    The Service Oriented Business Process and Separation of Concerns - Modelling paradigms for Architectures and Business Processes

    This paper deals with the contemporary concern of business service-orientation. The paper suggests that service-oriented Business Information Systems Development (BISD) permits quality aspects, -“ilities” e.g.; maintainability and modifiability, of BISs and BISD. In addition, this paper departs from viewing a Business Information System (BIS) as services and suggests Separation of Concerns (SoC) as a prerequisite for achieving a well formed digital ecosystem. Besides knowledge transfer the purpose of this paper is to indicate the importance of architecture incorporating SoC when service-orienting a business with the contemporary Service Oriented Architecture (SOA). By realizing business logic service orientation in a nationwide research project our conclusion indicates that separate implementation of decision logic and process logic provides two different species of digital services implementing SoC. Consequently, responsible and eligible digital services become deliverables, service-orienting a part of a business representing vital quality aspects of ISD. The viability of separate digital services ensuring SoC facilitates the well-formed digital ecosystem i.e. the service oriented business process and business logic reusability and modifiability. The originality of this paper is based on a non-technical departure of SOA and on separation of process and decision logic as a mean to decreased child deaths due to preventable diseases

    Business Rules Friendly or not so Business Rules Friendly Business Concepts Modelling - Early Experiences from a Business Rules Project on a Digital Vaccination Recommendation Service

    WHO estimated in 2002 that 1.4 million children in the world would die from preventable diseases (World Health Organization, 2010). Each child is, or should be, immunized according to their national immunization schedule, which regulates i.e. vaccines and doses. Problems emerge when immigrant children should be transferred from one national vaccination schedule into another. This problem is quite evident in Sweden (Statistics Sweden, 2010). Vaccination practitioners have a hard time figuring out how to immunize an immigrant child. As a remedy for this situation, the VacSam project develops a Business Rules (BR) centric digital service through principles of (BR Approach), for providing unique vaccination recommendations. The service is designed using a commercial BRMS (BR Management Systems), which require a business concepts model. Thus, the paper presents early findings on how a business concepts model could be designed more or less BR-friendly. We have discovered, by designing, that a rigid, relational model approach is not BR-friendly. Instead, the concepts model should be de-signed to provide rule clarity and understandability. Thus, we draw the conclusion that a con-cepts model designed for persistent data storing, favouring a good quality relational model, is not rule friendly

    Mediator regulates non-coding RNA transcription at fission yeast centromeres

    BACKGROUND: In fission yeast, centromeric heterochromatin is necessary for the fidelity of chromosome segregation. Propagation of heterochromatin in dividing cells requires RNA interference (RNAi) and transcription of centromeric repeats by RNA polymerase II during the S phase of the cell cycle. RESULTS: We found that the Med8-Med18-Med20 submodule of the Mediator complex is required for the transcriptional regulation of native centromeric dh and dg repeats and for the silencing of reporter genes inserted in centromeric heterochromatin. Mutations in the Med8-Med18-Med20 submodule did not alter Mediator occupancy at centromeres; however, they led to an increased recruitment of RNA polymerase II to centromeres and reduced levels of centromeric H3K9 methylation accounting for the centromeric desilencing. Further, we observed that Med18 and Med20 were required for efficient processing of dh transcripts into siRNA. Consistent with defects in centromeric heterochromatin, cells lacking Med18 or Med20 displayed elevated rates of mitotic chromosome loss. CONCLUSIONS: Our data demonstrate a role for the Med8-Med18-Med20 Mediator submodule in the regulation of non-coding RNA transcription at Schizosaccharomyces pombe centromeres. In wild-type cells this submodule limits RNA polymerase II access to the heterochromatic DNA of the centromeres. Additionally, the submodule may act as an assembly platform for the RNAi machinery or regulate the activity of the RNAi pathway. Consequently, Med8-Med18-Med20 is required for silencing of centromeres and proper mitotic chromosome segregation

    International Salmonella Typhimurium DT104 Infections, 1992–2001

    The incidence of multidrug-resistant (MDR) Salmonella Typhimurium infections in humans, and in particular MDR definitive phage type 104 (DT104), has increased substantially in many countries in the last 2 decades, often associated with increased illness. To examine the magnitude of this problem, a survey was conducted among countries with available antimicrobial resistance or phage typing surveillance data. A total of 29, primarily industrialized, countries participated in the survey, which covered the years 1992–2001. Overall, the incidence of MDR S. Typhimurium and DT104 increased continuously during this period, although the problem affected primarily Europe and North America. The increase appeared to have peaked in the United Kingdom but not in other countries. Also, the incidence of quinolone-resistant S. Typhimurium was increasing. This survey implies that MDR S. Typhimurium constitutes an increasing public health problem in large parts of the world and emphasizes the importance of surveillance and control programs

    Background Dementia has an enormous impact on the lives of individuals and families, and on health and social services, and this will increase as the population ages. The needs of people with dementia and their carers for information and support are inadequately addressed at all key points in the illness trajectory. Methods The Unit is working specifically on an evaluation of the impact of the Mental Capacity Act 2005, and will develop practice guidance to enhance concordance with the Act. Phase One of the study has involved baseline interviews with practitioners across a wide range of services to establish knowledge and expectations of the Act, and to consider change processes when new policy and legislation are implemented. Findings Phase 1, involving baseline interviews with 115 practitioners, identified variable knowledge and understanding about the principles of the Act. Phase 2 is exploring everyday decision-making by people with memory problems and their carers

    Automated Solid-Phase Subcloning Based on Beads Brought into Proximity by Magnetic Force

    In the fields of proteomics, metabolic engineering and synthetic biology there is a need for high-throughput and reliable cloning methods to facilitate construction of expression vectors and genetic pathways. Here, we describe a new approach for solid-phase cloning in which both the vector and the gene are immobilized to separate paramagnetic beads and brought into proximity by magnetic force. Ligation events were directly evaluated using fluorescent-based microscopy and flow cytometry. The highest ligation efficiencies were obtained when gene- and vector-coated beads were brought into close contact by application of a magnet during the ligation step. An automated procedure was developed using a laboratory workstation to transfer genes into various expression vectors and more than 95% correct clones were obtained in a number of various applications. The method presented here is suitable for efficient subcloning in an automated manner to rapidly generate a large number of gene constructs in various vectors intended for high throughput applications

    Two conserved modules of Schizosaccharomyces pombe Mediator regulate distinct cellular pathways

    Mediator is an evolutionary conserved coregulator complex required for transcription of almost all RNA polymerase II-dependent genes. The Schizosaccharomyces pombe Mediator consists of two dissociable components—a core complex organized into a head and middle domain as well as the Cdk8 regulatory subcomplex. In this work we describe a functional characterization of the S. pombe Mediator. We report the identification of the S. pombe Med20 head subunit and the isolation of ts alleles of the core head subunit encoding med17+. Biochemical analysis of med8ts, med17ts, Δmed18, Δmed20 and Δmed27 alleles revealed a stepwise head domain molecular architecture. Phenotypical analysis of Cdk8 and head module alleles including expression profiling classified the Mediator mutant alleles into one of two groups. Cdk8 module mutants flocculate due to overexpression of adhesive cell-surface proteins. Head domain-associated mutants display a hyphal growth phenotype due to defective expression of factors required for cell separation regulated by transcription factor Ace2. Comparison with Saccharomyces cerevisiae Mediator expression data reveals that these functionally distinct modules are conserved between S. pombe and S. cerevisiae

    Search for new particles in events with energetic jets and large missing transverse momentum in proton-proton collisions at root s=13 TeV

    A search is presented for new particles produced at the LHC in proton-proton collisions at root s = 13 TeV, using events with energetic jets and large missing transverse momentum. The analysis is based on a data sample corresponding to an integrated luminosity of 101 fb(-1), collected in 2017-2018 with the CMS detector. Machine learning techniques are used to define separate categories for events with narrow jets from initial-state radiation and events with large-radius jets consistent with a hadronic decay of a W or Z boson. A statistical combination is made with an earlier search based on a data sample of 36 fb(-1), collected in 2016. No significant excess of events is observed with respect to the standard model background expectation determined from control samples in data. The results are interpreted in terms of limits on the branching fraction of an invisible decay of the Higgs boson, as well as constraints on simplified models of dark matter, on first-generation scalar leptoquarks decaying to quarks and neutrinos, and on models with large extra dimensions. Several of the new limits, specifically for spin-1 dark matter mediators, pseudoscalar mediators, colored mediators, and leptoquarks, are the most restrictive to date.Peer reviewe
