    Back to the Future: Conserving Functional and Phylogenetic Diversity in the Amphibian-Climate Refuges

    Climate refuges have been used by several species over historical climate change. Ectothermic species often display good models for climate change studies because they are highly sensitive to temperature. Analysis of species loss with ecosystem and evolutionary values helps to understand environmental processes and climate change consequences. We determined the functional and phylogenetic diversity of amphibians in the Atlantic Forest hotspot, using multiple models representing present and future conditions. Through a novel approach, we predict species’ threat status by 2080, following the IUCN’s criterion B1. Our results estimate a drastic reduction in species richness, ecosystem functioning and evolutionary history at low latitudes and altitudes. We show that species will tend to disperse to the areas with milder temperatures (i.e., high latitudes/altitudes). Some of these areas are the same climate refuges that have been suggested for the Late Pleistocene. We highlight that 60% of amphibians can become threatened under predicted-future conditions. This work advances the knowledge on climate refuges for amphibian ecology and evolution, supporting complementary tools for conservation strategies

    Are you drowned in microplastic pollution? A brief insight on the current knowledge for early career researchers developing novel remediation strategies

    Microplastics (MPs) composed of different polymers with various shapes, within a vast granulometric distribution (1 μm - 5 mm) and with a wide variety of physicochemical surface and bulk characteristics spiral around the globe, with different atmospheric, oceanic, cryospheric, and terrestrial residence times, while interacting with other pollutants and biota. The challenges of microplastic pollution are related to the complex relationships between the microplastic generation mechanisms (physical, chemical, and biological), their physicochemical properties, their interactions with other pollutants and microorganisms, the changes in their properties with aging, and their small sizes that facilitate their diffusion and transportation between the air, water, land, and biota, thereby promoting their ubiquity. Early career researchers (ERCs) constitute an essential part of the scientific community committed to overcoming the challenges of microplastic pollution with their new ideas and innovative scientific perspectives for the development of remediation technologies. However, because of the enormous amount of scientific information available, it may be difficult for ERCs to determine the complexity of this environmental issue. This mini-review aims to provide a quick and updated overview of the essential insights of microplastic pollution to ERCs to help them acquire the background needed to develop highly innovative physical, chemical, and biological remediation technologies, as well as valorization proposals and environmental education and awareness campaigns. Moreover, the recommendations for the development of holistic microplastic pollution remediation strategies presented here can help ERCs propose technologies considering the environmental, social, and practical dimensions of microplastic pollution while fulfilling the current government policies to manage this plastic waste

    Concentraciones y movilidad relativa de isoformas de titina después de tres distintos entrenamientos de flexibilidad

    La flexibilidad muscular es un importante componente de aptitud física y está relacionada con el desempeño de actividades diarias y deportivas. Es determinada por factores intrínsecos y extrínsecos. En el caso de los intrínsecos, se destaca la concentración de titina en los músculos esqueléticos. El objetivo de este estudio fue comparar las concentra- ciones y la movilidad relativa de las isoformas T1 y T2 de titina después de 16 semanas de entrenamiento con diferentes ejercicios de flexibilidad. Para ello, 51 individuos entrenados, del sexo masculino con edades entre los 19 y 26 años, y con por lo menos 6 meses de entrenamiento físico, fueron divididos en tres grupos: elongación, flexibilización activa y flexibilización pasiva. La elongación no implicó ninguna modificación que pueda desvirtuar los porcentuales y la movilidad de las isoformas de titina. Los grupos que realizaron flexibilización presentaron disminución porcentual de la isoforma T1 y aumento porcentual de la isoforma T2, además del aumento de la movilidad relativa de estas dos isoformas de titina. La flexibilización pasiva implicó en mayores alteraciones en los porcentuales de las isoformas T1 (% = -11.30% para p < 0.05) y T2 (% = 21.45% para p < 0.05). La flexibilización activa promovió mayor aumento de movilidad relativa de las isoformas T1 (% = 26.53% para p < 0.05) y T2 (% = 10.96% para p < 0.05) de titina

    Genetic regulation of pituitary gland development in human and mouse

    Normal hypothalamopituitary development is closely related to that of the forebrain and is dependent upon a complex genetic cascade of transcription factors and signaling molecules that may be either intrinsic or extrinsic to the developing Rathke’s pouch. These factors dictate organ commitment, cell differentiation, and cell proliferation within the anterior pituitary. Abnormalities in these processes are associated with congenital hypopituitarism, a spectrum of disorders that includes syndromic disorders such as septo-optic dysplasia, combined pituitary hormone deficiencies, and isolated hormone deficiencies, of which the commonest is GH deficiency. The highly variable clinical phenotypes can now in part be explained due to research performed over the last 20 yr, based mainly on naturally occurring and transgenic animal models. Mutations in genes encoding both signaling molecules and transcription factors have been implicated in the etiology of hypopituitarism, with or without other syndromic features, in mice and humans. To date, mutations in known genes account for a small proportion of cases of hypopituitarism in humans. However, these mutations have led to a greater understanding of the genetic interactions that lead to normal pituitary development. This review attempts to describe the complexity of pituitary development in the rodent, with particular emphasis on those factors that, when mutated, are associated with hypopituitarism in humans

    AI is a viable alternative to high throughput screening: a 318-target study

    : High throughput screening (HTS) is routinely used to identify bioactive small molecules. This requires physical compounds, which limits coverage of accessible chemical space. Computational approaches combined with vast on-demand chemical libraries can access far greater chemical space, provided that the predictive accuracy is sufficient to identify useful molecules. Through the largest and most diverse virtual HTS campaign reported to date, comprising 318 individual projects, we demonstrate that our AtomNet® convolutional neural network successfully finds novel hits across every major therapeutic area and protein class. We address historical limitations of computational screening by demonstrating success for target proteins without known binders, high-quality X-ray crystal structures, or manual cherry-picking of compounds. We show that the molecules selected by the AtomNet® model are novel drug-like scaffolds rather than minor modifications to known bioactive compounds. Our empirical results suggest that computational methods can substantially replace HTS as the first step of small-molecule drug discovery

    Search for new physics in top quark production with additional leptons in proton-proton collisions at root s=13 TeV using effective field theory

    Events containing one or more top quarks produced with additional prompt leptons are used to search for new physics within the framework of an effective field theory (EFT). The data correspond to an integrated luminosity of 41.5 fb(-1) of proton-proton collisions at a center-of-mass energy of 13 TeV at the LHC, collected by the CMS experiment in 2017. The selected events are required to have either two leptons with the same charge or more than two leptons; jets, including identified bottom quark jets, are also required, and the selected events are divided into categories based on the multiplicities of these objects. Sixteen dimension-six operators that can affect processes involving top quarks produced with additional charged leptons are considered in this analysis. Constructed to target EFT effects directly, the analysis applies a novel approach in which the observed yields are parameterized in terms of the Wilson coefficients (WCs) of the EFT operators. A simultaneous fit of the 16 WCs to the data is performed and two standard deviation confidence intervals for the WCs are extracted; the standard model expectations for the WC values are within these intervals for all of the WCs probed.Peer reviewe

    Angular analysis of the decay B+ -> K+mu(+)mu(-) in proton-proton collisions at root s=8 TeV

    The angular distribution of the flavor-changing neutral current decay B+ -> K+mu(+)mu(-) is studied in proton-proton collisions at a center-of-mass energy of 8 TeV. The analysis is based on data collected with the CMS detector at the LHC, corresponding to an integrated luminosity of 20.5 fb(-1). The forward-backward asymmetry A(FB) of the dimuon system and the contribution F-H from the pseudoscalar, scalar, and tensor amplitudes to the decay width are measured as a function of the dimuon mass squared. The measurements are consistent with the standard model expectations.Peer reviewe
