A Pilot Study Comparing HPV-Positive and HPV-Negative Head and Neck Squamous Cell Carcinomas by Whole Exome Sequencing.
Background. Next-generation sequencing of cancers has identified important therapeutic targets and biomarkers. The goal of this pilot study was to compare the genetic changes in a human papillomavirus- (HPV-)positive and an HPV-negative head and neck tumor. Methods. DNA was extracted from the blood and primary tumor of a patient with an HPV-positive tonsillar cancer and those of a patient with an HPV-negative oral tongue tumor. Exome enrichment was performed using the Agilent SureSelect All Exon Kit, followed by sequencing on the ABI SOLiD platform. Results. Exome sequencing revealed slightly more mutations in the HPV-negative tumor (73) in contrast to the HPV-positive tumor (58). Multiple mutations were noted in zinc finger genes (ZNF3, 10, 229, 470, 543, 616, 664, 638, 716, and 799) and mucin genes (MUC4, 6, 12, and 16). Mutations were noted in MUC12 in both tumors. Conclusions. HPV-positive HNSCC is distinct from HPV-negative disease in terms of evidence of viral infection, p16 status, and frequency of mutations. Next-generation sequencing has the potential to identify novel therapeutic targets and biomarkers in HNSCC
A case report and genetic characterization of a massive acinic cell carcinoma of the parotid with delayed distant metastases.
We describe the presentation, management, and clinical outcome of a massive acinic cell carcinoma of the parotid gland. The primary tumor and blood underwent exome sequencing which revealed deletions in CDKN2A as well as PPP1R13B, which induces p53. A damaging nonsynonymous mutation was noted in EP300, a histone acetylase which plays a role in cellular proliferation. This study provides the first insights into the genetic underpinnings of this cancer. Future large-scale efforts will be necessary to define the mutational landscape of salivary gland malignancies to identify therapeutic targets and biomarkers of treatment failure

A new cell primo-culture method for freshwater benthic diatom communities
A new cell primo-culture method was developed for the benthic diatom community isolated from biofilm sampled in rivers. The approach comprised three steps: (1) scraping biofilm from river pebbles, (2) diatom isolation
from biofilm, and (3) diatom community culture. With a view to designing a method able to stimulate the growth of diatoms, to limit the development of other microorganisms, and to maintain in culture a community similar to the original natural one, different factors were tested in step 3:
cell culture medium (Chu No 10 vs Freshwater “WC” medium modified), cell culture vessel, and time of culture. The results showed that using Chu No 10 medium in an Erlenmeyer flask for cell culture was the optimal method,
producing enough biomass for ecotoxicological tests as well as minimising development of other microorganisms. After 96 h of culture, communities differed from the original communities sampled in the two rivers studied.
Species tolerant of eutrophic or saprobic conditions were favoured during culture. This method of diatom community culture affords the opportunity to assess, in vitro, the effects of different chemicals or effluents (water samples andindustrial effluents) on diatom communities, as well as on diatom cells, from a wide range of perspectives
Intra-hospital transport of neonatal intensive care patients: risk factors for complications
OBJECTIVE: Evaluate factors associated with hypothermia and increased need of oxygen and/or ventilatory support during intra-hospital transport of neonatal intensive care patients. METHODS: Prospective study of infants admitted to a single neonatal unit in need of at least one intra-hospital transport during weekdays, from 8:00AM to 05:00PM, from January 1997 to December 2000. Factors associated with hypothermia and increased need of oxygen and/or need of ventilatory support during and up to two hours after transport were studied by regression analyses. RESULTS: During the study period, 502 transports were analyzed. At the time of transport, the neonates had a mean weight and gestational age of, respectively, 2,000g and 35 weeks, and they were 22 days old. The main reasons for transport were: surgery and image exams. Hypothermia occurred in 17% of the transported infants and increased need of oxygen and/or ventilatory support in 7%. Factors associated with hypothermia were: duration of transportation >3h (OR=2.1; 95%CI=1.2-3.6), neurologic malformation (OR=1.7; 95%CI=1.1-2.5), transport performed in 1997 (OR=1.7; 95%CI=1.1-2.6) and weight at time of transport >3,500g (OR=0.3; 95%CI=0.16-0.68). Factors associated with increased need of oxygen and/or ventilation support were: gestational age at birth in weeks (OR=0.9; 95%CI=0.8-0.9), age in days at transportation (OR=1.0; 95%CI=1.0-1.1) and presence of gastrointestinal and genitourinary malformation (OR=3.1; 95%CI=1.6-6.2). CONCLUSIONS: Complications related to neonatal intra-hospital transports are frequent and associated with the patients characteristics and transport conditions.OBJETIVO: Estudar os fatores associados à hipotermia e ao aumento da necessidade de oxigênio e/ou suporte ventilatório durante o transporte intra-hospitalar de pacientes internados em Unidade de Terapia Intensiva neonatal. MÉTODOS: Estudo prospectivo de todos os pacientes internados na unidade neonatal que necessitaram de transporte intra-hospitalar de janeiro de 1997 a dezembro de 2000, entre segundas-feiras e sextas-feiras, das 8h às 17h. Fatores associados à hipotermia e ao aumento da necessidade de oxigênio e/ou de suporte ventilatório durante e até duas horas após o transporte foram estudados por meio de regressão logística. RESULTADOS: Foram realizados 502 transportes no período. Os pacientes tinham em média 2.000g, 35 semanas de idade gestacional ao nascer e 22 dias de vida. As principais indicações do transporte foram: cirurgia e realização de exames de imagem. A hipotermia ocorreu em 17% dos transportes e o aumento da necessidade de oxigênio e/ou de suporte ventilatório em 7%. Fatores associados à hipotermia foram: duração do transporte >3h (OR=2,1; IC95%=1,2-3,6), presença de malformações neurológicas (OR=1,7; IC95%=1,1-2,5), transporte realizado em 1997 (OR=1,7; IC95%=1,1-2,6) e peso no transporte >3.500g (OR=0,3; IC95%=0,16-0,68). Fatores de risco para o aumento da necessidade de oxigênio e/ou de suporte ventilatório foram: idade gestacional ao nascimento em semanas (OR=0,9; IC95%=0,8-0,9), idade em dias no transporte (OR=1,0; IC95%=1,0-1,1) e presença de malformações gastrintestinais e geniturinárias (OR=3,1; IC95%=1,6-6,2). Multi-messenger observations of a binary neutron star merger
On 2017 August 17 a binary neutron star coalescence candidate (later designated GW170817) with merger time 12:41:04 UTC was observed through gravitational waves by the Advanced LIGO and Advanced Virgo detectors. The Fermi Gamma-ray Burst Monitor independently detected a gamma-ray burst (GRB 170817A) with a time delay of ~1.7 s with respect to the merger time. From the gravitational-wave signal, the source was initially localized to a sky region of 31 deg2 at a luminosity distance of 40+8-8 Mpc and with component masses consistent with neutron stars. The component masses were later measured to be in the range 0.86 to 2.26 Mo. An extensive observing campaign was launched across the electromagnetic spectrum leading to the discovery of a bright optical transient (SSS17a, now with the IAU identification of AT 2017gfo) in NGC 4993 (at ~40 Mpc) less than 11 hours after the merger by the One- Meter, Two Hemisphere (1M2H) team using the 1 m Swope Telescope. The optical transient was independently detected by multiple teams within an hour. Subsequent observations targeted the object and its environment. Early ultraviolet observations revealed a blue transient that faded within 48 hours. Optical and infrared observations showed a redward evolution over ~10 days. Following early non-detections, X-ray and radio emission were discovered at the transient’s position ~9 and ~16 days, respectively, after the merger. Both the X-ray and radio emission likely arise from a physical process that is distinct from the one that generates the UV/optical/near-infrared emission. No ultra-high-energy gamma-rays and no neutrino candidates consistent with the source were found in follow-up searches. These observations support the hypothesis that GW170817 was produced by the merger of two neutron stars in NGC4993 followed by a short gamma-ray burst (GRB 170817A) and a kilonova/macronova powered by the radioactive decay of r-process nuclei synthesized in the ejecta
Detection of serum MMP-7 and MMP-9 in cholangiocarcinoma patients: evaluation of diagnostic accuracy
<p>Abstract</p> <p>Background</p> <p>Cholangiocarcinoma is an aggressive tumor with a tendency for local invasion and distant metastases. Timely diagnosis is very important because surgical resection (R0) remains the only hope for a cure. However, at present, there is no available tumor marker that can differentiate cholangiocarcinoma from benign bile duct disease. Previous studies have demonstrated that matrix metalloproteinase (MMP)-7 and MMP-9 are frequently expressed in cholangiocarcinoma specimens.</p> <p>Methods</p> <p>This study was designed to determine whether the serum levels of MMP-7 and MMP-9 can discriminate cholangiocarcinoma patients from benign biliary tract disease patients in comparison to carcinoembryonic antigen (CEA) and carbohydrate antigen 19-9 (CA19-9). We measured the level of CEA, CA19-9, MMP-7 and MMP-9 in the serum of 44 cholangiocarcinoma and 36 benign biliary tract diseases patients.</p> <p>Results</p> <p>Among the serum levels of CEA, CA19-9, MMP-7 and MMP-9, only the serum MMP-7 level was significantly higher in the patients with cholangiocarcinoma (8.9 ± 3.43 ng/ml) compared to benign biliary tract disease patients (5.9 ± 3.03 ng/ml) (<it>p </it>< 0.001). An receiver operating characteristic (ROC) curve analysis revealed that the detection of the serum MMP-7 level is reasonably accurate in differentiating cholangiocarcinoma from benign biliary tract disease patients (area under curve = 0.73; 95% CI = 0.614–0.848). While the areas under the curve of the ROC curves for CEA, CA19-9 and MMP-9 were 0.63 (95% CI = 0.501–0.760), 0.63 (95% CI = 0.491–0.761) and 0.59 (95% CI = 0.455–0.722), respectively.</p> <p>Conclusion</p> <p>Serum MMP-7 appears to be a valuable diagnostic marker in the discrimination of cholangiocarcinoma from benign biliary tract disease. Further prospective studies for serum MMP-7 measurement should be carried out to further investigate the potential of this molecule as a biomarker of cholangiocarcinoma.</p
Simultaneous Effects of Light Intensity and Phosphorus Supply on the Sterol Content of Phytoplankton
Sterol profiles of microalgae and their change with environmental conditions are of great interest in ecological food web research and taxonomic studies alike. Here, we investigated effects of light intensity and phosphorus supply on the sterol content of phytoplankton and assessed potential interactive effects of these important environmental factors on the sterol composition of algae. We identified sterol contents of four common phytoplankton genera, Scenedesmus, Chlamydomonas, Cryptomonas and Cyclotella, and analysed the change in sterol content with varying light intensities in both a high-phosphorus and a low-phosphorus approach. Sterol contents increased significantly with increasing light in three out of four species. Phosphorus-limitation reversed the change of sterol content with light intensity, i.e., sterol content decreased with increasing light at low phosphorus supply. Generally sterol contents were lower in low-phosphorus cultures. In conclusion, both light and phosphorus conditions strongly affect the sterol composition of algae and hence should be considered in ecological and taxonomic studies investigating the biochemical composition of algae. Data suggest a possible sterol limitation of growth and reproduction of herbivorous crustacean zooplankton during summer when high light intensities and low phosphorus supply decrease sterol contents of algae
Wettability Modification of Nanomaterials by Low-Energy Electron Flux
Controllable modification of surface free energy and related properties (wettability, hygroscopicity, agglomeration, etc.) of powders allows both understanding of fine physical mechanism acting on nanoparticle surfaces and improvement of their key characteristics in a number of nanotechnology applications. In this work, we report on the method we developed for electron-induced surface energy and modification of basic, related properties of powders of quite different physical origins such as diamond and ZnO. The applied technique has afforded gradual tuning of the surface free energy, resulting in a wide range of wettability modulation. In ZnO nanomaterial, the wettability has been strongly modified, while for the diamond particles identical electron treatment leads to a weak variation of the same property. Detailed investigation into electron-modified wettability properties has been performed by the use of capillary rise method using a few probing liquids. Basic thermodynamic approaches have been applied to calculations of components of solid–liquid interaction energy. We show that defect-free, low-energy electron treatment technique strongly varies elementary interface interactions and may be used for the development of new technology in the field of nanomaterials
‘You wouldn’t get that from watching TV!’: Exploring audience responses to virtual reality non-fiction in the home
Consumer virtual reality (VR) headsets (e.g. Oculus Go) have brought VR non-fiction (VRNF) within reach of at-home audiences. However, despite increase in VR hardware sales and enthusiasm for the platform among niche audiences at festivals, mainstream audience interest in VRNF is not yet proven. This is despite a growing body of critically acclaimed VRNF, some of which is freely available. In seeking to understand a lack of engagement with VRNF by mainstream audiences, we need to be aware of challenges relating to the discovery of content and bear in mind the cost, inaccessibility and known limitations of consumer VR technology. However, we also need to set these issues within the context of the wider relationships between technology, society and the media, which have influenced the uptake of new media technologies in the past. To address this work, this article provides accounts by members of the public of their responses to VRNF as experienced within their households. We present an empirical study – one of the first of its kind – exploring these questions through qualitative research facilitating diverse households to experience VRNF at home, over several months. We find considerable enthusiasm for VR as a platform for non-fiction, but we also find this enthusiasm tempered by ethical concerns relating to both the platform and the content, and a pervasive tension between the platform and the home setting. Reflecting on our findings, we suggest that VRNF currently fails to meet any ‘supervening social necessity’ (Winston, 1996, Technologies of Seeing: Photography, Cinematography and Television. British: BFI.) that would pave the way for widespread domestic uptake, and we reflect on future directions for VR in the home
