    Boosting inhibition control process by knitting at school

    Across two experiments, the presented research explored the impact of a knitting bout on elementary school pupils’ inhibition abilities. They proposed an accurate measure of the pupils’ inhibition abilities through the use of a stop-signal paradigm. In order to take into account, the differentiation between cool and hot inhibitions abilities, the emotional content of the stimuli was manipulated across experiments. Neutral materials were used in Experiment 1 when emotionally charged materials were in Experiment 2. The findings of both experiments highlighted a beneficial impact of the knitting bout on children’s inhibition abilities. While the results of Experiment 1 showed an optimization of inhibition abilities for the knitting session group in comparison to the control group, Experiment 2 revealed a disappearance of the effect of the emotional content on these abilities as well. Proposals as to why EF could be sensitive to knitting practice are discussed

    International audienceThis paper presents an interactive web simulation (SimDiabolo), designed by education and computer education researchers. The activity, proposed to students from 15 to 17 years old, is about designing an experimental procedure to highlight alcoholic fermentation in biology. In order to help students in this complex task, fixed and adaptive scaffolds have been designed and implemented in the simulation. The anthropological theory of the didactic has been used for modeling errors and designing scaffolds. The simulation can also be connected to present software FORMID, which can modify the actual script implemented in SimDiabolo and offer different scripts with different scaffolds. Experimentations will take place this year in biology class in high school (France).Nous proposons dans cette communication le résultat de la co-construction entre didacticiens des sciences et informaticiens, d'une simulation web interactive (SimDiabolo) à destination d'élèves de 15-17 ans. Elle propose actuellement une activité de conception expérimentale sur le thème de la fermentation alcoolique. Pour aider les élèves dans cette tâche complexe nous avons intégré des étayages fixes et adaptatifs. Les différentes aides ont été conçues à partir d'une analyse didactique de l'erreur selon le cadre de la théorie anthropologique du didactique. Grâce à la connexion avec la suite logicielle FORMID, il est possible de scénariser différentes situations d'utilisation de SimDiabolo et notamment de proposer des niveaux d'étayage différenciés. Les différents scénarios seront testés in situ dans des classes de lycée en France cette année

    Calcium Inhibits Promotion by Hot Dog of 1,2-Dimethylhydrazine-Induced Mucin-Depleted Foci in Rat Colon

    Epidemiology suggests that processed meat is associated with colorectal cancer risk, but few experimental studies support this association. We have shown that a model of cured meat made in a pilot workshop promotes preneoplastic lesions, mucin-depleted foci (MDF) in the colon of rats. This study had two aims: to check if real store-bought processed meats also promote MDF, and to test if calcium carbonate, which suppresses heme-induced promotion, can suppress promotion by processed meat. A 14-day study was done to test the effect of nine purchased cured meats on fecal and urinary biomarkers associated with heme-induced carcinogenesis promotion. Fecal water from rats given hot dog or fermented raw dry sausage was particularly cytotoxic. These two cured meats were thus given to rats pretreated with 1,2-dimethylhydrazine, to evaluate their effect on colorectal carcinogenesis. After a 100-d feeding period, fecal apparent total N-nitroso compounds (ATNC) were assayed and colons were scored for MDF. Hot dog diet increased fecal ATNC and the number of MDF per colon compared with the no-meat control diet (3.0±1.7 vs. 1.2±1.4, P<0.05). In a third study, addition of calcium carbonate (150 µmol/g) to the hot dog diet decreased the number of MDF/colon and fecal ATNC compared with the hot dog diet without calcium carbonate (1.2 ± 1.1 vs. 2.3 ± 1.4, respectively, P<0.05). This is the first experimental evidence that a widely consumed processed meat promotes colon carcinogenesis in rats. It also shows that dietary prevention of this detrimental effect is possible

    A central role for heme iron in colon carcinogenesis associated with red meat intake

    Epidemiology shows that red and processed meat intake is associated with an increased risk of colorectal cancer. Heme iron, heterocyclic amines and endogenous N-nitroso compounds (NOC) are proposed to explain this effect, but their relative contribution is unknown. Our study aimed at determining, at nutritional doses, which is the main factor involved and proposing a mechanism of cancer promotion by red meat. The relative part of heme iron (1% in diet), heterocyclic amines (PhIP+MeIQx, 50+25 μg/kg in diet) and NOC (induced by NaNO2+NaNO3 0.17+0.23 g/l of drinking water) was determined by a factorial design and preneoplastic endpoints in chemically-induced rats and validated on tumors in Min mice. The molecular mechanisms (genotoxicity, cytotoxicity) were analyzed in vitro in normal and Apc- deficient cell lines and confirmed on colon mucosa. Heme iron increased the number of preneoplastic lesions but dietary heterocyclic amines and NOC had no effect on carcinogenesis in rats. Dietary hemoglobin increased tumor load in Min mice (control diet: 67±39 mm2; 2,5% hemoglobin diet: 114±47 mm2, p=0.004). In vitro, fecal water from rats given hemoglobin was rich in aldehydes and was cytotoxic to normal cells, but not to premalignant cells. The aldehydes 4-hydroxynonenal and 4-hydroxyhexenal were more toxic to normal versus mutated cells and were only genotoxic to normal cells. Genotoxicity was also observed in colon mucosa of mice given hemoglobin. These results highlight the role of heme iron in the promotion of colon cancer by red meat and suggest that heme iron could initiate carcinogenesis through lipid peroxidation

    Post-translational modifications and mass spectrometry detection

    In this review, we provide a comprehensive bibliographic overview of the role of mass spectrometry and the recent technical developments in the detection of post-translational modifications (PTMs). We briefly describe the principles of mass spectrometry for detecting PTMs and the protein and peptide enrichment strategies for PTM analysis, including phosphorylation, acetylation and oxidation. This review presents a bibliographic overview of the scientific achievements and the recent technical development in the detection of PTMs is provided. In order to ascertain the state of the art in mass spectrometry and proteomics methodologies for the study of PTMs, we analyzed all the PTM data introduced in the Universal Protein Resource (UniProt) and the literature published in the last three years. The evolution of curated data in UniProt for proteins annotated as being post-translationally modified is also analyzed. Additionally, we have undertaken a careful analysis of the research articles published in the years 2010 to 2012 reporting the detection of PTMs in biological samples by mass spectrometry. © 2013 Elsevier Inc

    On the role of 4-hydroxynonenal in health and disease

    AbstractPolyunsaturated fatty acids are susceptible to peroxidation and they yield various degradation products, including the main α,β-unsaturated hydroxyalkenal, 4-hydroxy-2,3-trans-nonenal (HNE) in oxidative stress. Due to its high reactivity, HNE interacts with various macromolecules of the cell, and this general toxicity clearly contributes to a wide variety of pathological conditions. In addition, growing evidence suggests a more specific function of HNE in electrophilic signaling as a second messenger of oxidative/electrophilic stress. It can induce antioxidant defense mechanisms to restrain its own production and to enhance the cellular protection against oxidative stress. Moreover, HNE-mediated signaling can largely influence the fate of the cell through modulating major cellular processes, such as autophagy, proliferation and apoptosis. This review focuses on the molecular mechanisms underlying the signaling and regulatory functions of HNE. The role of HNE in the pathophysiology of cancer, cardiovascular and neurodegenerative diseases is also discussed
