    Design of Air Thermal Recovery Experiment Device and Analysis of Thermal Efficiency

    AbstractIt was significant for energy save and environment protection that heat recovering high temperature and high humidity air directly discharged into the atmosphere. Based on high temperature and high humidity mine retention tower and return air fluid, the air heat recovery simulation experiment device was designed, and air heat recovery experiment was researched by using spray heat exchanger. Heat exchange efficiency was tested by experiment with certain water spray coefficient and different nozzle number, and experiment result that was error analysis and data correction through compared with theoretical analysis results. Result showed that the air heat recovery simulation device can recycle heat of air fluid stably, and rise temperature of spray cold water to about 15°C. The heat exchange efficiency would be 90% with certain water spray coefficient, which is basically identical with theoretical analysis. Energy saving effect of this device was remarkable with high heat exchange efficiency

    Using Generalized Procrustes Analysis (GPA) for normalization of cDNA microarray data

    <p>Abstract</p> <p>Background</p> <p>Normalization is essential in dual-labelled microarray data analysis to remove non-biological variations and systematic biases. Many normalization methods have been used to remove such biases within slides (Global, Lowess) and across slides (Scale, Quantile and VSN). However, all these popular approaches have critical assumptions about data distribution, which is often not valid in practice.</p> <p>Results</p> <p>In this study, we propose a novel assumption-free normalization method based on the Generalized Procrustes Analysis (GPA) algorithm. Using experimental and simulated normal microarray data and boutique array data, we systemically evaluate the ability of the GPA method in normalization compared with six other popular normalization methods including Global, Lowess, Scale, Quantile, VSN, and one boutique array-specific housekeeping gene method. The assessment of these methods is based on three different empirical criteria: across-slide variability, the Kolmogorov-Smirnov (K-S) statistic and the mean square error (MSE). Compared with other methods, the GPA method performs effectively and consistently better in reducing across-slide variability and removing systematic bias.</p> <p>Conclusion</p> <p>The GPA method is an effective normalization approach for microarray data analysis. In particular, it is free from the statistical and biological assumptions inherent in other normalization methods that are often difficult to validate. Therefore, the GPA method has a major advantage in that it can be applied to diverse types of array sets, especially to the boutique array where the majority of genes may be differentially expressed.</p

    Mural granulosa cell gene expression associated with oocyte developmental competence

    <p>Abstract</p> <p>Background</p> <p>Ovarian follicle development is a complex process. Paracrine interactions between somatic and germ cells are critical for normal follicular development and oocyte maturation. Studies have suggested that the health and function of the granulosa and cumulus cells may be reflective of the health status of the enclosed oocyte. The objective of the present study is to assess, using an <it>in vivo </it>immature rat model, gene expression profile in granulosa cells, which may be linked to the developmental competence of the oocyte. We hypothesized that expression of specific genes in granulosa cells may be correlated with the developmental competence of the oocyte.</p> <p>Methods</p> <p>Immature rats were injected with eCG and 24 h thereafter with anti-eCG antibody to induce follicular atresia or with pre-immune serum to stimulate follicle development. A high percentage (30-50%, normal developmental competence, NDC) of oocytes from eCG/pre-immune serum group developed to term after embryo transfer compared to those from eCG/anti-eCG (0%, poor developmental competence, PDC). Gene expression profiles of mural granulosa cells from the above oocyte-collected follicles were assessed by Affymetrix rat whole genome array.</p> <p>Results</p> <p>The result showed that twelve genes were up-regulated, while one gene was down-regulated more than 1.5 folds in the NDC group compared with those in the PDC group. Gene ontology classification showed that the up-regulated genes included lysyl oxidase (<it>Lox</it>) and nerve growth factor receptor associated protein 1 (<it>Ngfrap1</it>), which are important in the regulation of protein-lysine 6-oxidase activity, and in apoptosis induction, respectively. The down-regulated genes included glycoprotein-4-beta galactosyltransferase 2 (<it>Ggbt2</it>), which is involved in the regulation of extracellular matrix organization and biogenesis.</p> <p>Conclusions</p> <p>The data in the present study demonstrate a close association between specific gene expression in mural granulosa cells and the developmental competence of oocytes. This finding suggests that the most differentially expressed gene, lysyl oxidase, may be a candidate biomarker of oocyte health and useful for the selection of good quality oocytes for assisted reproduction.</p

    Application and Mechanism of High-Sensitivity Indicator Film for Monitoring Fish Freshness

    The sensitivity of freshness indicator labels/films has become an important research direction of intelligent food packaging. In this study, a high-sensitivity indicator film containing gelatin and Fe2+ was prepared by electrospinning using blueberry anthocyanins as the indicator and zein as the matrix for monitoring fish freshness. The validity and sensitivity of the indicator film for detecting the freshness of silver carp were tested and the potential mechanism was elucidated. The results of pH sensitivity, ammonia sensitivity and anthocyanin release showed that the addition of gelatin and Fe2+ improved the sensitivity of the indicator film to pH and ammonia, and contributed to better binding of the anthocyanins in the film. There was a strong correlation between the color response (P = (L* + a* + b* + R + G + B)/a*) of the film and the content of total volatile basic nitrogen (TVB-N) content in fish meat as a freshness indicator (R2 > 0.98). In conclusion, the prepared indicator film can effectively monitor fish freshness, and the hydrogen bond interactions between anthocyanins and gelatin/Fe2+ may affect the color response characteristics and sensitivity of the indicator film

    Defining Global Neuroendocrine Gene Expression Patterns Associated with Reproductive Seasonality in Fish

    Many vertebrates, including the goldfish, exhibit seasonal reproductive rhythms, which are a result of interactions between external environmental stimuli and internal endocrine systems in the hypothalamo-pituitary-gonadal axis. While it is long believed that differential expression of neuroendocrine genes contributes to establishing seasonal reproductive rhythms, no systems-level investigation has yet been conducted. gamma2 receptor, calmodulin, and aromatase b by independent samplings of goldfish brains from six seasonal time points and real-time PCR assays.Using both theoretical and experimental strategies, we report for the first time global gene expression patterns throughout a breeding season which may account for dynamic neuroendocrine regulation of seasonal reproductive development

    SARS-CoV-2 bivalent mRNA vaccine with broad protection against variants of concern

    IntroductionThe severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Omicron variant has rapidly spread around the globe. With a substantial number of mutations in its Spike protein, the SARS-CoV-2 Omicron variant is prone to immune evasion and led to the reduced efficacy of approved vaccines. Thus, emerging variants have brought new challenges to the prevention of COVID-19 and updated vaccines are urgently needed to provide better protection against the Omicron variant or other highly mutated variants.Materials and methodsHere, we developed a novel bivalent mRNA vaccine, RBMRNA-405, comprising a 1:1 mix of mRNAs encoding both Delta-derived and Omicron-derived Spike proteins. We evaluated the immunogenicity of RBMRNA-405 in BALB/c mice and compared the antibody response and prophylactic efficacy induced by monovalent Delta or Omicron-specific vaccine with the bivalent RBMRNA-405 vaccine in the SARSCoV-2 variant challenge.ResultsResults showed that the RBMRNA-405 vaccine could generate broader neutralizing antibody responses against both Wuhan-Hu-1 and other SARS-CoV-2 variants, including Delta, Omicron, Alpha, Beta, and Gamma. RBMRNA-405 efficiently blocked infectious viral replication and lung injury in both Omicron- and Delta-challenged K18-ACE2 mice.ConclusionOur data suggest that RBMRNA-405 is a promising bivalent SARS-CoV-2 vaccine with broad-spectrum efficacy for further clinical development

    Large expert-curated database for benchmarking document similarity detection in biomedical literature search

    Document recommendation systems for locating relevant literature have mostly relied on methods developed a decade ago. This is largely due to the lack of a large offline gold-standard benchmark of relevant documents that cover a variety of research fields such that newly developed literature search techniques can be compared, improved and translated into practice. To overcome this bottleneck, we have established the RElevant LIterature SearcH consortium consisting of more than 1500 scientists from 84 countries, who have collectively annotated the relevance of over 180 000 PubMed-listed articles with regard to their respective seed (input) article/s. The majority of annotations were contributed by highly experienced, original authors of the seed articles. The collected data cover 76% of all unique PubMed Medical Subject Headings descriptors. No systematic biases were observed across different experience levels, research fields or time spent on annotations. More importantly, annotations of the same document pairs contributed by different scientists were highly concordant. We further show that the three representative baseline methods used to generate recommended articles for evaluation (Okapi Best Matching 25, Term Frequency-Inverse Document Frequency and PubMed Related Articles) had similar overall performances. Additionally, we found that these methods each tend to produce distinct collections of recommended articles, suggesting that a hybrid method may be required to completely capture all relevant articles. The established database server located at https://relishdb.ict.griffith.edu.au is freely available for the downloading of annotation data and the blind testing of new methods. We expect that this benchmark will be useful for stimulating the development of new powerful techniques for title and title/abstract-based search engines for relevant articles in biomedical research.Peer reviewe

    Biomarkers-based Biosensing and Bioimaging with Graphene for Cancer Diagnosis

    At the onset of cancer, specific biomarkers get elevated or modified in body fluids or tissues. Early diagnosis of these biomarkers can greatly improve the survival rate or facilitate effective treatment with different modalities. Potential nanomaterial-based biosensing and bioimaging are the main techniques in nanodiagnostics because of their ultra-high selectivity and sensitivity. Emerging graphene, including two dimensional (2D) graphene films, three dimensional (3D) graphene architectures and graphene hybrids (GHs) nanostructures, are attracting increasing interests in the field of biosensing and bioimaging. Due to their remarkable optical, electronic, and thermal properties; chemical and mechanical stability; large surface area; and good biocompatibility, graphene-based nanomaterials are applicable alternatives as versatile platforms to detect biomarkers at the early stage of cancer. Moreover, currently, extensive applications of graphene-based biosensing and bioimaging has resulted in promising prospects in cancer diagnosis. We also hope this review will provide critical insights to inspire more exciting researches to address the current remaining problems in this field

    Research on the combined effects of traditional Chinese medicine compound Based on physical movement Theory

    Methamphetamine (METH) is a powerful psychostimulant. Chronic obsessive and compulsive usage of METH caused great harm to human health (physical and psychological) and social stability issues all over the world. Currently, no drugs have been used to treat and prevent METH addiction. Our previous studies demonstrated that Chinese medicine compound (JDYH) can inhibit METH-induced sensitization in mice. To investigate the combined effects of JDYH on physical movement in C57BL/6 male mice. In the experiment, mice chronic treatment with JDYH (14.12-56.48 g/kg) for 28-30 days, physical movement activity and body weight was measured. The body weight and physical movement have no change after being treated with low-dose (14.12 g/kg) and mid-dose (28.24 g/kg) of chronic treatment with JDYH. These results suggest that JDYH (14.12-28.24 g/kg) for 21days were not inhibit physical movement in mice
