    Recent results of H-mode studies on asdex

    In a comparative study of various confinement regimes the H-mode demonstrated the best performance. Confinement enhancement factors (above ITER 89-P L-mode scaling) in the range of 1.6 fH 2.8 have been achieved with values depending on the divertor configuration, the wall condition, ELM behaviour and the plasma ion spicies. Long-pulse H-phases, withELMs, of up to 3.5s with constant confinement time, recycling and impurity characteristics are archived. H* -mode operation is possible without a loss of current scaling at qa values as low as 2.2. The B-limit is the same with and without ELMs. Murakami parameters are similar in H-and L-modes

    Machine-learning prediction of tumor antigen immunogenicity in the selection of therapeutic epitopes

    Current tumor neoantigen calling algorithms primarily rely on epitope/major histocompatibility complex (MHC) binding affinity predictions to rank and select for potential epitope targets. These algorithms do not predict for epitope immunogenicity using approaches modeled from tumor-specific antigen data. Here, we describe peptide-intrinsic biochemical features associated with neoantigen and minor histocompatibility mismatch antigen immunogenicity and present a gradient boosting algorithm for predicting tumor antigen immunogenicity. This algorithm was validated in two murine tumor models and demonstrated the capacity to select for therapeutically active antigens. Immune correlates of neoantigen immunogenicity were studied in a pan-cancer data set from The Cancer Genome Atlas and demonstrated an association between expression of immunogenic neoantigens and immunity in colon and lung adenocarcinomas. Lastly, we present evidence for expression of an out-of-frame neoantigen that was capable of driving antitumor cytotoxic T-cell responses. With the growing clinical importance of tumor vaccine therapies, our approach may allow for better selection of therapeutically relevant tumor-specific antigens, including nonclas-sic out-of-frame antigens capable of driving antitumor immunity

    Current status of turbulent dynamo theory: From large-scale to small-scale dynamos

    Several recent advances in turbulent dynamo theory are reviewed. High resolution simulations of small-scale and large-scale dynamo action in periodic domains are compared with each other and contrasted with similar results at low magnetic Prandtl numbers. It is argued that all the different cases show similarities at intermediate length scales. On the other hand, in the presence of helicity of the turbulence, power develops on large scales, which is not present in non-helical small-scale turbulent dynamos. At small length scales, differences occur in connection with the dissipation cutoff scales associated with the respective value of the magnetic Prandtl number. These differences are found to be independent of whether or not there is large-scale dynamo action. However, large-scale dynamos in homogeneous systems are shown to suffer from resistive slow-down even at intermediate length scales. The results from simulations are connected to mean field theory and its applications. Recent work on helicity fluxes to alleviate large-scale dynamo quenching, shear dynamos, nonlocal effects and magnetic structures from strong density stratification are highlighted.     CVD diamond coated silicon nitride self-mated systems : tribological behaviour under high loads

    Friction and wear behaviour of self-mated chemical vapour deposited (CVD) diamond films coating silicon nitride ceramics (Si3N4) were investigated in ambient atmosphere. The tribological tests were conducted in a reciprocal motion ball-on-flat type tribometer under applied normal loads up to 80 N (~10 GPa). Several characterisation techniques - including scanning electron microscopy (SEM), atomic force microscopy (AFM) and micro-Raman studies - were used in order to assess the quality, stress state and wear resistance of the coatings. In addition, a novel method is presented to estimate the wear coefficient of the diamond coated flat specimens from AFM and optical microscopy (OM) observations of the wear tracks

    Standards for plant synthetic biology: A common syntax for exchange of DNA parts

Inventors in the field of mechanical and electronic engineering can access multitudes of components and, thanks to standardization, parts from different manufacturers can be used in combination with each other. The introduction of BioBrick standards for the assembly of characterized DNA sequences was a landmark in microbial engineering, shaping the field of synthetic biology. Here, we describe a standard for Type IIS restriction endonuclease-mediated assembly, defining a common syntax of 12 fusion sites to enable the facile assembly of eukaryotic transcriptional units. This standard has been developed and agreed by representatives and leaders of the international plant science and synthetic biology communities, including inventors, developers and adopters of Type IIS cloning methods. Our vision is of an extensive catalogue of standardized, characterized DNA parts that will accelerate plant bioengineering

    Contribution of machining to the fatigue behaviour of metal matrix composites (MMCs) of varying reinforcement size

    The high cycle constant stress amplitude fatigue performance of metal matrix composite (MMC) components machined by a milling process was investigated in this study as a function of machining speed, feed rate and reinforcement particle size. The presence of reinforcement and particle size were found to be the most influential factors that affected the fatigue life. In contrast to this, the effect of feed and speed on tool-particle interaction, strain hardening and heat generation during milling of MMCs were balanced in such a way that the contributions of feed and speed on fatigue life were negligible. The interactions of different parameters contributed significantly to the fatigue life which indicated that the modelling of fatigue life based on these three parameters was relatively complex. The fatigue life of the machined MMC samples increased with decreasing particle size and increasing feed. However, the fatigue life was not influenced by speed variation. The presence of smaller or no particles induced a complete separation of failed samples, in contrast to that of specimens containing larger reinforcing particles where crack growth was arrested or deflected by the reinforcing particles

    Flavor Decomposition of the Polarized Quark Distributions in the Nucleon from Inclusive and Semi-inclusive Deep-inelastic Scattering

    Spin asymmetries of semi-inclusive cross sections for the production of positively and negatively charged hadrons have been measured in deep-inelastic scattering of polarized positrons on polarized hydrogen and 3He targets, in the kinematic range 0.023<x<0.6 and 1 GeV^2<Q^2<10 GeV^2. Polarized quark distributions are extracted as a function of x for up $(u+u_bar) and down (d+d_bar) flavors. The up quark polarization is positive and the down quark polarization is negative in the measured range. The polarization of the sea is compatible with zero. The first moments of the polarized quark distributions are presented. The isospin non-singlet combination Delta_q_3 is consistent with the prediction based on the Bjorken sum rule.     Time-integrated luminosity recorded by the BABAR detector at the PEP-II e+e- collider

    This article is the Preprint version of the final published artcile which can be accessed at the link below.We describe a measurement of the time-integrated luminosity of the data collected by the BABAR experiment at the PEP-II asymmetric-energy e+e- collider at the ϒ(4S), ϒ(3S), and ϒ(2S) resonances and in a continuum region below each resonance. We measure the time-integrated luminosity by counting e+e-→e+e- and (for the ϒ(4S) only) e+e-→μ+μ- candidate events, allowing additional photons in the final state. We use data-corrected simulation to determine the cross-sections and reconstruction efficiencies for these processes, as well as the major backgrounds. Due to the large cross-sections of e+e-→e+e- and e+e-→μ+μ-, the statistical uncertainties of the measurement are substantially smaller than the systematic uncertainties. The dominant systematic uncertainties are due to observed differences between data and simulation, as well as uncertainties on the cross-sections. For data collected on the ϒ(3S) and ϒ(2S) resonances, an additional uncertainty arises due to ϒ→e+e-X background. For data collected off the ϒ resonances, we estimate an additional uncertainty due to time dependent efficiency variations, which can affect the short off-resonance runs.
