shift, which is proportional to the slope of phase, can be achieved around the CPA-LPs. The study may find great applications in highly sensitive sensors

    Numerical Investigation on Dynamic Response of RC T-Beams Strengthened with CFRP under Impact Loading

    To precisely evaluate the retrofitting effectiveness of Carbon Fiber Reinforced Plastic (CFRP) sheets on the impact response of reinforced concrete (RC) T-beams, a non-linear finite element model was developed to simulate the structural response of T-beams with CFRP under impact loads. The numerical model was firstly verified by comparing the numerical simulation results with the experimental data, i.e., impact force, reaction force, and mid-span displacement. The strengthening effect of CFRP was analyzed from the section damage evaluation. Then the impact force, mid-span displacement, and failure mode of CFRP-strengthened RC T-beams were studied in comparison with those of un-strengthened T-beams. In addition, the influence of the impact resistance of T-beams strengthened with FRP was investigated in terms of CFRP strengthening mode, CFRP strengthening sizes, CFRP layers and FRP material types. The numerical simulation results indicate that the overall stiffness of the T-beams was improved significantly due to external CFRP strips. Compared with the un-strengthened beam, the maximum mid-span displacement of the CFRP-strengthened beam was reduced by 7.9%. Additionally, the sectional damage factors of the whole span of the CFRP-strengthened beam were reduced to less than 0.3, indicating that the impact resistance of the T-beams was effectively enhanced

    The Spillover Influence of Household Waste Sorting on Green Consumption Behavior by Mediation of Environmental Concern: Evidence from Rural China

    The spillover effect of environmental behavior has been of wide concern in recent years. The purpose of this paper is to explore the influence of household waste sorting on green consumption (behavioral spillover) and the possible psychological mechanisms involved in such spillover of environmental concern. Though it is important, insufficient attention has been paid to exploring the relationship, and the process of its formation, between waste sorting and green consumption. To narrow this gap, survey data collected in 2018 from 688 rural households from Shaanxi Province in western China were used. The propensity score matching method was employed to measure the effect of waste sorting on rural households’ green consumption. The mediating model was employed to investigate the path of influence in the relationship between waste sorting and green consumption. The results showed that waste sorting behavior positively spilled over into green consumption, with a net effect of 0.205. Environmental concern has a mediating effect on the relationship between waste sorting and green consumption behavior, with a mediating effect of 0.3177. In summary, household waste sorting behavior has a spillover effect on green consumption behavior as a result of the mediation effect of environmental concern. The results of this article fill in our knowledge on the spillover effects of waste sorting behavior in developing countries. Policy makers and regulators should vigorously advocate and implement waste sorting behavior, increase farmers’ concern for the environment, and promote their participation in green consumption behavior, so as to maximize the spillover effect

    Generation and characterization of a monoclonal antibody against human BCL6 for immunohistochemical diagnosis.

    BackgroundHuman B-cell lymphoma 6 (BCL6) gene, usually coding protein of 706 amino acids, is closely associated with large B cell lymphoma. Researches showed that protein mutation or change of expression levels usually happened in the mounting non-hodgkin lymphoma (NHL). Thus BCL6 is considered to be involved in germinal center (GC)-derived lymphoma.ResultsThe BCL61-350 gene codons were optimized for prokaryotic system. After expression of BCL61-350 in E. coli, the BCL61-350 protein was purified with Ni column. Then the BCL61-350 protein, mixing with QuickAntibody-Mouse5W adjuvant, was injected into Balb/c mice. After immunization and cell fusion, a stable cell line named 1E6A4, which can secrete anti-BCL6 antibody, was obtained. The isotype of 1E6A4 mAb was determined as IgG2a, and the affinity constant reached 5.12×1010 L/mol. Furthermore, the specificity of the mAb was determined with ELISA, western blot and immunohistochemistry. Results indicated that the 1E6A4 mAb was able to detect BCL6 specifically and sensitively.ConclusionsBCL61-350 antigen has been successfully generated with an effective and feasible method, and a highly specific antibody named 1E6A4 against BCL6 has been screened and characterized in this study, which was valuable in clinical diagnosis

    Effect of Potato Pulp Pectic Polysaccharide on the Stability of Acidified Milk Drinks

    In order to broaden the application of potato pulp pectic polysaccharide (PPP) in stabilizing acidified milk drinks (AMDs) and investigate the stabilizing effect and physical properties of AMDs prepared with PPP, a comparative study was made among PPP, commercial high methoxyl pectin (HMP) and low methoxyl pectin (LMP). The zeta potential, rheology, particle size and serum separation of AMDs were evaluated after preparing with PPP, HMP and LMP, respectively. Results indicated that PPP led to lower serum separation than LMP (14.65% for AMDs prepared with 0.5% PPP compared to 25.05% for AMDs prepared with 0.5% LMP), but still higher than HMP (9.09% for AMDs prepared with 0.5% HMP). However, narrower particle size distribution and lower viscosity of AMDs was achieved by PPP than by LMP and HMP. PPP can electrostatically adsorb on the surface of casein and its abundant neutral sugar side chains would provide steric hindrance to prevent casein flocculation in AMDs. Our results might provide some new ideas for the application of PPP in improving the stability of AMDs
