Las proteínas del plasma seminal incrementan la viabilidad espermática post-descongelación del semen de toros Sanmartinero
RESUMEN
Objetivo. El objetivo de este trabajo fue evaluar el efecto de la adición de proteínas del plasma seminal sobre el porcentaje de espermatozoides bovinos viables post-descongelación. Materiales y métodos. Los espermatozoides se congelaron usando dos medios (citrato-fructosa-yema y Bioxcell®) y la obtención de proteínas de plasma seminal de bajo peso molecular se realizó por medio de cromatografía líquida de baja presión. Las proteínas de interés eluyeron en las fracciones 21-25 y se sometieron a electroforésis en una y dos dimensiones. Los espermatozoides se incubaron a 37°C durante una hora, con 0.5, 1.0, 1.5 y 2.0 mg de la fracción 21-25. Se incluyeron dos tratamientos adicionales: uno con proteínas totales del plasma seminal y otro sin proteína. Resultados. La electroforésis bidimensional de las fracciones confirmó la presencia de siete puntos de proteína de bajo peso molecular (14-16 kDa y punto Isoeléctrico de 5.0 - 5.5). La adición de estas proteínas aumentó 20% (p<0.05), el porcentaje de espermatozoides viables post-descongelación en muestras congeladas en medio citrato-fructosa-yema (con dosis de 1 ó 1.5 mg de proteína/106 espermatozoides), y 25% (p<0.05) en muestras congeladas en medio Bioxcell® (con dosis de 0.5 mg de proteína/106 espermatozoides). Conclusiones. Los resultados de esta investigación sugieren el posible uso de proteínas de bajo peso molecular del plasma seminal, para disminuir el efecto deletéreo de la criopreservación en los espermatozoide
Antimicrobial resistance among migrants in Europe: a systematic review and meta-analysis
BACKGROUND: Rates of antimicrobial resistance (AMR) are rising globally and there is concern that increased migration is contributing to the burden of antibiotic resistance in Europe. However, the effect of migration on the burden of AMR in Europe has not yet been comprehensively examined. Therefore, we did a systematic review and meta-analysis to identify and synthesise data for AMR carriage or infection in migrants to Europe to examine differences in patterns of AMR across migrant groups and in different settings. METHODS: For this systematic review and meta-analysis, we searched MEDLINE, Embase, PubMed, and Scopus with no language restrictions from Jan 1, 2000, to Jan 18, 2017, for primary data from observational studies reporting antibacterial resistance in common bacterial pathogens among migrants to 21 European Union-15 and European Economic Area countries. To be eligible for inclusion, studies had to report data on carriage or infection with laboratory-confirmed antibiotic-resistant organisms in migrant populations. We extracted data from eligible studies and assessed quality using piloted, standardised forms. We did not examine drug resistance in tuberculosis and excluded articles solely reporting on this parameter. We also excluded articles in which migrant status was determined by ethnicity, country of birth of participants' parents, or was not defined, and articles in which data were not disaggregated by migrant status. Outcomes were carriage of or infection with antibiotic-resistant organisms. We used random-effects models to calculate the pooled prevalence of each outcome. The study protocol is registered with PROSPERO, number CRD42016043681. FINDINGS: We identified 2274 articles, of which 23 observational studies reporting on antibiotic resistance in 2319 migrants were included. The pooled prevalence of any AMR carriage or AMR infection in migrants was 25·4% (95% CI 19·1-31·8; I2 =98%), including meticillin-resistant Staphylococcus aureus (7·8%, 4·8-10·7; I2 =92%) and antibiotic-resistant Gram-negative bacteria (27·2%, 17·6-36·8; I2 =94%). The pooled prevalence of any AMR carriage or infection was higher in refugees and asylum seekers (33·0%, 18·3-47·6; I2 =98%) than in other migrant groups (6·6%, 1·8-11·3; I2 =92%). The pooled prevalence of antibiotic-resistant organisms was slightly higher in high-migrant community settings (33·1%, 11·1-55·1; I2 =96%) than in migrants in hospitals (24·3%, 16·1-32·6; I2 =98%). We did not find evidence of high rates of transmission of AMR from migrant to host populations. INTERPRETATION: Migrants are exposed to conditions favouring the emergence of drug resistance during transit and in host countries in Europe. Increased antibiotic resistance among refugees and asylum seekers and in high-migrant community settings (such as refugee camps and detention facilities) highlights the need for improved living conditions, access to health care, and initiatives to facilitate detection of and appropriate high-quality treatment for antibiotic-resistant infections during transit and in host countries. Protocols for the prevention and control of infection and for antibiotic surveillance need to be integrated in all aspects of health care, which should be accessible for all migrant groups, and should target determinants of AMR before, during, and after migration. DNA multigene characterization of Fasciola hepatica and Lymnaea neotropica and its fascioliasis transmission capacity in Uruguay, with historical correlation, human report review and infection risk analysis
Fascioliasis is a highly pathogenic zoonotic disease emerging in recent decades, in part due to the effects of climate and global changes. South America is the continent presenting more numerous human fascioliasis endemic areas and the highest Fasciola hepatica infection prevalences and intensities known in humans. These serious public health scenarios appear mainly linked to altitude areas in Andean countries, whereas lowland areas of non-Andean countries, such as Uruguay, only show sporadic human cases or outbreaks. To understand this difference, we characterized F. hepatica from cattle and horses and lymnaeids of Uruguay by sequencing of ribosomal DNA ITS-2 and ITS-1 spacers and mitochondrial DNA cox1, nad1 and 16S genes. Results indicate that vectors belong to Lymnaea neotropica instead of to Lymnaea viator, as always reported from Uruguay. Our correlation of fasciolid and lymnaeid haplotypes with historical data on the introduction and spread of livestock species into Uruguay allow to understand the molecular diversity detected. We study the life cycle and transmission features of F. hepatica by L. neotropica of Uruguay under standardized experimental conditions to enable a comparison with the transmission capacity of F. hepatica by Galba truncatula at very high altitude in Bolivia. Results demonstrate that although L. neotropica is a highly efficient vector in the lowlands, its transmission capacity is markedly lower than that of G. truncatula in the highlands. On this baseline, we review the human fascioliasis cases reported in Uruguay and analyze the present and future risk of human infection in front of future climate change estimations
Surgical site infection after gastrointestinal surgery in high-income, middle-income, and low-income countries: a prospective, international, multicentre cohort study
Background: Surgical site infection (SSI) is one of the most common infections associated with health care, but its importance as a global health priority is not fully understood. We quantified the burden of SSI after gastrointestinal surgery in countries in all parts of the world.
Methods: This international, prospective, multicentre cohort study included consecutive patients undergoing elective or emergency gastrointestinal resection within 2-week time periods at any health-care facility in any country. Countries with participating centres were stratified into high-income, middle-income, and low-income groups according to the UN's Human Development Index (HDI). Data variables from the GlobalSurg 1 study and other studies that have been found to affect the likelihood of SSI were entered into risk adjustment models. The primary outcome measure was the 30-day SSI incidence (defined by US Centers for Disease Control and Prevention criteria for superficial and deep incisional SSI). Relationships with explanatory variables were examined using Bayesian multilevel logistic regression models. This trial is registered with ClinicalTrials.gov, number NCT02662231.
Findings: Between Jan 4, 2016, and July 31, 2016, 13 265 records were submitted for analysis. 12 539 patients from 343 hospitals in 66 countries were included. 7339 (58·5%) patient were from high-HDI countries (193 hospitals in 30 countries), 3918 (31·2%) patients were from middle-HDI countries (82 hospitals in 18 countries), and 1282 (10·2%) patients were from low-HDI countries (68 hospitals in 18 countries). In total, 1538 (12·3%) patients had SSI within 30 days of surgery. The incidence of SSI varied between countries with high (691 [9·4%] of 7339 patients), middle (549 [14·0%] of 3918 patients), and low (298 [23·2%] of 1282) HDI (p < 0·001). The highest SSI incidence in each HDI group was after dirty surgery (102 [17·8%] of 574 patients in high-HDI countries; 74 [31·4%] of 236 patients in middle-HDI countries; 72 [39·8%] of 181 patients in low-HDI countries). Following risk factor adjustment, patients in low-HDI countries were at greatest risk of SSI (adjusted odds ratio 1·60, 95% credible interval 1·05–2·37; p=0·030). 132 (21·6%) of 610 patients with an SSI and a microbiology culture result had an infection that was resistant to the prophylactic antibiotic used. Resistant infections were detected in 49 (16·6%) of 295 patients in high-HDI countries, in 37 (19·8%) of 187 patients in middle-HDI countries, and in 46 (35·9%) of 128 patients in low-HDI countries (p < 0·001).
Interpretation: Countries with a low HDI carry a disproportionately greater burden of SSI than countries with a middle or high HDI and might have higher rates of antibiotic resistance. In view of WHO recommendations on SSI prevention that highlight the absence of high-quality interventional research, urgent, pragmatic, randomised trials based in LMICs are needed to assess measures aiming to reduce this preventable complication
Global disparities in surgeons’ workloads, academic engagement and rest periods: the on-calL shIft fOr geNEral SurgeonS (LIONESS) study
: The workload of general surgeons is multifaceted, encompassing not only surgical procedures but also a myriad of other responsibilities. From April to May 2023, we conducted a CHERRIES-compliant internet-based survey analyzing clinical practice, academic engagement, and post-on-call rest. The questionnaire featured six sections with 35 questions. Statistical analysis used Chi-square tests, ANOVA, and logistic regression (SPSS® v. 28). The survey received a total of 1.046 responses (65.4%). Over 78.0% of responders came from Europe, 65.1% came from a general surgery unit; 92.8% of European and 87.5% of North American respondents were involved in research, compared to 71.7% in Africa. Europe led in publishing research studies (6.6 ± 8.6 yearly). Teaching involvement was high in North America (100%) and Africa (91.7%). Surgeons reported an average of 6.7 ± 4.9 on-call shifts per month, with European and North American surgeons experiencing 6.5 ± 4.9 and 7.8 ± 4.1 on-calls monthly, respectively. African surgeons had the highest on-call frequency (8.7 ± 6.1). Post-on-call, only 35.1% of respondents received a day off. Europeans were most likely (40%) to have a day off, while African surgeons were least likely (6.7%). On the adjusted multivariable analysis HDI (Human Development Index) (aOR 1.993) hospital capacity > 400 beds (aOR 2.423), working in a specialty surgery unit (aOR 2.087), and making the on-call in-house (aOR 5.446), significantly predicted the likelihood of having a day off after an on-call shift. Our study revealed critical insights into the disparities in workload, access to research, and professional opportunities for surgeons across different continents, underscored by the HDI
Taking the pulse of Earth's tropical forests using networks of highly distributed plots
Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science. La investigación participativa en Bienestar y Comportamiento animal en el trópico de América: oportunidades para nuevo conocimiento aplicado
En la presente conferencia se hace una reflexión sobre la importancia de reconocer algunas particularidades de la región tropical de América, que invitan a repensar una agenda adecuada para el conocimiento y formación en los temas del Bienestar y Comportamiento Animal (BA/CA). La creatividad en el trabajo interdisciplinario y el respeto a formas más participativas de hacer la investigación, son dos bases poderosas que pueden lograr nuevos productos de conocimiento con potencial de aplicación inmediata. La investigación participativa emplea procedimientos del método científico con respeto y valoración de fuentes de conocimiento y los sujetos de investigación diferentes al investigador. La combinación respetuosa a través del Diálogo de Saberes sistematizado permite sinergias que pueden superar en resultados los métodos convencionales de investigación, con la ventaja de ser rápidamente apropiables por las comunidades y sectores productivos. A pesar de que el Bienestar y Comportamiento Animal (BA/CA), son asuntos de creciente interés entre los consumidores organizados del mundo desarrollado y algunas instituciones internacionales como la FAO lo consideren importante dentro de los criterios de calidad de los productos para consumo humano, así como de la sostenibilidad de los sistemas productivos, el estado de avance del conocimiento, divulgación y aplicación en la producción animal en América tropical es bajo. En América tropical tenemos la presencia simultánea de los tres grandes sistemas de producción animal reconocidos en el mundo: el industrial o intensivo (avicultura, porcicultura y en menor escala otras especies); el de pastoreo con sus grandes extremos (extensivos, semi-intensivos y rotacionales); y los sistemas mixtos que combinan agricultura, manejo de bosques y otros ecosistemas naturales con actividades ganaderas. En el primer grupo de sistemas, el que más energía fósil y tierras fértiles emplea (cultivos de cereales y soya), las condiciones de BA/CA están afectadas por el confinamiento y privación de las libertades individuales, la manipulación genética extrema, la contaminación del aire y las aguas, así como el uso indiscriminado de sustancias de síntesis industrial, como los promotores de crecimiento manejadas como uso cotidiano y preventivo de enfermedades. En el segundo grupo, es bien conocida la enorme ocupación de territorio (la más alta del continente), sus bajos índices productivos, la falta de garantías sobre la satisfacción de la sed y la buena nutrición, alta exposición a la radiación solar y estrés climático, además de algunas prácticas violentas y dolorosas. En la región existen tradiciones, formas culturales de relación con los animales y profundas raíces de conocimiento empírico que en el caso de varios ecosistemas tienen claves estratégicas para el manejo sustentable de enormes territorios, así como el de animales en sistemas de producción adaptados con el uso de recursos tropicales. Es necesario reconocer en los animales productivos, un ser vivo, con sensibilidad y fragilidad como individuo y grupo, condiciones que no son obstáculo para una adecuada productividad, por el contrario, el bienestar es un factor determinante de la producción que además genera arraigo, orgullo y felicidad en los productores y garantía de salud y buena nutrición en los consumidores. Buscar un camino diferente y auténtico al diseñar agendas apropiadas de investigación, formación y divulgación conducirá a procesos virtuosos para fortalecer la cultura rural con nuevos conocimientos emergidos en procesos de interdisciplinariedad con participación de muchos actores sociales y lo más importante, con aplicación inmediata como requiere la ciencia para el desarrollo sustentable
Proteínas del plasma seminal y calidad espermática de toros Romosinuano y Costeño con Cuernos
The aim of this study was to determine by SDS PAGE the protein profile of the seminal plasma of Romosinuano (ROMO) and Costeño con Cuernos (CCC) bulls and their relationship with sperm quality parameters in fresh and thawed semen. Three ejaculates per bull were collected by electro-ejaculate from 10 ROMO and 10 CCC sires. Viability (VF), mass motility (MMF), individual rapid progressive motility (MIRPF) and sperm concentration (CE) were determined in fresh semen. Seminal plasma (PS) was obtained by double centrifugation. Individual motility of rapid progressives (MIRPPC), progressivity (PRPC), rapid progressives (RPPC) and medium progressive (MPPC) were determined in thawed semen, in both cases using the CASA computerized analysis system. Additionally, viability (VPC) and membrane integrity were determined by the hypoosmotic test. Proteins were separated by polyacrylamide gel electrophoresis (SDS PAGE). The data was analyzed by Student’s T test and the presence of proteins by Chi square. The relationship between proteins and seminal quality parameters using Pearson’s correlation test and principal components of forward regression. In fresh semen, differences (p<0.05) were found between the two breeds for CE and MIRPF, and in thawed semen for VPC. Ten protein bands were evidenced in ROMO bulls and 7 in CCC bulls. The 13, 83 and 121 kDa bands were positively related and the 15 kDa negatively with VF from ROMO bulls. Protein bands 21, 25, 102 and 174 were associated with both MF and CE in ROMO bulls. In conclusion, the electrophoretic profiles of seminal plasma proteins from Romosinuano and Costeño con Cuernos bulls were established. The relationships between the bands detected with the sperm quality could partly explain the fertilizing capacity of the sperm of these breeds.El objetivo del estudio fue determinar mediante SDS PAGE el perfil de proteínas del plasma seminal de toros criollos Romosinuano (ROMO) y Costeño con Cuernos (CCC) y su relación con los parámetros de calidad espermática en semen fresco y descongelado. Se colectaron tres eyaculados por toro mediante electro-eyaculación de 10 reproductores ROMO y 10 CCC. En semen fresco se determinó viabilidad (VF), motilidad masal (MMF), motilidad individual rápida progresiva (MIRPF) y concentración espermática (CE). El plasma seminal (PS) se obtuvo mediante doble centrifugación. En el semen descongelado se determinó motilidad individual de rápidos progresivos (MIRPPC), progresividad (PRPC), rápidos progresivos (RPPC) y medios progresivos (MPPC), en ambos casos usando el sistema de análisis computarizado CASA. Adicionalmente se determinó la viabilidad (VPC), e integridad de la membrana mediante el test hipoosmótico. Las proteínas se separaron por electroforesis en geles de poliacrilamida (SDS PAGE). Los datos se analizaron mediante prueba de T de student y la presencia de proteínas por Chi cuadrado. La relación entre las proteínas y los parámetros de calidad seminal mediante la prueba de correlación de Pearson y componentes principales de regresión hacia adelante. En semen fresco se evidenciaron diferencias (p<0.05) entre las dos razas para CE y MIRPF, y en semen congelado-descongelado para VPC. Se evidenciaron 10 bandas de proteína en toros ROMO y 7 en toros CCC. Las bandas de 13, 83 y 121 kDa se relacionaron positivamente y la de 15 kDa negativamente con VF de toros ROMO. Las bandas de proteínas de 21, 25, 102 y 174 se relacionaron tanto con MF y CE en toros ROMO. En conclusión, se establecieron los perfiles electroforéticos de proteínas del plasma seminal de toros ROMO y CCC. Las relaciones entre las bandas detectadas con la calidad espermática podrían explicar en parte la capacidad fecundante del espermatozoide de estas razas
