    Evidences pointing that bone marrow originating tumor cells with lung-colonizyng ability, rather than tumor cells remaining within the bone marrow, are related to a mesenchymal stem cell phenotype

    A critical challenge in the clinical management of osteosarcoma (OS) is the appearance of lung metastasis. This bone marrow ? associated tumor represents the most frequent bone tumor in pediatrics and young adult populations. In this context, 20% of OS patients are diagnosed with metastatic OS, but a high percentage of the remaining cases diagnosed as without metastasis could already present micrometastasis undetectable through conventional methods. Our previous results indicated that a differential gene expression distinguished OS cells with higher ability to home into the lungs. Interestingly, molecular differences were subtle at the level of cellular content but more prominent at the level of the secretory compartment. These molecular features were reproduced by a functional behavior relevant to a colonizing ability to the lungs. In order to gain insight into spatial arrangements of OS cells that diverged in their lung colonizing ability, that would shed light into advantages to colonize the lungs, and relate to metastatic mechanisms, we analyzed 3D cultures of OS cells that diverged in their lung homing behavior. We observed that OS cells that remain at the primary tumor site had a lesser ability to establish 3D growth, while cells leaving the tumor and colonizing the lungs established 3D growth successfully; this last feature was shared by mesenchymal stem cells (MSC). This would point that cell-cell contact was a prominent feature in lung colonizing cells, Since our previous results demonstrated that the secretome of divergent OS is the cellular compartment that mostly distinguished the ability to home into the lungs, we analyzed we analyzed GOs in the secretory compartment in divergent OS cells and bone marrow MSC. Related to a similarity between MSC and lung-colonizing OS cells, MSC share the original niche where the bone tumor arises, and related to possible closeness similarity between MSC and OS cells, we demonstrated that the cells that leave the primary tumor rather that the cells remaining at the primary niche of residence for OS, share similarity with MSC.Fil: Gutierrez, Luciana Mariel.     Use of a customized app to assess communication in children with ASD: AppTEA

    El uso de dispositivos digitales para tratar a personas con trastorno del espectro autista (TEA) tiene buena aceptación, pero hasta el momento no se han registrado investigaciones en Argentina que analicen empíricamente su aporte en cuanto a los comportamientos comunicativos. El objetivo de esta investigación consistió en evaluar la implicación de los dispositivos digitales por lo que respecta a dos parámetros conductuales: la postura y el foco atencional de niños con TEA. Para llevarlo a cabo se analizaron 211 sesiones (vídeos) de 11 niños (edad, Media = 6.45 años, DE = 1.30) durante la utilización de la aplicación AppTEA y dispositivos tradicionales en interacciones con profesionales, en el contexto de unas actividades terapéuticas. Como resultado se encontró que en las sesiones, los niveles y las tendencias de postura y el foco atencional fueron similares, independientemente de los dispositivos empleados. Por lo tanto, si bien las tecnologías y los softwares específicos pueden complementar y apoyar los objetivos terapéuticos, no pueden sustituir el cuidado humano y el apoyo social y terapéutico para los niños.Introduction: There are many well-established digital tools for the treatment of people with Autism Spectrum Disorder (ASD), but so far there no investigations in Argentina that have empirically analyzed how these tools contribute to communicative behaviors. Objectives: The objective was to evaluate the implications of digital devices for two behavioral parameters, the posture and attentional focus of children with ASD. Methods: 211 sessions with 11 children (Age, M= 6.45 years, SD = 1.30) were analyzed. The sessions consisted of the use of the AppTEA application and traditional devices in interactions with professionals, in the context of therapeutic activities. Results: It was found that in the sessions with and without the tablet, the levels and trends of posture and attention focus were similar, regardless of the devices used. Conclusion: Although specific technologies and software can complement and support therapeutic objectives, they cannot replace human care and the social and therapeutic support to children.Fil: Guzman, Guido Benjamin.     Stem: associated features in tumor cells able to colonize secondary tumor sites

    Osteosarcoma (OS), the most frequent bone tumor in pediatrics, presents critical clinical challenges in lung metastasis and chemoresistance emergence. Understanding OS switch into a metastatic phenotype and the interaction OS-stromal cells relevant in the new niche, would help in developing better diagnostic and therapeutic tools. In order to distinguish aspects that would allow OS cells to leave the bone niche and survive in a new tissue environment, we evaluated behavioral features acquired by OS cells with ability to establishsecondary tumor growth in the lungs, approaching the degree of differentiation, doxorubicin (doxo) exclusion and distribution properties and molecular signatures. Our results indicate that lung-colonizing OS cells diminished its osteoblastic potential while modified the intracellular localization of chemodrugs. In this way, doxo switched from a nuclear to a cytoplasmatic distribution in cells with lung colonizing ability (0,884±0,015 SAOS2;0,546±0,131 LM7). These features coincided with a higher level of expression of stemrelated genes and lower expression of differentiation-associated markers even at basal conditions in the metastatic cells. On the other hand, the higher osteogenic activity of OS cells with non-colonizing features was even reflected as a paracrine osteo-inductive effect. In addition, OS cells with high and low lung-colonizing capacities have opposite impact in mesenchymal stem cells (MSCs). Further, OS cells colonized-mouse lungs had a greater chemoattractive induction on MSCs. A major acquisition in tumor cells with metastatic features is a switch into a stem-like state that could favor their survival in the pulmonary niche, opening new possibilities for specific chemotherapeutic schemes.     Mapping development and health effects of cooking with solid fuels in low-income and middle-income countries, 2000-18 : a geospatial modelling study

    Background More than 3 billion people do not have access to clean energy and primarily use solid fuels to cook. Use of solid fuels generates household air pollution, which was associated with more than 2 million deaths in 2019. Although local patterns in cooking vary systematically, subnational trends in use of solid fuels have yet to be comprehensively analysed. We estimated the prevalence of solid-fuel use with high spatial resolution to explore subnational inequalities, assess local progress, and assess the effects on health in low-income and middle-income countries (LMICs) without universal access to clean fuels.Methods We did a geospatial modelling study to map the prevalence of solid-fuel use for cooking at a 5 km x 5 km resolution in 98 LMICs based on 2.1 million household observations of the primary cooking fuel used from 663 population-based household surveys over the years 2000 to 2018. We use observed temporal patterns to forecast household air pollution in 2030 and to assess the probability of attaining the Sustainable Development Goal (SDG) target indicator for clean cooking. We aligned our estimates of household air pollution to geospatial estimates of ambient air pollution to establish the risk transition occurring in LMICs. Finally, we quantified the effect of residual primary solid-fuel use for cooking on child health by doing a counterfactual risk assessment to estimate the proportion of deaths from lower respiratory tract infections in children younger than 5 years that could be associated with household air pollution.Findings Although primary reliance on solid-fuel use for cooking has declined globally, it remains widespread. 593 million people live in districts where the prevalence of solid-fuel use for cooking exceeds 95%. 66% of people in LMICs live in districts that are not on track to meet the SDG target for universal access to clean energy by 2030. Household air pollution continues to be a major contributor to particulate exposure in LMICs, and rising ambient air pollution is undermining potential gains from reductions in the prevalence of solid-fuel use for cooking in many countries. We estimated that, in 2018, 205000 (95% uncertainty interval 147000-257000) children younger than 5 years died from lower respiratory tract infections that could be attributed to household air pollution.Interpretation Efforts to accelerate the adoption of clean cooking fuels need to be substantially increased and recalibrated to account for subnational inequalities, because there are substantial opportunities to improve air quality and avert child mortality associated with household air pollution.     Analysis of shared heritability in common disorders of the brain

    ience, this issue p. eaap8757 Structured Abstract INTRODUCTION Brain disorders may exhibit shared symptoms and substantial epidemiological comorbidity, inciting debate about their etiologic overlap. However, detailed study of phenotypes with different ages of onset, severity, and presentation poses a considerable challenge. Recently developed heritability methods allow us to accurately measure correlation of genome-wide common variant risk between two phenotypes from pools of different individuals and assess how connected they, or at least their genetic risks, are on the genomic level. We used genome-wide association data for 265,218 patients and 784,643 control participants, as well as 17 phenotypes from a total of 1,191,588 individuals, to quantify the degree of overlap for genetic risk factors of 25 common brain disorders. RATIONALE Over the past century, the classification of brain disorders has evolved to reflect the medical and scientific communities' assessments of the presumed root causes of clinical phenomena such as behavioral change, loss of motor function, or alterations of consciousness. Directly observable phenomena (such as the presence of emboli, protein tangles, or unusual electrical activity patterns) generally define and separate neurological disorders from psychiatric disorders. Understanding the genetic underpinnings and categorical distinctions for brain disorders and related phenotypes may inform the search for their biological mechanisms. RESULTS Common variant risk for psychiatric disorders was shown to correlate significantly, especially among attention deficit hyperactivity disorder (ADHD), bipolar disorder, major depressive disorder (MDD), and schizophrenia. By contrast, neurological disorders appear more distinct from one another and from the psychiatric disorders, except for migraine, which was significantly correlated to ADHD, MDD, and Tourette syndrome. We demonstrate that, in the general population, the personality trait neuroticism is significantly correlated with almost every psychiatric disorder and migraine. We also identify significant genetic sharing between disorders and early life cognitive measures (e.g., years of education and college attainment) in the general population, demonstrating positive correlation with several psychiatric disorders (e.g., anorexia nervosa and bipolar disorder) and negative correlation with several neurological phenotypes (e.g., Alzheimer's disease and ischemic stroke), even though the latter are considered to result from specific processes that occur later in life. Extensive simulations were also performed to inform how statistical power, diagnostic misclassification, and phenotypic heterogeneity influence genetic correlations. CONCLUSION The high degree of genetic correlation among many of the psychiatric disorders adds further evidence that their current clinical boundaries do not reflect distinct underlying pathogenic processes, at least on the genetic level. This suggests a deeply interconnected nature for psychiatric disorders, in contrast to neurological disorders, and underscores the need to refine psychiatric diagnostics. Genetically informed analyses may provide important "scaffolding" to support such restructuring of psychiatric nosology, which likely requires incorporating many levels of information. By contrast, we find limited evidence for widespread common genetic risk sharing among neurological disorders or across neurological and psychiatric disorders. We show that both psychiatric and neurological disorders have robust correlations with cognitive and personality measures. Further study is needed to evaluate whether overlapping genetic contributions to psychiatric pathology may influence treatment choices. Ultimately, such developments may pave the way toward reduced heterogeneity and improved diagnosis and treatment of psychiatric disorders

    Mapping geographical inequalities in access to drinking water and sanitation facilities in low-income and middle-income countries, 2000-17

    Background: Universal access to safe drinking water and sanitation facilities is an essential human right, recognised in the Sustainable Development Goals as crucial for preventing disease and improving human wellbeing. Comprehensive, high-resolution estimates are important to inform progress towards achieving this goal. We aimed to produce high-resolution geospatial estimates of access to drinking water and sanitation facilities. Methods: We used a Bayesian geostatistical model and data from 600 sources across more than 88 low-income and middle-income countries (LMICs) to estimate access to drinking water and sanitation facilities on continuous continent-wide surfaces from 2000 to 2017, and aggregated results to policy-relevant administrative units. We estimated mutually exclusive and collectively exhaustive subcategories of facilities for drinking water (piped water on or off premises, other improved facilities, unimproved, and surface water) and sanitation facilities (septic or sewer sanitation, other improved, unimproved, and open defecation) with use of ordinal regression. We also estimated the number of diarrhoeal deaths in children younger than 5 years attributed to unsafe facilities and estimated deaths that were averted by increased access to safe facilities in 2017, and analysed geographical inequality in access within LMICs. Findings: Across LMICs, access to both piped water and improved water overall increased between 2000 and 2017, with progress varying spatially. For piped water, the safest water facility type, access increased from 40·0% (95% uncertainty interval [UI] 39·4–40·7) to 50·3% (50·0–50·5), but was lowest in sub-Saharan Africa, where access to piped water was mostly concentrated in urban centres. Access to both sewer or septic sanitation and improved sanitation overall also increased across all LMICs during the study period. For sewer or septic sanitation, access was 46·3% (95% UI 46·1–46·5) in 2017, compared with 28·7% (28·5–29·0) in 2000. Although some units improved access to the safest drinking water or sanitation facilities since 2000, a large absolute number of people continued to not have access in several units with high access to such facilities (>80%) in 2017. More than 253 000 people did not have access to sewer or septic sanitation facilities in the city of Harare, Zimbabwe, despite 88·6% (95% UI 87·2–89·7) access overall. Many units were able to transition from the least safe facilities in 2000 to safe facilities by 2017; for units in which populations primarily practised open defecation in 2000, 686 (95% UI 664–711) of the 1830 (1797–1863) units transitioned to the use of improved sanitation. Geographical disparities in access to improved water across units decreased in 76·1% (95% UI 71·6–80·7) of countries from 2000 to 2017, and in 53·9% (50·6–59·6) of countries for access to improved sanitation, but remained evident subnationally in most countries in 2017. Interpretation: Our estimates, combined with geospatial trends in diarrhoeal burden, identify where efforts to increase access to safe drinking water and sanitation facilities are most needed. By highlighting areas with successful approaches or in need of targeted interventions, our estimates can enable precision public health to effectively progress towards universal access to safe water and sanitation

    Production of He-4 and (4) in Pb-Pb collisions at root(NN)-N-S=2.76 TeV at the LHC

    Results on the production of He-4 and (4) nuclei in Pb-Pb collisions at root(NN)-N-S = 2.76 TeV in the rapidity range vertical bar y vertical bar <1, using the ALICE detector, are presented in this paper. The rapidity densities corresponding to 0-10% central events are found to be dN/dy4(He) = (0.8 +/- 0.4 (stat) +/- 0.3 (syst)) x 10(-6) and dN/dy4 = (1.1 +/- 0.4 (stat) +/- 0.2 (syst)) x 10(-6), respectively. This is in agreement with the statistical thermal model expectation assuming the same chemical freeze-out temperature (T-chem = 156 MeV) as for light hadrons.     A challenge in osteosarcoma (OS) is lung metastasis occurrence. OS, a bone marrow (BM)–associated tumor, is the most frequent pediatric bone tumor. A large percentage of patients diagnosed without metastasis would already present micrometastasis non-detectable by conventional methods. To identify metastasis associated-molecular signatures, we approached proteomics analysis demonstrating differential gene expression distinguishing OS cells with divergent lung-homing ability. Interestingly, subtle molecular differences distinguished the cellular content of both OS cell types. Major differences occurred in the secretome, and were associated to a functional behavior relevant to lungcolonization. To gain insight into spatial arrangements, we analyzed non-scaffold, anchorage-independent three-dimentional (3D) OS cell organizations. This would allow a more accurate comprehension of advantages to metastasize. We demonstrated that OS cells homing into the lungs had an increased ability to establish 3D structures, anattribute shared with mesenchymal stem cells (MSC). This would point that cellto-cell contact is a prominent feature in lung-colonizing cells. Since our previous results demonstrated that the secretome of divergent OS cells mostly distinguished lung-homing ability, we analyzed gene ontology (GO) terms in the secretome from OS cells and BM-derived MSC. MSC share the original OS niche. Surprisingly and related to cell hierarchy, we demonstrated that OS cells leaving the primary tumor rather than those remaining at the primary niche, share molecular and GO terms with MSC. Further, lung-colonizing OS cells had higher levels of expression of stem-related genes and lower expression of differentiation markers.
