Large scale multifactorial likelihood quantitative analysis of BRCA1 and BRCA2 variants: An ENIGMA resource to support clinical variant classification
The multifactorial likelihood analysis method has demonstrated utility for quantitative assessment of variant pathogenicity for multiple cancer syndrome genes. Independent data types currently incorporated in the model for assessing BRCA1 and BRCA2 variants include clinically calibrated prior probability of pathogenicity based on variant location and bioinformatic prediction of variant effect, co-segregation, family cancer history profile, co-occurrence with a pathogenic variant in the same gene, breast tumor pathology, and case-control information. Research and clinical data for multifactorial likelihood analysis were collated for 1,395 BRCA1/2 predominantly intronic and missense variants, enabling classification based on posterior probability of pathogenicity for 734 variants: 447 variants were classified as (likely) benign, and 94 as (likely) pathogenic; and 248 classifications were new or considerably altered relative to ClinVar submissions. Classifications were compared with information not yet included in the likelihood model, and evidence strengths aligned to those recommended for ACMG/AMP classification codes. Altered mRNA splicing or function relative to known nonpathogenic variant controls were moderately to strongly predictive of variant pathogenicity. Variant absence in population datasets provided supporting evidence for variant pathogenicity. These findings have direct relevance for BRCA1 and BRCA2 variant evaluation, and justify the need for gene-specific calibration of evidence types used for variant classification

Introduction the prevalence of malnutrition in children and its impact on clinical outcomes is underrecognized by clinicians in Italy as well as worldwide. A novel definition of pediatric malnutrition has been recently proposed by a working group of the Academy of Nutrition and Dietetics and American Society for Parenteral and Enteral Nutrition (A.S.P.E.N.), based on the correlation between illness and the use of zscores of anthropometric measurements. Aim to investigate the prevalence of malnutrition and related nutritional support among hospitalized children in Italy, in a nationwide survey performed in a single day (16/4/2015). Methods an open access website (http://nday.biomedia.net) was used to collected data from 73 hospitals and 101 wards in 14 Italian regions (1994 patients). Anonymous information was collected on hospitals' characteristics, patient's anthropometry, admission diagnosis, presence of chronic diseases and use of nutritional support: oral nutritional supplements (ONS), enteral nutrition (EN) or parenteral nutrition (PN). Z-scores of anthropometric measurements, calculated with Epi Info 7.1.5, defined nutritional status: wasting was identified by BMI or Weight-for-Length z-score (<â1 mild, <â2 moderate, <â3 severe), stunting by Height-for-Age Z-score <â2. WHO 2006 and CDC 2000 growth charts were used respectively for children younger and older than 2 years old. Results 1790 complete records were obtained for hospitalized patients aged 0â20 years, with median age 6.16 (0.1â20 years and 53.3% males). 52.9% were aged 0â6 years and 58.8% of children suffered from chronic diseases. Wasting was detected in 28.7% of the total sample with higher occurrence observed in age ranges 0â6 and 14â20 years, while 17.3% of patients showed stunting; surprisingly almost 27% of them were aged 0â2. A ranking of the admission diagnosis with the highest rate of malnutrition was complied. The prevalence of wasting was significantly (p < 0.005) higher amongst children with chronic diseases (34.1% vs. 27.1%); stunting prevalence tripled in patients with chronic disease (24.5% vs. 8.3%). Only 23.5% of malnourished children (17%, 25.6% and 36.7%, respectively mild, moderate and severe malnutrition) received nutritional support: 11.7% received oral nutrition supplements (ONS, modular or complete), 11.5% enteral nutrition (EN, 6.4% via nasogastric tube, 5.1% via gastrostomy) and 6.8 % received parenteral nutrition (PN); in some patients a combination of two. Nutritional support is more commonly used among stunting patients, 39.5% of children under treatment. Conclusion Malnutrition of any grade was observed in nearly 1/3 and stunting in 17% of the reported hospitalized children, and it is likely to be underrecognized as the nutritional support reached only a small part of the malnourished children

THE VIRGO INTERFEROMETER FOR GRAVITATIONAL WAVE DETECTION
The Virgo interferometer for gravitational wave detection is described. During the commissioning phase that followed the first scientific data taking run an unprecedented sensitivity was obtained in the range 10-60 Hz. Since then an upgrade program has begun, with the aim of increasing the sensitivity, mainly through the introduction of fused silica wires to suspend mirrors and by increasing the Finesse of the Fabry-Perot cavities. Plans until the shutdown for the construction of the Advanced Virgo detector are given as well as the status of the upgrade
A THERMAL COMPENSATION SYSTEM FOR THE GRAVITATIONAL WAVE DETECTOR VIRGO
Thermal lensing due to the absorption of the laser beam in core optics of gravitational wave interferometers can represent a strong limitation to their operation and sensitivity. This effect has already been observed in the present detectors and will become more relevant in the future upgraded interferometers, due to the much higher circulating power. A thermal compensation system, based on a CO2 laser projector, has been installed in Virgo, allowing to increase the interferometer input power from 7 to 17 W. The thermal compensation system can introduce displacement noise by means of several mechanisms. This noise depends on the CO2 laser intensity fluctuations and on the power needed to compensate thermal effects. To make the displacement noise compliant with Virgo specifications, a feedback system to reduce the CO2 laser intensity fluctuations has been implemented
Search for High-energy Neutrinos from Binary Neutron Star Merger GW170817 with ANTARES, IceCube, and the Pierre Auger Observatory
The Advanced LIGO and Advanced Virgo observatories recently discovered
gravitational waves from a binary neutron star inspiral. A short gamma-ray
burst (GRB) that followed the merger of this binary was also recorded by the
Fermi Gamma-ray Burst Monitor (Fermi-GBM), and the Anticoincidence Shield for
the Spectrometer for the International Gamma-Ray Astrophysics Laboratory
(INTEGRAL), indicating particle acceleration by the source. The precise
location of the event was determined by optical detections of emission
following the merger. We searched for high-energy neutrinos from the merger in
the GeV--EeV energy range using the ANTARES, IceCube, and Pierre Auger
Observatories. No neutrinos directionally coincident with the source were
detected within s around the merger time. Additionally, no MeV
neutrino burst signal was detected coincident with the merger. We further
carried out an extended search in the direction of the source for high-energy
neutrinos within the 14-day period following the merger, but found no evidence
of emission. We used these results to probe dissipation mechanisms in
relativistic outflows driven by the binary neutron star merger. The
non-detection is consistent with model predictions of short GRBs observed at a

First narrow-band search for continuous gravitational waves from known pulsars in advanced detector data
International audienceSpinning neutron stars asymmetric with respect to their rotation axis are potential sources of continuous gravitational waves for ground-based interferometric detectors. In the case of known pulsars a fully coherent search, based on matched filtering, which uses the position and rotational parameters obtained from electromagnetic observations, can be carried out. Matched filtering maximizes the signal-to-noise (SNR) ratio, but a large sensitivity loss is expected in case of even a very small mismatch between the assumed and the true signal parameters. For this reason, narrow-band analysis methods have been developed, allowing a fully coherent search for gravitational waves from known pulsars over a fraction of a hertz and several spin-down values. In this paper we describe a narrow-band search of 11Â pulsars using data from Advanced LIGOâs first observing run. Although we have found several initial outliers, further studies show no significant evidence for the presence of a gravitational wave signal. Finally, we have placed upper limits on the signal strain amplitude lower than the spin-down limit for 5 of the 11 targets over the bands searched; in the case of J1813-1749 the spin-down limit has been beaten for the first time. For an additional 3 targets, the median upper limit across the search bands is below the spin-down limit. This is the most sensitive narrow-band search for continuous gravitational waves carried out so far
