    Experimental Conversion of Liver to Pancreas

    AbstractBackground: The liver and the pancreas arise from adjacent regions of endoderm in embryonic development. Pdx1 is a key transcription factor that is essential for the development of the pancreas and is not expressed in the liver. The aim of this study was to determine whether a gene overexpression protocol based on Pdx1 would be able to cause conversion of liver to pancreas.Results: We show that a modified form of Pdx1, carrying the VP16 transcriptional activation domain, can cause conversion of liver to pancreas, both in vivo and in vitro. Transgenic Xenopus tadpoles carrying the construct TTR-Xlhbox8-VP16:Elas-GFP were prepared. Xlhbox8 is the Xenopus homolog of Pdx1, the TTR (transthyretin) promoter directs expression to the liver, and the GFP is under the control of an elastase promoter and provides a real-time visible marker of pancreatic differentiation. In the transgenic tadpoles, part or all of the liver is converted to pancreas, containing both exocrine and endocrine cells, while liver differentiation products are lost from the regions converted to pancreas. The timing of events is such that the liver is differentiating by the time Xlhbox8-VP16 is expressed, so we consider this a transdifferentiation event rather than a reprogramming of embryonic development. Furthermore, this same construct will bring about transdifferentiation of human hepatocytes in culture, with formation of both exocrine and endocrine cells.Conclusions: We consider that the conversion of liver to pancreas could be the basis of a new type of therapy for insulin-dependent diabetes. Although expression of the transgene is transient, once the ectopic pancreas is established, it persists thereafter

    Spin-orbit coupling in bulk GaAs

    We study the spin-orbit coupling in the whole Brillouin zone for GaAs using both the sp3s∗d5sp^3s^{\ast}d^5 and sp3s∗sp^3s^{\ast} nearest-neighbor tight-binding models. In the Γ\Gamma-valley, the spin splitting obtained is in good agreement with experimental data. We then further explicitly present the coefficients of the spin splitting in GaAs LL and XX valleys.     Amplification of evanescent waves in a lossy left-handed material slab

    We carry out finite-difference time-domain (FDTD) simulations, with a specially-designed boundary condition, on pure evanescent waves interacting with a lossy left-handed material (LHM) slab. Our results provide the first full-wave numerical evidence for the amplification of evanescent waves inside a LHM slab of finite absorption. The amplification is due to the interactions between the evanescent waves and the coupled surface polaritons at the two surfaces of the LHM slab and the physical process can be described by a simple model.Comment: 4 pages, 2 figure

    Solar wind pressure pulse‐driven magnetospheric vortices and their global consequences

    We report the in situ observation of a plasma vortex induced by a solar wind dynamic pressure enhancement in the nightside plasma sheet using multipoint measurements from Time History of Events and Macroscale Interactions during Substorms (THEMIS) satellites. The vortex has a scale of 5–10 Re and propagates several Re downtail, expanding while propagating. The features of the vortex are consistent with the prediction of the Sibeck (1990) model, and the vortex can penetrate deep (~8 Re ) in the dawn‐dusk direction and couple to field line oscillations. Global magnetohydrodynamics simulations are carried out, and it is found that the simulation and observations are consistent with each other. Data from THEMIS ground magnetometer stations indicate a poleward propagating vortex in the ionosphere, with a rotational sense consistent with the existence of the vortex observed in the magnetotail.     All-trans retinoic acid suppresses exocrine differentiation and branching morphogenesis in the embryonic pancreas

    Recent evidence has shown that retinoic acid (RA) signalling is required for early pancreatic development in zebrafish and frog but its role in later development in mammals is less clear cut. In the present study, we determined the effects of RA on the differentiation of the mouse embryonic pancreas. Addition of all-trans retinoic acid (atRA) to embryonic pancreatic cultures induced a number of changes. Branching morphogenesis and exocrine differentiation were suppressed and there was premature formation of endocrine cell clusters (although the total area of ÎČ cells was not different in control and atRA-treated buds). We investigated the mechanism of these changes and found that the premature formation of ÎČ cells was associated with the early expression of high-level Pdx1 in the endocrine cell clusters. In contrast, the suppressive effect of RA on exocrine differentiation may be due to a combination of two mechanisms (i) up-regulation of the extracellular matrix component laminin and (ii) enhancement of apoptosis. We also demonstrate that addition of fibroblast growth factor (FGF)-10 is able to partially prevent apoptosis and rescue exocrine differentiation and branching morphogenesis in atRA-treated cultures but not in mice lacking the FGF receptor 2-IIIb, suggesting the effects of FGF-10 are mediated through this receptor

    The Fueling and Evolution of AGN: Internal and External Triggers

    In this chapter, I review the fueling and evolution of active galactic nuclei (AGN) under the influence of internal and external triggers, namely intrinsic properties of host galaxies (morphological or Hubble type, color, presence of bars and other non-axisymmetric features, etc) and external factors such as environment and interactions. The most daunting challenge in fueling AGN is arguably the angular momentum problem as even matter located at a radius of a few hundred pc must lose more than 99.99 % of its specific angular momentum before it is fit for consumption by a BH. I review mass accretion rates, angular momentum requirements, the effectiveness of different fueling mechanisms, and the growth and mass density of black BHs at different epochs.     FGF receptor genes and breast cancer susceptibility: results from the Breast Cancer Association Consortium

    Background:Breast cancer is one of the most common malignancies in women. Genome-wide association studies have identified FGFR2 as a breast cancer susceptibility gene. Common variation in other fibroblast growth factor (FGF) receptors might also modify risk. We tested this hypothesis by studying genotyped single-nucleotide polymorphisms (SNPs) and imputed SNPs in FGFR1, FGFR3, FGFR4 and FGFRL1 in the Breast Cancer Association Consortium. Methods:Data were combined from 49 studies, including 53 835 cases and 50 156 controls, of which 89 050 (46 450 cases and 42 600 controls) were of European ancestry, 12 893 (6269 cases and 6624 controls) of Asian and 2048 (1116 cases and 932 controls) of African ancestry. Associations with risk of breast cancer, overall and by disease sub-type, were assessed using unconditional logistic regression. Results:Little evidence of association with breast cancer risk was observed for SNPs in the FGF receptor genes. The strongest evidence in European women was for rs743682 in FGFR3; the estimated per-allele odds ratio was 1.05 (95 confidence interval=1.02-1.09, P=0.0020), which is substantially lower than that observed for SNPs in FGFR2. Conclusion:Our results suggest that common variants in the other FGF receptors are not associated with risk of breast cancer to the degree observed for FGFR2. © 2014 Cancer Research UK

The primary focus is the theoretical and experimental developments of spin relaxation and dephasing in both spin precession in time domain and spin diffusion and transport in spacial domain.     Identifying Neuroimaging and Proteomic Biomarkers for MCI and AD via the Elastic Net

    poster abstractAbstract Multi-modal neuroimaging and biomarker data provide exciting opportunities to enhance our understanding of phenotypic characteristics associated with complex disorders. This study focuses on integrative analysis of structural MRI data and proteomic data from an RBM panel to examine their predictive power and identify relevant biomarkers in a large MCI/AD cohort. MRI data included volume and thickness measures of 98 regions estimated by FreeSurfer. RBM data included 146 proteomic analytes extracted from plasma and serum. A sparse learning model, elastic net logistic regression, was proposed to classify AD and MCI, and select disease-relevant biomarkers. A linear support vector machine coupled with feature selection was employed for comparison. Combining RBM and MRI data yielded improved prediction rates: HC vs AD (91.9%), HC vs MCI (90.5%) and MCI vs AD (86.5%). Elastic net identified a small set of meaningful imaging and proteomic biomarkers. The elastic net has great power to optimize the sparsity of feature selection while maintaining high predictive power. Its application to multi-modal imaging and biomarker data has considerable potential for discovering biomarkers and enhancing mechanistic understanding of AD and MCI

    Measurement of the Atmospheric Muon Spectrum from 20 to 3000 GeV

    The absolute muon flux between 20 GeV and 3000 GeV is measured with the L3 magnetic muon spectrometer for zenith angles ranging from 0 degree to 58 degree. Due to the large exposure of about 150 m2 sr d, and the excellent momentum resolution of the L3 muon chambers, a precision of 2.3 % at 150 GeV in the vertical direction is achieved. The ratio of positive to negative muons is studied between 20 GeV and 500 GeV, and the average vertical muon charge ratio is found to be 1.285 +- 0.003 (stat.) +- 0.019 (syst.).Comment: Total 32 pages, 9Figure
