The Work of Columbia University\u27s Latin Lab, with Emphasis on Medellin\u27s Borderlands , Clara Irazabal
Abstract:
In recent years the renaissance of the city of Medellin has achieved great recognition. Just weeks ago, Medellin hosted the United Nations\u27 World Urban Forum 7, which celebrated its urban planning transformation. According to most architectural and urban planning debates, this Colombian city has successfully been investing in infrastructural and social projects to improve the conditions in blighted neighborhoods as well as in the city as a whole. At the same time, some of the issues derived from the design of the new urban environment and the management of planning processes remain both unattended and understudied.
This presentation will introduce the experience that Columbia University’s Latin Lab had in Medellin. In Spring 2013, graduate planning students examined Medellin\u27s urban-rural borders and critically assessed ongoing neighborhood upgrading projects, by assuming a local community (Comuna 8) as their “client” and co-constructing planning alternative solutions with it. The lecture will present how the Lab\u27s experience critically engaged collaborative urban planning, design, risk management, and food security issues, bringing together research and proposal-making into a comprehensive project that aims to expand sustainability and justice.
Contesting Neoliberal Urbanization: The Mexican Case , Heidi Sohn
Abstract:
The advance and encroachment of neoliberal ideologies in all realms of our contemporary world have had significant impact upon the built environment. Although almost no aspect of the urbanization process has been exempt of the effects of urban policies implemented under the guise of liberalization and deregulation operating in the interest of private capital, it has been the social housing sector, which has arguably been the one where some the most important transformations have been registered, and which without a doubt will have long-termed effects upon cities. The neoliberal urban model follows quite similar formulas throughout the globe: the privatization of public interest being one common denominator that deeply affects the understanding and implementation of schemes aimed at providing the lower-income population and urban poor with viable alternatives for housing. Particularly interesting in this case are the elevated failure rates that are becoming patent in most of these schemes, and the problems that this phenomenon represents for the city.
Within this framework, the present paper will briefly introduce the background to the contemporary Mexican case, presenting an explanation of the forces at work in the dissolution of public agendas within the social housing sector and its absorption into the private sector of construction developers. Furthermore, it will investigate the culprits that render this process as corrupt and failed, highlighting the possible consequences and challenges that lay ahead. Two specific cases will be introduced and discussed as examples of this process: the housing development known as Las Américas, in Ecatepec de Morelos, State of México, and that of Riberas del Bravo, Ciudad Juárez, Chihuahua.
The main aim of this paper, however, is to reveal some of the possibilities of intervention for spatial planners and designers, and their role in the processes of re-conception, re-composition and recovery of housing developments erected under the neoliberal precepts that have become untenable today
Introduction: Commoning as Differentiated Publicness
Contemporary commoning practices do not constitute a mere alternative, but instead comprise a qualitative threshold: a moment of critical differentiation. As such, they call out for the development of a set of renewed methodological, analytical and synthetic tools and devices that are better equipped to understand the in-between as a ‘thirding’: as a form of differentiated publicness. The editorial introduction offers a platform of negotiation, which far from disregarding the already established approaches to the thematic in question, aims at expanding their scope, complementing them with non-dialectical readings. By presenting non-hierarchical understandings of urban practices, as well as fostering the intersection of different trajectories and discourses, the introduction to this issue strives to provide a fertile ground for the encounter of the multidimensional and relational potentials of contemporary commoning practices.
Mediating the Spatiality of Conflicts:
Conflict, when dislodged from its conventional understanding as a process and system of war and destruction exclusively, may be apprehended as an experimental method for analysis and synthesis, as a potent resource for pedagogy, for disruptive design and for the production of theory. In this sense, conflict produces more than the eradication of (the possibility of) life and its supporting structures: conflict produces transitional spaces at different scales, of differentiated material ecologies and site-specific meanings in relation to their global position. Conflicts are both locations and explanations of often ‘seductive’ images of destruction offered by popular (and other) media: ruined architectures, dead (or barely alive) bodies, forced migratory movements, impermanent infrastructures and settlements, as well as the tracing and construction of borders, real-estate driven post-war reconstruction processes, etc.
The emphasis on the mediatic aspect in the concept of conflict, may be seen as a way of triggering trans- and interdisciplinary discussions, conversations and encounters that serve as a negotiation between conditions of violence and new — or alternative — possibilities for everyday life. Artistic mediations could be as effective as violence in resolving conflicts, but operate through other means and through other channels, thus truly producing new power relations and alternative ways of political struggle. This exposing of conflict and violence through the artistic work is an activist act, but more importantly an artistic and technological mediation. The agency of the artistic work in terms of conflict, then, is situated in the capacity of visualising the conflict, creating awareness of its consequences, its side-effects, its collateral damage. And the creating of awareness and the becoming of fertile ground for protest and the creation of alternative realities.
At a three-day conference held at the TU Delft on November 6-8, 2019 researchers, scholars, activists, practitioners and artists presented individual papers that addressed the relationships between spatiality, mediation and conflict from a variety of perspectives. In addition to academic paper contributions, the conference welcomed other proposals in different formats and media: audio-visual material (film, video, photography), digital or physical archives, experimental design proposals, installations, performances, etc.
The thematic core of the conference explored new — or innovative — theoretical and methodological approaches and insights on: (1) Spaces of conflict as transitional spaces of material interactions between violence and everyday life; and (2) Spaces of memory as transformative space of violence)

DEEPEN: A negation detection system for clinical text incorporating dependency relation into NegEx
In Electronic Health Records (EHRs), much of valuable information regarding patients’ conditions is embedded in free text format. Natural language processing (NLP) techniques have been developed to extract clinical information from free text. One challenge faced in clinical NLP is that the meaning of clinical entities is heavily affected by modifiers such as negation. A negation detection algorithm, NegEx, applies a simplistic approach that has been shown to be powerful in clinical NLP. However, due to the failure to consider the contextual relationship between words within a sentence, NegEx fails to correctly capture the negation status of concepts in complex sentences. Incorrect negation assignment could cause inaccurate diagnosis of patients’ condition or contaminated study cohorts. We developed a negation algorithm called DEEPEN to decrease NegEx’s false positives by taking into account the dependency relationship between negation words and concepts within a sentence using Stanford dependency parser. The system was developed and tested using EHR data from Indiana University (IU) and it was further evaluated on Mayo Clinic dataset to assess its generalizability. The evaluation results demonstrate DEEPEN, which incorporates dependency parsing into NegEx, can reduce the number of incorrect negation assignment for patients with positive findings, and therefore improve the identification of patients with the target clinical findings in EHRs
Two independent proteomic approaches provide a comprehensive analysis of the synovial fluid proteome response to Autologous Chondrocyte Implantation
Background: Autologous chondrocyte implantation (ACI) has a failure rate of approximately 20%, but it is yet to be fully understood why. Biomarkers are needed that can pre-operatively predict in which patients it is likely to fail, so that alternative or individualised therapies can be offered. We previously used label-free quantitation (LF) with a dynamic range compression proteomic approach to assess the synovial fluid (SF) of ACI responders and non-responders. However, we were able to identify only a few differentially abundant proteins at baseline. In the present study, we built upon these previous findings by assessing higher-abundance proteins within this SF, providing a more global proteomic analysis on the basis of which more of the biology underlying ACI success or failure can be understood. Methods: Isobaric tagging for relative and absolute quantitation (iTRAQ) proteomic analysis was used to assess SF from ACI responders (mean Lysholm improvement of 33; n = 14) and non-responders (mean Lysholm decrease of 14; n = 13) at the two stages of surgery (cartilage harvest and chondrocyte implantation). Differentially abundant proteins in iTRAQ and combined iTRAQ and LF datasets were investigated using pathway and network analyses. Results: iTRAQ proteomic analysis confirmed our previous finding that there is a marked proteomic shift in response to cartilage harvest (70 and 54 proteins demonstrating ≥ 2.0-fold change and p < 0.05 between stages I and II in responders and non-responders, respectively). Further, it highlighted 28 proteins that were differentially abundant between responders and non-responders to ACI, which were not found in the LF study, 16 of which were altered at baseline. The differential expression of two proteins (complement C1s subcomponent and matrix metalloproteinase 3) was confirmed biochemically. Combination of the iTRAQ and LF proteomic datasets generated in-depth SF proteome information that was used to generate interactome networks representing ACI success or failure. Functional pathways that are dysregulated in ACI non-responders were identified, including acute-phase response signalling. Conclusions: Several candidate biomarkers for baseline prediction of ACI outcome were identified. A holistic overview of the SF proteome in responders and non-responders to ACI has been profiled, providing a better understanding of the biological pathways underlying clinical outcome, particularly the differential response to cartilage harvest in non-responders
Materials for Pharmaceutical Dosage Forms: Molecular Pharmaceutics and Controlled Release Drug Delivery Aspects
Controlled release delivery is available for many routes of administration and offers many advantages (as microparticles and nanoparticles) over immediate release delivery. These advantages include reduced dosing frequency, better therapeutic control, fewer side effects, and, consequently, these dosage forms are well accepted by patients. Advances in polymer material science, particle engineering design, manufacture, and nanotechnology have led the way to the introduction of several marketed controlled release products and several more are in pre-clinical and clinical development
Pharmacokinetic aspects of retinal drug delivery
Drug delivery to the posterior eye segment is an important challenge in ophthalmology, because many diseases affect the retina and choroid leading to impaired vision or blindness. Currently, intravitreal injections are the method of choice to administer drugs to the retina, but this approach is applicable only in selected cases (e.g. anti-VEGF antibodies and soluble receptors). There are two basic approaches that can be adopted to improve retinal drug delivery: prolonged and/or retina targeted delivery of intravitreal drugs and use of other routes of drug administration, such as periocular, suprachoroidal, sub-retinal, systemic, or topical. Properties of the administration route, drug and delivery system determine the efficacy and safety of these approaches. Pharmacokinetic and pharmacodynamic factors determine the required dosing rates and doses that are needed for drug action. In addition, tolerability factors limit the use of many materials in ocular drug delivery. This review article provides a critical discussion of retinal drug delivery, particularly from the pharmacokinetic point of view. This article does not include an extensive review of drug delivery technologies, because they have already been reviewed several times recently. Instead, we aim to provide a systematic and quantitative view on the pharmacokinetic factors in drug delivery to the posterior eye segment. Integrated genomic characterization of oesophageal carcinoma
Oesophageal cancers are prominent worldwide; however, there are few targeted therapies and survival rates for these cancers remain dismal. Here we performed a comprehensive molecular analysis of 164 carcinomas of the oesophagus derived from Western and Eastern populations. Beyond known histopathological and epidemiologic distinctions, molecular features differentiated oesophageal squamous cell carcinomas from oesophageal adenocarcinomas. Oesophageal squamous cell carcinomas resembled squamous carcinomas of other organs more than they did oesophageal adenocarcinomas. Our analyses identified three molecular subclasses of oesophageal squamous cell carcinomas, but none showed evidence for an aetiological role of human papillomavirus. Squamous cell carcinomas showed frequent genomic amplifications of CCND1 and SOX2 and/or TP63, whereas ERBB2, VEGFA and GATA4 and GATA6 were more commonly amplified in adenocarcinomas. Oesophageal adenocarcinomas strongly resembled the chromosomally unstable variant of gastric adenocarcinoma, suggesting that these cancers could be considered a single disease entity. However, some molecular features, including DNA hypermethylation, occurred disproportionally in oesophageal adenocarcinomas. These data provide a framework to facilitate more rational categorization of these tumours and a foundation for new therapies
