    Alkamides Activate Jasmonic Acid Biosynthesis and Signaling Pathways and Confer Resistance to Botrytis cinerea in Arabidopsis thaliana

    Alkamides are fatty acid amides of wide distribution in plants, structurally related to N-acyl-L-homoserine lactones (AHLs) from Gram-negative bacteria and to N- acylethanolamines (NAEs) from plants and mammals. Global analysis of gene expression changes in Arabidopsis thaliana in response to N-isobutyl decanamide, the most highly active alkamide identified to date, revealed an overrepresentation of defense-responsive transcriptional networks. In particular, genes encoding enzymes for jasmonic acid (JA) biosynthesis increased their expression, which occurred in parallel with JA, nitric oxide (NO) and H2O2 accumulation. The activity of the alkamide to confer resistance against the necrotizing fungus Botrytis cinerea was tested by inoculating Arabidopsis detached leaves with conidiospores and evaluating disease symptoms and fungal proliferation. N-isobutyl decanamide application significantly reduced necrosis caused by the pathogen and inhibited fungal proliferation. Arabidopsis mutants jar1 and coi1 altered in JA signaling and a MAP kinase mutant (mpk6), unlike salicylic acid- (SA) related mutant eds16/sid2-1, were unable to defend from fungal attack even when N-isobutyl decanamide was supplied, indicating that alkamides could modulate some necrotrophic-associated defense responses through JA-dependent and MPK6-regulated signaling pathways.     Nuevos aportes sobre alteraciones cognitivas y metabólico-cerebrales por la administración de ketamina en dosis subanestésicas en ratas Rattus norvegicus (estudio preliminar)

    La ketamina antagonista del receptor NMDA fue creada en la década del 60 como anestésico disociativo para niños y adultos; rápidamente ocupo un lugar fundamental en medicina veterinaria. Con el tiempo se le atribuyeron otro tipo de utilidades como analgésico, antidepresivo e incluso anticonvulsivante para algún tipo de estado epiléptico refractario. En nuestro laboratorio lo utilizamos como anestésico para la colocación de cánulas intracerebrales en dosis de 70 mg/kg.2 En el caso de su uso como analgésico para dolor de origen central y neuropático la dosis es muy por debajo de la utilizada para llegar al plano quirúrgico. Por lo que es fundamental saber si estas dosis conllevan consecuencias a nivel cognitivo y qué cambios se producen a nivel metabólico en las estructuras cerebrales implicadas en dichos procesos. En trabajos previos demostramos que la ketamina no produce alteración en el patrón de movimientos con dosis inferiores a 5 mg/ kg pero provoca detrimento de la memoria de trabajo asociado a alteraciones metabólicas de la corteza prefrontal e hipocampo.     Mortality of emergency abdominal surgery in high-, middle- and low-income countries

    Background: Surgical mortality data are collected routinely in high-income countries, yet virtually no low- or middle-income countries have outcome surveillance in place. The aim was prospectively to collect worldwide mortality data following emergency abdominal surgery, comparing findings across countries with a low, middle or high Human Development Index (HDI). Methods: This was a prospective, multicentre, cohort study. Self-selected hospitals performing emergency surgery submitted prespecified data for consecutive patients from at least one 2-week interval during July to December 2014. Postoperative mortality was analysed by hierarchical multivariable logistic regression. Results: Data were obtained for 10 745 patients from 357 centres in 58 countries; 6538 were from high-, 2889 from middle- and 1318 from low-HDI settings. The overall mortality rate was 1⋅6 per cent at 24 h (high 1⋅1 per cent, middle 1⋅9 per cent, low 3⋅4 per cent; P < 0⋅001), increasing to 5⋅4 per cent by 30 days (high 4⋅5 per cent, middle 6⋅0 per cent, low 8⋅6 per cent; P < 0⋅001). Of the 578 patients who died, 404 (69⋅9 per cent) did so between 24 h and 30 days following surgery (high 74⋅2 per cent, middle 68⋅8 per cent, low 60⋅5 per cent). After adjustment, 30-day mortality remained higher in middle-income (odds ratio (OR) 2⋅78, 95 per cent c.i. 1⋅84 to 4⋅20) and low-income (OR 2⋅97, 1⋅84 to 4⋅81) countries. Surgical safety checklist use was less frequent in low- and middle-income countries, but when used was associated with reduced mortality at 30 days. Conclusion: Mortality is three times higher in low- compared with high-HDI countries even when adjusted for prognostic factors. Patient safety factors may have an important role.     Taking the pulse of Earth's tropical forests using networks of highly distributed plots

    Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science.     Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Abstract Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries

    Effect of the joint administration of ketamine and fluoxetine in positive calbindine interneurons of the basilateral nucleus of amígdala in ratus novergicus: preliminary results

    There are different pharmacological treatments for depression, limited in their effectiveness and many have very long latency periods. Current research points to NMDA antagonists as possible therapeutic targets of this disorder. The objective of this work was to assess at a behavioral level the synergistic interactions between the antidepressant of the SSRIs group fluoxetine, and ketamine, a noncompetitive NMDA receptor antagonist and correlate the findings with structural changes in the basolateral nucleus of the tonsil with respect to positive calbindin Interneurons. Holtzman rats that were treated with fluoxetine were used. 10, 15 and 20 mg / kg and ketamine. 2.5, 5 and 10 mg / kg and subsequently were evaluated in the forced swim test each group consisted of n = 20. Fluoxetine in doses of 10 and 15 mg / kg and ketamine in doses of 2.5 and 5 mg / kg did not produce a significant decrease in immobility time, On the contrary, if significant decreases were observed with fluoxetine 20 mg / kg (p <0.01) and ketamine 10 mg / kg (p <0.05). Likewise, there was a significant decrease in immobility time when fluoxetine 10 mg / kg + ketamine 2.5 mg / kg (p <0.01) and fluoxetine 15 mg / kg + ketamine 5 mg / kg (p <0,0001). At 24 hours after the test, positive calbindin interneurons of the basolateral nucleus of the tonsil were analyzed by immunohistochemistry. Each group consisted of an n = 3. It was observed that the saline group presented significant differences with the group treated with 20 mg / kg of fluoxetine (p = 0.001), with the group treated with 5 mg / kg of fluoxetine (p = 0.0001) and with the group treated with 5 mg / kg of fluoxetine + 2.5 mg / kg of ketamine (p = 0.0001), the latter being the one with the least positive calbindin interneurons. Ketamine groups have not yet been analyzed. These results indicate that co-administration of fluoxetine and ketamine can induce a more potent antidepressant activity than when used alone. In addition, the decrease in the density of cositive calbindin interneurons in the basolateral nucleus of the amygdala could be directly related to the action of drugs on these neuronal populations, thereby increasing neuronal plasticity and being able to restore excitatory and inhibitory balance.Fil: Guevara, M.     <p>The islet amyloid polypeptide (IAPP) or amylin maintains a key role in metabolism. This 37-residues-peptide could form pancreatic amyloids, which are a characteristic feature of diabetes mellitus type 2. However, some species do not form amyloid fibril structures. By employing a biomimetic approach, we generated an extensive panel of optimized sequences of IAPP, which could drastically reduce aggregation propensity. A structural and cellular characterization analysis was performed on the C-terminal domain with the highest aggregation propensity. This allowed the observation of an aggregative phenomenon dependent of the lipid environment. Evaluation of the new F<sub>23</sub>R variant demonstrated inhibition of β-sheet structure and, therefore, amyloid formation on the native C-terminal, phenomenon that was associated with functional optimization in calcium and cholesterol management coupled with the optimization of insulin secretion by beta cells. When F<sub>23</sub>R variant was evaluated in microglia cells, a model of amyloidosis, cytotoxic conditions were not registered. In addition, it was found that C-terminal sequences of IAPP could modulate cholesterol metabolism in hepatocytes through regulation of SREBP-2, apoA-1, ABCA1, and LDLR, mechanism that may represent a new function of IAPP on the metabolism of cholesterol, increasing the LDL endocytosis in hepatocytes. Optimized sequences with only one residue modification in the C-terminal core aggregation could diminish β-sheet formation and represent a novel strategy adaptable to other pharmacological targets. Our data suggest a new IAPP function associated with rearrangements on metabolism of cholesterol in hepatocytes.</p
