    Effective Knockdown of Gene Expression in Primary Microglia With siRNA and Magnetic Nanoparticles Without Cell Death or Inflammation.

    Microglia, the resident immune cells of the brain, have multiple functions in physiological and pathological conditions, including Alzheimer's disease (AD). The use of primary microglial cell cultures has proved to be a valuable tool to study microglial biology under various conditions. However, more advanced transfection methodologies for primary cultured microglia are still needed, as current methodologies provide low transfection efficiency and induce cell death and/or inflammatory activation of the microglia. Here, we describe an easy, and effective method based on the Glial-Mag method (OZ Biosciences) using magnetic nanoparticles and a magnet to successfully transfect primary microglia cells with different small interfering RNAs (siRNAs). This method does not require specialist facilities or specific training and does not induce cell toxicity or inflammatory activation. We demonstrate that this protocol successfully decreases the expression of two key genes associated with AD, the triggering receptor expressed in myeloid cells 2 (TREM2) and CD33, in primary microglia cell cultures

    La presente investigación busca desarrollar una propuesta de gestión de cambios y analizar su influencia en los costos y cronograma utilizando la metodología EVM en estructuras de concreto armado del proyecto multifamiliar las flores, SJL, Lima 2021, la metodología de la investigación es de tipo aplicada, método causal explicativo, el diseño es No Experimental – transversal y el enfoque es cuantitativo, el cual busca describir las ventajas aportadas por la propuesta de un procedimiento de gestión de cambios mediante la metodología del EVM en el desarrollo constructivo de las partidas de concreto armado. Del caso estudiado se obtuvieron los siguientes resultados; la implementación de un procedimiento de cambios y su validez utilizando la fórmula de alfa de Cronbach obteniendo un resultado de 0.9823 lo cual significa que es excelente confiable y aplicable al desarrollo del caso de estudio. También se analizó el desempeño de la obra mediante el EVM, para conocer el estado del desarrollo constructivo en base al cálculo de las variaciones, índices de desempeño de costo y cronograma y los pronósticos del costo al cierre de las partidas de concreto armado. Se concluye que el uso de un procedimiento para la gestión de cambios y utilizando EVM permite obtener datos para la toma de decisiones correctivas en miras al cumplimiento de las líneas base planificadas de un proyecto.This research seeks to develop a change management proposal and analyze its influence on costs and schedule using the EVM methodology in reinforced concrete structures of the Las Flores multifamily building, SJL, Lima 2021, the research methodology is of an applied type, explanatory causal method, the design is Non-Experimental - transversal and the approach is quantitative, which seeks to describe the advantages provided by the proposal of a change management procedure through the EVM methodology in the constructive development of the reinforced concrete items. The following results were obtained from the case studied; the implementation of a change procedure and its validity using Cronbach's alpha formula obtaining a result of 0.9823 which means that it is excellently reliable and applicable to the development of the case study. The performance of the work was also analyzed through the EVM, to know the status of the construction development based on the calculation of the variations, cost and schedule performance indices and the forecasts of the cost at the closing of the reinforced concrete items. It is concluded that the use of a change management procedure and using EVM allows obtaining data for corrective decision making in order to comply with the planned baselines of a project

    Amiloidosis de lengua asociado a mieloma múltiple: reporte de caso

    Resumen: La amiloidosis es una enfermedad de causa desconocida, que se caracteriza por el acúmulo de material proteico amorfo, que se deposita en diversos órganos y tejidos, produciendo atrofia y alteraciones funcionales de ellos. En la cavidad oral habitualmente se encuentra macroglosia. Se presenta el caso de un varón de 78 años con mieloma múltiple que comienza con crecimiento progresivo de la lengua que le impedía respirar, deglutir y le producía disfonía, al estudio histopatológico es diagnosticado con amiloidosis secundaria localizada en lengua asociado a mieloma múltiple. Los pacientes que tienen mieloma múltiple tienen una asociación con el desarrollo de una amiloidosis primaria; caso contrario sucede con la amiloidosis secundaria que son raras y más extrañas en el caso de una localizada. Es importante reconocer que el estudio histopatológico e inmunohistoquímico nos dan el diagnóstico definitivo de la amiloidosis y el tipo de esta.   DOI: https://doi.org/10.25176/RFMH.v17.n2.84

    Repositioning of the global epicentre of non-optimal cholesterol

    High blood cholesterol is typically considered a feature of wealthy western countries(1,2). However, dietary and behavioural determinants of blood cholesterol are changing rapidly throughout the world(3) and countries are using lipid-lowering medications at varying rates. These changes can have distinct effects on the levels of high-density lipoprotein (HDL) cholesterol and non-HDL cholesterol, which have different effects on human health(4,5). However, the trends of HDL and non-HDL cholesterol levels over time have not been previously reported in a global analysis. Here we pooled 1,127 population-based studies that measured blood lipids in 102.6 million individuals aged 18 years and older to estimate trends from 1980 to 2018 in mean total, non-HDL and HDL cholesterol levels for 200 countries. Globally, there was little change in total or non-HDL cholesterol from 1980 to 2018. This was a net effect of increases in low- and middle-income countries, especially in east and southeast Asia, and decreases in high-income western countries, especially those in northwestern Europe, and in central and eastern Europe. As a result, countries with the highest level of non-HDL cholesterol-which is a marker of cardiovascular riskchanged from those in western Europe such as Belgium, Finland, Greenland, Iceland, Norway, Sweden, Switzerland and Malta in 1980 to those in Asia and the Pacific, such as Tokelau, Malaysia, The Philippines and Thailand. In 2017, high non-HDL cholesterol was responsible for an estimated 3.9 million (95% credible interval 3.7 million-4.2 million) worldwide deaths, half of which occurred in east, southeast and south Asia. The global repositioning of lipid-related risk, with non-optimal cholesterol shifting from a distinct feature of high-income countries in northwestern Europe, north America and Australasia to one that affects countries in east and southeast Asia and Oceania should motivate the use of population-based policies and personal interventions to improve nutrition and enhance access to treatment throughout the world.Peer reviewe

    [Esp]Este estudio compara la evolución de los modelos de predicción de fracaso empresarial desarrollados a lo largo de los años. Se expone como alternativa el uso de análisis cualitativo comparativo (QCA) a través del modelo de conjuntos difusos fsQCA. El modelo se ha aplicado a una muestra de 10380 empresas del sector industrial en la región de Murcia escogidas por su código de actividad económica (CNAE-2009). Se han utilizado las variables de un centenar de estudios para realizar una selección basada en su peso y criterio de multicolinealidad. El estudio arroja una serie de configuraciones que distinguen las variables más relevantes que han podido desencadenar la quiebra sobre nuestra muestra. El modelo fsQCA ha sido desarrollado con el software fsQCA 3.0, y la síntesis del estudio ha sido desarrollada con R estudio. [Eng]This study compares the evolution of business failure prediction models developed over the years. The use of Qualitative Comparative Analysis (QCA) through the fsQCA fuzzy-set model is presented as an alternative. The model has been applied to a sample of 10380 companies in the industrial sector in the region of Murcia chosen by their economic activity code (CNAE-2009).The variables from a hundred studies have been used to make a selection based on their weight and multicollinearity criteria. The study reveals a series of settings that distinguish the most relevant variables that could have triggered the bankruptcy of our sample.
