    DEATH IN NEW HEAVEN: A HISTORICAL–GRAMMATICAL INQUIRY

    The book of Isaiah is one of the largest prophetic books in the Old Testament. One of the most confusing texts in this prophetic book is found in 65:20. It reads, “No more shall an infant from there live but a few days, nor an old man who has not fulfilled his days; for the child shall die one hundred years old, but the sinner being one hundred years old shall be accursed.” There are four main interpretations of this text which includes the following: (1). The prophecy pertains to Isaiah’s near future which is the restoration of Judah; (2) The prophecy refers to Isaiah’s distant future, which is the restoration of God’s church or God’s people; (3) The prophecy alludes  to the restoration of God’s people in the millennium; and (4) The prophecy speaks of the final restoration during or after the millennium. It appears that the proponents of the three main interpretations of Isaiah 65:20 fails to consider the historical and literary context of this important prophecy. What is evident from this study is that longevity of life promise in this prophecy is primarily for the people of Judah in the eighth century. Therefore, its fulfillment would take place within the time, place, and circumstances of the prophetic writers.     Students’ Career Choices: A Guide For Senior High School Preparation

    This study was conducted to determine what Senior High School (SHS) track and strands Adventist University of the Philippines (AUP) should offer starting school year 2016-2017 based on the career choices of 160 AUP Academy grades 7 to 9 students. DepEd mandates four tracks in SHS under the K+12 program, namely: Academic, Sports, Arts and Design, and Technical-Vocational (Tech-Voc)- Livelihood tracks. For each track, specialization strands are offered. Descriptive statistics were used. Variables on profile, awareness of track offering and interpersonal, intrinsic and extrinsic motivation were assessed. Results showed that 98% of the respondents would proceed to SHS. A total of 49% would pursue courses under the academic track while 13% would take sports, 8% will take the arts and design track, while 3% will take the track on technical-vocational and livelihood. A total of 95% were planning to still enroll in AUP for collegiate degrees. The researchers recommend that AUP prepare and enhance facilities for the academic track, particularly in the sciences, engineering, and business strands. It should also prepare programs and facilities in sports and arts, but also device strategies to make other strands attractive. Likewise, AUP should maintain its Adventist Christian standards, while parents continue to support their children’s career choices.Key words: Assessment, Career Choices, Junior High School, K-12 Program, Senior High School, Strands, Track

    Molecular imaging aims to visualize the cellular and molecular processes occurring in living tissues, and for the imaging of specific molecules in vivo, the development of reporter probes and dedicated imaging equipment is most important. Reporter genes can be used to monitor the delivery and magnitude of therapeutic gene transfer, and the time variation involved. Imaging technologies such as micro-PET, SPECT, MRI and CT, as well as optical imaging systems, are able to non-invasively detect, measure, and report the simultaneous expression of multiple meaningful genes. It is believed that recent advances in reporter probes, imaging technologies and gene transfer strategies will enhance the effectiveness of gene therapy trials

    Direct application of plasmid DNA containing type I interferon transgenes to vaginal mucosa inhibits HSV-2 mediated mortality

    The application of naked DNA containing type I interferon (IFN) transgenes is a promising potential therapeutic approach for controlling chronic viral infections. Herein, we detail the application of this approach that has been extensively used to restrain ocular HSV-1 infection, for antagonizing vaginal HSV-2 infection. We show that application of IFN-α1, -α5, and –β transgenes to vaginal mouse lumen 24 hours prior to HSV-2 infection reduces HSV-2 mediated mortality by 2.5 to 3-fold. However, other type I IFN transgenes (IFN- α4, -α5, -α6, and –α9) are non effectual against HSV-2. We further show that the efficacy of IFN-α1 transgene treatment is independent of CD4+ T lymphocytes. However, in mice depleted of CD8+ T lymphocytes, the ability of IFN-α1 transgene treatment to antagonize HSV-2 was lost

    Comparison of immune response generated against Japanese encephalitis virus envelope protein expressed by DNA vaccines under macrophage associated versus ubiquitous expression promoters

    <p>Abstract</p> <p>Background</p> <p>Japanese encephalitis virus (JEV) is the leading cause of viral encephalitis, with ~50,000 cases reported annually worldwide. Vaccination is the only measure for prevention. Recombinant vaccines are an efficient and safe alternative for formalin inactivated or live attenuated vaccines. Nowadays, incorporation of molecular adjuvants has been the main strategy for melioration of vaccines. Our attempt of immunomodulation is based on targeting antigen presenting cells (APC) "majorly macrophages" by using macrosialin promoter. We have compared the immune response of the constructed plasmids expressing JEV envelope (E) protein under the control of aforesaid promoter and cytomegalovirus (CMV) immediate early promoter in mouse model. Protection of immunized mice from lethal challenge with JEV was also studied.</p> <p>Results</p> <p>The E protein was successfully expressed in the macrophage cell line and was detected using immunofluorescence assay (IFA) and Western blotting. APC expressing promoter showed comparable expression to CMV promoter. Immunization of mice with either of the plasmids exhibited induction of variable JEV neutralizing antibody titres and provided protection from challenge with a lethal dose of JEV. Immune splenocytes showed proliferative response after stimulation with the JEV antigen (Ag), however, it was higher for CMV promoter. The magnitude of immunity provided by APC dominant promoter was non-significantly lower in comparison to CMV promoter. More importantly, immune response directed by APC promoter was skewed towards Th1 type in comparison to CMV promoter, this was evaluated by cytokine secretion profile of immune splenocytes stimulated with JEV Ag.</p> <p>Conclusions</p> <p>Thus, our APC-expressing DNA vaccination approach induces comparable immunity in comparison to ubiquitous promoter construct. The predominant Th1 type immune responses provide opportunities to further test its potency suitable for response in antiviral or anticancer vaccines.</p

    Classification of current anticancer immunotherapies

Many immunotherapeutic regimens are now approved by the US Food and Drug Administration and the European Medicines Agency for use in cancer patients, and many others are being investigated as standalone therapeutic interventions or combined with conventional treatments in clinical studies. Immunotherapies may be subdivided into "passive" and "active" based on their ability to engage the host immune system against cancer. Since the anticancer activity of most passive immunotherapeutics (including tumor-targeting monoclonal antibodies) also relies on the host immune system, this classification does not properly reflect the complexity of the drug-host-tumor interaction. Alternatively, anticancer immunotherapeutics can be classified according to their antigen specificity. While some immunotherapies specifically target one (or a few) defined tumor-associated antigen(s), others operate in a relatively non-specific manner and boost natural or therapy-elicited anticancer immune responses of unknown and often broad specificity.
