    Combined Preconditioning and Postconditioning Provides Synergistic Protection against Liver Ischemic Reperfusion Injury

    Hepatic Ischemia and Reperfusion Injury (IRI) is a major cause of liver damage during liver surgery and transplantation. Ischemic preconditioning and postconditioning are strategies that can reduce IRI. In this study, different combined types of pre- and postconditioning procedures were tested in a murine warm hepatic IRI model to evaluate their protective effects. Proanthocyanidins derived from grape seed was used before ischemia process as pharmacological preconditioning to combine with technical preconditioning and postconditioning. Three pathways related to IRI, including reactive oxygen species (ROS) generation, pro-inflammatory cytokines release and hypoxia responses were examined in hepatic IRI model. Individual and combined pre- and postconditioning protocols significantly reduce liver injury by decreasing the liver ROS and cytokine levels, as well as enhancing the hypoxia tolerance response. Our data also suggested that in addition to individual preconditioning or postconditioning, the combination of these two treatments could reduce liver ischemia/reperfusion injury more effectively by increasing the activity of ROS scavengers and antioxidants. The utilization of grape seed proanthocyanidins (GSP) could improve the oxidation resistance in combined pre- and postconditioning groups. The combined protocol also further increased the liver HIF-1 alpha protein level, but had no effect on pro-inflammatory cytokines release compared to solo treatment

    Effective Spectral Emissivity of Gas Turbine Blades for Optical Pyrometry

    Turbine blade temperature measurements are important for monitoring the turbine engine performance to protect the hot components from damage due to excess temperatures. However, the reflected radiation from the blades and the surrounding environment complicate the blade temperature measurements by optical pyrometers. This study characterizes the effect of the reflected radiation on the effective spectral emissivity of a three-dimensional turbine blade in a confined turbine space for optical pyrometry temperature measurements. The effective spectral emissivity distribution on a threedimensional blade was numerically determined for various wavelengths (0.8-15.0 lm) and actual blade surface emissivities for a specified turbine blade model. When the actual spectral emissivity of the blade surface is assumed to be 0.5, the effective spectral emissivity varies from 0.5 to 0.538 at the longer wavelength of 10.0 lm and further increases from 0.5 to 1.396 at the shorter wavelength of 0.9 lm. The results show that the effective emissivity distributions at shorter wavelengths differ greatly from those at longer wavelengths. There are also obvious differences between the effective spectral emissivity and the actual surface emissivity at shorter wavelengths. The effect of the effective emissivity on the temperature measurement accuracy, when using the optical pyrometry, was also investigated for various wavelengths (0.8-15.0 lm). The results show that the radiation reflected from the blades has less effect on the temperature measurements than on the effective emissivity, especially at the shorter wavelengths of 0.8-3.0 lm. However, the temperature measurements still need to be corrected using the effective spectral emissivity to improve the temperature calculation accuracy. This analysis provides guidelines for choosing the optimum measurement wavelengths for optical pyrometry in turbine engines

    Numerical studies on wake and turbulence characteristics of aquaculture nets

    This paper aims to understand the drag coefficient discrepancy between the equivalent-twine and twisted-twine nets based on their wake and turbulence characteristics. To that end, we conduct unsteady Reynolds-averaged Navier-Stokes (URANS) and the second-moment (Reynolds stress, RSM) simulations at a Reynolds number, Re=4.5×103, based on the effective diameter of the net twine, which corresponds to the subcritical flow regime. Then, the vortex structures and the turbulence statistics are assessed at AOA=90°. The results highlight that the wake interactions for the twisted-twine net are relatively strong compared to the equivalent-twine net, due to the disturbance of the helixes on the twisted twines. In comparison to the classical Karman vortex, the overall vortex shedding of these two nets is well organized. Symmetric vortices form behind the equivalent-twine net, while single vortices form behind the twisted-twine net. Moreover, the Reynolds normal and shear stresses show symmetric and anti-symmetric profiles. The addition of helixes to smooth circular cylinders changes the flow development, leading to a decrease of turbulence kinetic energy. With this understanding, engineers need to be carefully select the net type for preliminary design of marine aquaculture cages to avoid over- or underestimation of the drag forces

    Developmental iodine deficiency and hypothyroidism impair neural development in rat hippocampus: involvement of doublecortin and NCAM-180

    <p>Abstract</p> <p>Background</p> <p>Developmental iodine deficiency results in inadequate thyroid hormone (TH), which damages the hippocampus. Here, we explored the roles of hippocampal doublecortin and neural cell adhesion molecule (NCAM)-180 in developmental iodine deficiency and hypothyroidism.</p> <p>Methods</p> <p>Two developmental rat models were established with either an iodine-deficient diet, or propylthiouracil (PTU)-adulterated water (5 ppm or 15 ppm) to impair thyroid function, in pregnant rats from gestational day 6 until postnatal day (PN) 28. Silver-stained neurons and protein levels of doublecortin and NCAM-180 in several hippocampal subregions were assessed on PN14, PN21, PN28, and PN42.</p> <p>Results</p> <p>The results show that nerve fibers in iodine-deficient and 15 ppm PTU-treated rats were injured on PN28 and PN42. Downregulation of doublecortin and upregulation of NCAM-180 were observed in iodine-deficient and 15 ppm PTU-treated rats from PN14 on. These alterations were irreversible by the restoration of serum TH concentrations on PN42.</p> <p>Conclusion</p> <p>Developmental iodine deficiency and hypothyroidism impair the expression of doublecortin and NCAM-180, leading to nerve fiber malfunction and thus impairments in hippocampal development.</p

    High Incidence and Endemic Spread of NDM-1-Positive Enterobacteriaceae in Henan Province, China

    The emergence and spread of New Delhi metallo-β-lactamase 1 (NDM-1)-producing carbapenem-resistant Enterobacteriaceae (CRE) present an urgent threat to human health. In China, the blaNDM-1 gene has been reported mostly in Acinetobacter spp. but is rarely found in Enterobacteriaceae. Here, we report a high incidence and endemic spread of NDM-1-producing CRE in Henan Province in China. Sixteen (33.3%) of the 48 CRE isolates obtained from patients during June 2011 to July 2012 were positive for blaNDM-1, and the gene was found to be carried on plasmids of various sizes (∼55 to ∼360 kb). These plasmids were readily transferrable to recipient Escherichia coli by conjugation, conferred resistance to multiple antibiotics, and belonged to multiple replicon types. The blaNDM-1-positive CRE isolates were genetically diverse, and six new multilocus sequence typing (MLST) sequence types were linked to the carriage of NDM-1. Five of the isolates were classified as extensively drug-resistant (XDR) isolates, four of which also carried the fosA3 gene conferring resistance to fosfomycin, an alternative drug for treating infections by CRE. In each blaNDM-1-positive CRE isolate, the blaNDM-1 gene was downstream of an intact ISAba125 element and upstream of the bleMBL gene. Furthermore, gene environment analysis suggested the possible transmission of blaNDM-1-containing sequences from Acinetobacter spp. to Klebsiella pneumoniae and Klebsiella oxytoca. These findings reveal the emergence and active transmission of NDM-1-positive CRE in China and underscore the need for heightened measures to control their further spread

    Large expert-curated database for benchmarking document similarity detection in biomedical literature search

    Document recommendation systems for locating relevant literature have mostly relied on methods developed a decade ago. This is largely due to the lack of a large offline gold-standard benchmark of relevant documents that cover a variety of research fields such that newly developed literature search techniques can be compared, improved and translated into practice. To overcome this bottleneck, we have established the RElevant LIterature SearcH consortium consisting of more than 1500 scientists from 84 countries, who have collectively annotated the relevance of over 180 000 PubMed-listed articles with regard to their respective seed (input) article/s. The majority of annotations were contributed by highly experienced, original authors of the seed articles. The collected data cover 76% of all unique PubMed Medical Subject Headings descriptors. No systematic biases were observed across different experience levels, research fields or time spent on annotations. More importantly, annotations of the same document pairs contributed by different scientists were highly concordant. We further show that the three representative baseline methods used to generate recommended articles for evaluation (Okapi Best Matching 25, Term Frequency-Inverse Document Frequency and PubMed Related Articles) had similar overall performances. Additionally, we found that these methods each tend to produce distinct collections of recommended articles, suggesting that a hybrid method may be required to completely capture all relevant articles. The established database server located at https://relishdb.ict.griffith.edu.au is freely available for the downloading of annotation data and the blind testing of new methods. We expect that this benchmark will be useful for stimulating the development of new powerful techniques for title and title/abstract-based search engines for relevant articles in biomedical research.Peer reviewe
