    EFFECT OF THERMAL PROCESSING ON ANTIBACTERIAL ACTIVITY OF MULTIFLORAL HONEYS

    In this study, the effects of thermal processing in the antibacterial activity of four multifloral honeys were determined. The thermal treatments were carried out at 30, 40, 50, 60, 70 and 80C, and the following characteristics were determined: total phenols and antibacterial activity of three gram-positive bacteria (Bacillus subtilis, Staphylococcus aureus and Listeria monocytogenes) and three gram-negative bacteria (Escherichia coli, Salmonella typhimurium and Pseudomonas aeruginosa). The results showed that the behavior of total phenols in relation to temperature depended on the floral sources of honey. The honeys presented curves of either linear or quadratic responses of the antibacterial activity in relation to the thermal process, depending on the honey's floral sources and the kind of bacterium to inhibit. The results suggest that the honey's antibacterial activities and behavior with the temperature are different depending on the honey's floral sources and the kind of bacteria that they inhibit.Fundacao para a Ciencia e a Tecnologiainfo:eu-repo/semantics/publishedVersio

    Caracterización fitoquímica y capacidad antioxidante de especies de Briofitas colectadas en el NOA

    Las briofitas conforman un amplio grupo integrado por unas 22.000 especies deplantas pequeñas que crecen habitualmente en zonas húmedas sobre el suelo,troncos de árboles y rocas, son plantas embrionarias no vasculares que incluyenmusgos, hepáticas y antocerotes.No se han realizado muchos estudios de fitoquímica y bioactividades en briofitas, esto se debe a que son morfológicamente muy pequeñas y difíciles de recoger en grandes cantidades como muestra pura. Sin embargo, algunas de ellas han tenido tradicionalmente un uso medicinal.El objetivo de este trabajo es realizar una caracterización fitoquímica de extractos de 9 especies de briofitas colectadas en el Noroeste Argentino.Se prepararon extractos acuosos y etanólicos de las 9 especies a estudiar: Plagiochasma intermedium; Pleurochaete luteola; Marchantia papillata; Marchantiaplicata; Targionia hypophylla; Saitobryum lorentzii; Marchantia chenopoda; Anoectangium aestivum; Anoectangium euchloron. A estos extractos se les determinó el contenido de compuestos fenólicos utilizando el método colorimétrico de FolinCiocalteau, el contenido de flavonoides totales usando AlCl₃, y la capacidad antioxidante de los compuestos con el ensayo del radical catiónico ABTS.Los extractos etanólicos de Pleurochaete luteola, Marchantia papillata, Anoectangium aestivum y Anoectangium euchloron presentaron significativamente más compuestos fenólicos; además, Anoectangium aestivum, Anoectangium euchloron contenían más flavonoides que el resto de las especias estudiadas. Pleurochaete luteola y Anoectangium euchloron mostraron mayor capacidad antioxidante. En los extractos acuosos Plagiochasma intermedium y Marchantia papillata presentaron mayor contenido de compuestos fenólicos, pero en concentraciones menores a las de las muestras etanólicas. Anoectangium aestivum y Anoectangium euchloron tuvieron mayor contenido de flavonoides totales. Los extractos acuosos de Plagiochasma intermedium y Pleurochaete luteola resultaron muy buenos antioxidantes con potencia superior a los extractos etanólicos.Los resultados de este trabajo permitieron definir por primera vez que estas especies de briofitas contienen polifenoles y son potencialmente útiles como antioxidantes naturales.Fil: Jimenez, Ignacio.     Application of six detection methods for analysis of paralytic shellfish toxins in shellfish from four regions within Latin America

    With the move away from use of mouse bioassay (MBA) to test bivalve mollusc shellfish for paralytic shellfish poisoning (PSP) toxins, countries around the world are having to adopt non-animal-based alternatives that fulfil ethical and legal requirements. Various assays have been developed which have been subjected to single-laboratory and multi-laboratory validation studies, gaining acceptance as official methods of analysis and approval for use in some countries as official control testing methods. The majority of validation studies conducted to date do not, however, incorporate shellfish species sourced from Latin America. Consequently, this study sought to investigate the performance of five alternative PSP testing methods together with the MBA, comparing the PSP toxin data generated both qualitatively and quantitatively. The methods included a receptor binding assay (RBA), two liquid chromatography with fluorescence detection (LC-FLD) methods including both pre-column and post-column oxidation, liquid chromatography with tandem mass spectrometry (LC-MS/MS) and a commercial lateral flow assay (LFA) from Scotia. A total of three hundred and forty-nine shellfish samples from Argentina, Mexico, Chile and Uruguay were assessed. For the majority of samples, qualitative results compared well between methods. Good statistical correlations were demonstrated between the majority of quantitative results, with a notably excellent correlation between the current EU reference method using pre-column oxidation LC-FLD and LC-MS/MS. The LFA showed great potential for qualitative determination of PSP toxins, although the findings of high numbers of false-positive results and two false negatives highlighted that some caution is still needed when interpreting results. This study demonstrated that effective replacement methods are available for countries that no longer wish to use the MBA, but highlighted the importance of comparing toxin data from the replacement method using local shellfish species of concern before implementing new methods in official control testing programs

    Bioactive compounds and antibacterial activities in crystallized honey liquefied with ultrasound

    The effect of ultrasound on the crystal size, phenols, flavonoids, Maillard products and antibacterial activity of crystallized honeys was studied. Three multifloral honeys (M), one monofloral (MO) and one honeydew (HD) honey were used. Ultrasound was performed at 42 kHz for different times (0, 5, 10 and 15 min). The antibacterial activities were tested against Salmonella typhimurium, Bacillus subtilis, Pseudomonas aeruginosa, Listeria mono cytogenes, Staphylococcus aureus and Escherichia coli. In all honeys, the parameters analyzed had significant dif ferences ((P < 0.05)). After 15 min of ultrasound the HD had increments of 44 mg of gallic acid/100 g of honey in phenols, and some M showed increase in flavonoids (5.64 mg of quercitin /100 g of honey) and improvement in inhibition against Salmonella typhimurium was 13.1%. In some honeys the correlation between phenols or fla vonoids and antibacterial activity were significant ((P < 0.05)). No correlation was found between Maillard products and antibacterial activity.     A malignant hyperthermia–inducing mutation in RYR1 (R163C): alterations in Ca2+ entry, release, and retrograde signaling to the DHPR

    Bidirectional signaling between the sarcolemmal L-type Ca2+ channel (1,4-dihydropyridine receptor [DHPR]) and the sarcoplasmic reticulum (SR) Ca2+ release channel (type 1 ryanodine receptor [RYR1]) of skeletal muscle is essential for excitation–contraction coupling (ECC) and is a well-understood prototype of conformational coupling. Mutations in either channel alter coupling fidelity and with an added pharmacologic stimulus or stress can trigger malignant hyperthermia (MH). In this study, we measured the response of wild-type (WT), heterozygous (Het), or homozygous (Hom) RYR1-R163C knock-in mouse myotubes to maintained K+ depolarization. The new findings are: (a) For all three genotypes, Ca2+ transients decay during prolonged depolarization, and this decay is not a consequence of SR depletion or RYR1 inactivation. (b) The R163C mutation retards the decay rate with a rank order WT > Het > Hom. (c) The removal of external Ca2+ or the addition of Ca2+ entry blockers (nifedipine, SKF96365, and Ni2+) enhanced the rate of decay in all genotypes. (d) When Ca2+ entry is blocked, the decay rates are slower for Hom and Het than WT, indicating that the rate of inactivation of ECC is affected by the R163C mutation and is genotype dependent (WT > Het > Hom). (e) Reduced ECC inactivation in Het and Hom myotubes was shown directly using two identical K+ depolarizations separated by varying time intervals. These data suggest that conformational changes induced by the R163C MH mutation alter the retrograde signal that is sent from RYR1 to the DHPR, delaying the inactivation of the DHPR voltage sensor

    TRY plant trait database – enhanced coverage and open access

    Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait‐based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for ‘plant growth form’. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait–environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives
