    Quality assessment of truffle-inoculated seedlings in Italy: proposing revised parameters for certification

    Aim of study: the main aims of this study were to evaluate the quality of truffle-inoculated seedlings produced by commercial nurseries in Italy and to identify their minimum requisites in terms of plant age, health, homogeneity, and cut-off percentage of inoculated Tuber and non-Tuber ectomycorrhizae, based on the analysis of an extensive sample of seedlings subjected to quality control and certification.Area of study: truffle-inoculated seedlings produced by Italian commercial nurseries.Material and Methods: analysis of truffle-inoculated seedlings for health and quality standards; recording of presence of inoculated Tuber spp. and other concurrent fungi according to the official Italian method for certification; selective amplification of ectomycorrhizal DNA by PCR species-specific primers.Main results: We showed that mycorrhization levels in truffle-inoculated seedlings increased with time after truffle-spore inoculation. The highest mean percentage of the inoculated Tuber spp., but also the highest presence of contaminants, were recorded after three years. The mycorrhization level of Tuber melanosporum and T. aestivum was higher in Corylus and Ostrya seedlings than in Q. ilex and Q. pubescens, but the latter two host species showed the lowest presence of other ectomycorrhizal fungi. Mycorrhization level distribution in truffle-inoculated seedlings of suitable batches differed very little from the distribution in only all suitable seedlings. Truffle seedlings with other Tuber spp. were very few and even absent after three years.     Characterizing microbial communities associated with northern root-knot nematode (Meloidogyne hapla) occurrence and soil health

    The northern root-knot nematode (Meloidogyne hapla) causes extensive damage to agricultural crops globally. In addition, M. hapla populations with no known genetic or morphological differences exhibit parasitic variability (PV) or reproductive potential based on soil type. However, why M. hapla populations from mineral soil with degraded soil health conditions have a higher PV than populations from muck soil is unknown. To improve our understanding of soil bio-physicochemical conditions in the environment where M. hapla populations exhibited PV, this study characterized the soil microbial community and core- and indicator-species structure associated with M. hapla occurrence and soil health conditions in 15 Michigan mineral and muck vegetable production fields. Bacterial and fungal communities in soils from where nematodes were isolated were characterized with high throughput sequencing of 16S and internal transcribed spacer (ITS) rDNA. Our results showed that M. hapla-infested, as well as disturbed and degraded muck fields, had lower bacterial diversity (observed richness and Shannon) compared to corresponding mineral soil fields or non-infested mineral fields. Bacterial and fungal community abundance varied by soil group, soil health conditions, and/or M. hapla occurrence. A core microbial community was found to consist of 39 bacterial and 44 fungal sub-operational taxonomic units (OTUs) across all fields. In addition, 25 bacteria were resolved as indicator OTUs associated with M. hapla presence or absence, and 1,065 bacteria as indicator OTUs associated with soil health conditions. Out of the 1,065 bacterial OTUs, 73.9% indicated stable soil health, 8.4% disturbed, and 0.4% degraded condition; no indicators were common to the three categories. Collectively, these results provide a foundation for an in-depth understanding of the environment where M. hapla exists and conditions associated with parasitic variability

    Search for stopped gluinos in pp collisions at ?s=7TeV

    The results of the first search for long-lived gluinos produced in 7 TeV pp collisions at the CERN Large Hadron Collider are presented. The search looks for evidence of long-lived particles that stop in the CMS detector and decay in the quiescent periods between beam crossings. In a dataset with a peak instantaneous luminosity of 1×1032cm-2s-1, an integrated luminosity of 10pb-1, and a search interval corresponding to 62 hours of LHC operation, no significant excess above background was observed. Limits at the 95% confidence level on gluino pair production over 13 orders of magnitude of gluino lifetime are set. For a mass difference m g-m?10>100GeV/c2, and assuming BR(gâ€șg?10)= 100%, mg<370GeV/c2 are excluded for lifetimes from 10”s to 1000 s. © 2010 The American Physical Society

    Study of Z boson production in PbPb collisions at √s NN=2.76TeV

The number of opposite-sign muon pairs observed in the 60-120GeV/c2 invariant mass range is 39, corresponding to a yield per unit of rapidity (y) and per minimum bias event of [33.8±5.5(stat)±4.4(syst)]×10-8, in the |y|<2.0 range. Rapidity, transverse momentum, and centrality dependencies are also measured.
