    Baseline pathological data of the wedge clam Donax trunculus from the Tyrrhenian Sea (Mediterranean Basin)

    In recent years, a collapse in Donax trunculus fishing yields has occurred in the Tyrrhenian Sea (Mediterranean Basin). There is little information available on the impact disease may have had on D. trunculus populations. For the first time, a pathological survey was performed on the natural beds of the bivalve on the Campania and Lazio coasts, western Italy. Detected pathogens and related diseases were analysed, and their prevalence and mean intensity values were calculated. Viral particles, Chlamydia-like organisms, ciliates, coccidians, microcells and trematodes were observed. An unknown ciliate was linked to severe inflammatory and necrotic lesions in the digestive gland. Metacercariae of the trematode Postmonorchis sp. were also strongly represented in almost all samples, reaching high levels of infection; however, none of the pathogens described required the World Organisation for Animal Health to be notified. Initial results indicated that further surveys related to environmental data are necessary in order to assess the relevance of these early observations in managing the declining D. trunculus population in the Tyrrhenian Sea.postprin

    The disease perivellosa massive fibrin deposition (MPFD), is a condition characterized by uncontrolled mainly fibrin deposition intervillous space. The incidence worldwide is 0.028% per 1000 live births, there is only one case report where this condition is associated with trisomy 21, in our country there are no reports of this disease. The MPFD has high morbidity, obstetric mortality, recurrence, as well as neurodevelopmental significance of newborns. The etiology until the moment is unknown, difficult diagnosis and management for the obstetrician. The aim is to report MPFD association with trisomy 21 (T21) and a review of the medical literature regarding this condition

    Rac1 as a therapeutic target in ovarian cancer

    Rac1 is a high value therapeutic target for cancer based on its tumor promoting activities, yet clinical applications targeting Rac1 are in their infancy. High expression and hyperactivation of Rac1 in ovarian cancer, along with our identification of R-ketorolac as a novel Rac1 and Cdc42 selective inhibitor with translational potential, prompt us to test the hypothesis that targeting Rac1 has therapeutic utility for ovarian cancer. Ascites tumor cell samples from ovarian cancer patients in a prospective study receiving racemic ketorolac for clinically indicated use in pain relief were previously reported to show time dependent reduction of Rac1 and Cdc42 activities post-treatment. New RNA seq data of these patient samples reveals significant changes of genes involved in cell adhesion, cytokine-mediated signaling and cytokine production pathways. Conversely, the identified downregulated genes were overexpressed and associated with worse survival in ovarian cancer patients analyzed through The Cancer Genome Atlas (TCGA). Among the downregulated genes in the NOD pathway are chemokines and pro-inflammatory cytokines. Follow-up cytokine panels from patients confirm that racemic ketorolac treatment reduces the levels of immunosuppressive cytokines IL-6, IL-10 and RANTES in ascites fluids. Together, these data indicate there may be a benefit to the anti-inflammatory activity of the S- enantiomer, as well as the GTPase inhibitory activity of the R- enantiomer of ketorolac for ovarian cancer treatment

    Reduction in BACE1 decreases body weight, protects against diet-induced obesity and enhances insulin sensitivity in mice

    Insulin resistance and impaired glucose homoeostasis are important indicators of Type 2 diabetes and are early risk factors of AD (Alzheimer's disease). An essential feature of AD pathology is the presence of BACE1 (β-site amyloid precursor protein-cleaving enzyme 1), which regulates production of toxic amyloid peptides. However, whether BACE1 also plays a role in glucose homoeostasis is presently unknown. We have used transgenic mice to analyse the effects of loss of BACE1 on body weight, and lipid and glucose homoeostasis. BACE1−/− mice are lean, with decreased adiposity, higher energy expenditure, and improved glucose disposal and peripheral insulin sensitivity than wild-type littermates. BACE1−/− mice are also protected from diet-induced obesity. BACE1-deficient skeletal muscle and liver exhibit improved insulin sensitivity. In a skeletal muscle cell line, BACE1 inhibition increased glucose uptake and enhanced insulin sensitivity. The loss of BACE1 is associated with increased levels of UCP1 (uncoupling protein 1) in BAT (brown adipose tissue) and UCP2 and UCP3 mRNA in skeletal muscle, indicative of increased uncoupled respiration and metabolic inefficiency. Thus BACE1 levels may play a critical role in glucose and lipid homoeostasis in conditions of chronic nutrient excess. Therefore strategies that ameliorate BACE1 activity may be important novel approaches for the treatment of diabetes

    Precipitation extremes over La Plata Basin –Review and new results from observations and climate simulations

    Monthly and daily precipitation extremes over La Plata Basin (LPB) are analyzed in the framework of the CLARIS-LPB Project. A review of the studies developed during the project and results of additional research are presented and discussed. Specific aspects of analysis are focused on large-scale versus local processes impacts on the intensity and frequency of precipitation extremes over LPB, and on the assessment of specific wet and dry spell indices and their changed characteristics in future climate scenarios. The analysis is shown for both available observations of precipitation in the region and ad-hoc global and regional models experiments. The Pacific, Indian and Atlantic Oceans can all impact precipitation intensity and frequency over LPB. In particular, considering the Pacific sector, different types of ENSO events (i.e. canonical vs Modoki or East vs Central) have different influences. Moreover, model projections indicate an increase in the frequency of precipitation extremes over LPB during El Niño and La Ninã events in future climate. Local forcings can also be important for precipitation extremes. Here, the feedbacks between soil moisture and extreme precipitation in LPB are discussed based on hydric conditions in the region and model sensitivity experiments. Concerning droughts, it was found that they were more frequent in the western than in the eastern sector of LPB during the period of 1962–2008. On the other hand, observations and model experiments agree in that the monthly wet extremes were more frequent than the dry extremes in the northern and southern LPB sectors during the period 1979–2001, with higher frequency in the south.Published211-2304A.     Nitrate and nitrite in drinking water affect antioxidant enzymes in erythrocytes of rats

    The present study evaluated the effect of short term intake of nitrite and nitrate drinking water on the antioxidant system and membrane damage of rat erythrocytes. Wistar rats were randomly divided into three groups as follows; the group I received only distilled water ad libitum; the group II was given water with nitrate (a dose of 124 mg/kg of nitrate-nitrogen) as drinking water and the group III was given nitrites dissolved in distilled water in a dose of 150 mg/kg for 7 days. At the end of the study, group III rats showed a significant decrease in activities of glutathione peroxidase (GPx), glucose 6-phosphate dehydrogenase (G6PDH) and catalase (CAT), while in group II rats, the activity of GPx and CAT were significantly reduced, but no significant changes in glutathione reductase activity and peroxynitrite levels were observed. On the other hand, malondialdehyde (MDA) was increased in both groups with respect to group I. Also, our major results indicate that all treatments changed methemoglobin levels and osmotic fragility in comparison to group I rats. The intensity of alterations was found more severe in rats of group III, followed by rats of group II. It can be concluded from these observations that nitrate or nitrite leads to alterations in the erythrocytes antioxidant defense status mainly throughout NADPH relate enzymes

    Influence of radiation and TiO2 concentration on the hydroxyl radicals generation in a photocatalytic LED reactor. Application to dodecylbenzenesulfonate degradation

    One of the main issues associated to the development of photocatalysis is the lack of adequate indexes that allow the comparison of the results obtained in different experimental setup designs. The hydroxyl radicals (o OH) generation rate is a key factor to determine the overall oxidation rate. In this work, using a Light Emitting Diodes (LEDs) reactor aimed to maximize light ef?ciency and minimize energy consumption, the o OH generated have been determined as a function of the radiation and catalyst concentration following an indirect method based on the reaction between o OH and dimethyl sulfoxide (DMSO) to produce formaldehyde. Finally, the methodology has been applied to analyze the degradation kinetics of the anionic surfactant dodecylbenzenesulfonate (DBS), frequently used in shampoo formulations and detergents for washing machines.     Measurement of the cross-section and charge asymmetry of WW bosons produced in proton-proton collisions at s=8\sqrt{s}=8 TeV with the ATLAS detector

    This paper presents measurements of the W+μ+νW^+ \rightarrow \mu^+\nu and WμνW^- \rightarrow \mu^-\nu cross-sections and the associated charge asymmetry as a function of the absolute pseudorapidity of the decay muon. The data were collected in proton--proton collisions at a centre-of-mass energy of 8 TeV with the ATLAS experiment at the LHC and correspond to a total integrated luminosity of 20.2~\mbox{fb^{-1}}. The precision of the cross-section measurements varies between 0.8% to 1.5% as a function of the pseudorapidity, excluding the 1.9% uncertainty on the integrated luminosity. The charge asymmetry is measured with an uncertainty between 0.002 and 0.003. The results are compared with predictions based on next-to-next-to-leading-order calculations with various parton distribution functions and have the sensitivity to discriminate between them.Comment: 38 pages in total, author list starting page 22, 5 figures, 4 tables, submitted to EPJC.     Search for chargino-neutralino production with mass splittings near the electroweak scale in three-lepton final states in √s=13 TeV pp collisions with the ATLAS detector

    A search for supersymmetry through the pair production of electroweakinos with mass splittings near the electroweak scale and decaying via on-shell W and Z bosons is presented for a three-lepton final state. The analyzed proton-proton collision data taken at a center-of-mass energy of √s=13  TeV were collected between 2015 and 2018 by the ATLAS experiment at the Large Hadron Collider, corresponding to an integrated luminosity of 139  fb−1. A search, emulating the recursive jigsaw reconstruction technique with easily reproducible laboratory-frame variables, is performed. The two excesses observed in the 2015–2016 data recursive jigsaw analysis in the low-mass three-lepton phase space are reproduced. Results with the full data set are in agreement with the Standard Model expectations. They are interpreted to set exclusion limits at the 95% confidence level on simplified models of chargino-neutralino pair production for masses up to 345 GeV
