Silver(I) and Copper(II) 1,10-Phenanthroline-5,6-dione Complexes as Promising Antivirulence Strategy against Leishmania: Focus on Gp63 (Leishmanolysin)
Leishmaniasis, caused by protozoa of the genus Leishmania, encompasses a group of neglected diseases with diverse clinical and epidemiological manifestations that can be fatal if not adequately and promptly managed/treated. The current chemotherapy options for this disease are expensive, require invasive administration and often lead to severe side effects. In this regard, our research group has previously reported the potent anti-Leishmania activity of two coordination compounds (complexes) derived from 1,10-phenanthroline-5,6-dione (phendione): [Cu(phendione)3 ].(ClO4 )2 .4H2O and [Ag(phendione)2 ].ClO4 . The present study aimed to evaluate the effects of these complexes on leishmanolysin (gp63), a virulence factor produced by all Leishmania species that plays multiple functions and is recognized as a potential target for antiparasitic drugs. The results showed that both Ag-phendione (â74.82 kcal/mol) and Cu-phendione (â68.16 kcal/mol) were capable of interacting with the amino acids comprising the active site of the gp63 protein, exhibiting more favorable interaction energies compared to phendione alone (â39.75 kcal/mol) or 1,10-phenanthroline (â45.83 kcal/mol; a classical gp63 inhibitor) as judged by molecular docking assay. The analysis of kinetic parameters using the fluorogenic substrate Z-Phe-Arg-AMC indicated Vmax and apparent Km values of 0.064 ”M/s and 14.18 ”M, respectively, for the released gp63. The effects of both complexes on gp63 proteolytic activity were consistent with the in silico assay, where Ag-phendione exhibited the highest gp63 inhibition capacity against gp63, with an IC50 value of 2.16 ”M and the lowest inhibitory constant value (Ki = 5.13 ”M), followed by Cu-phendione (IC50 = 163 ”M and Ki = 27.05 ”M). Notably, pretreatment of live L. amazonensis promastigotes with the complexes resulted in a significant reduction in the expression of gp63 protein, including the isoforms located on the parasite cell surface. Both complexes markedly decreased the in vitro association indexes between L. amazonensis promastigotes and THP-1 human macrophages; however, this effect was reversed by the addition of soluble gp63 molecules to the interaction medium. Collectively, our findings highlight the potential use of these potent complexes in antivirulence therapy against Leishmania, offering new insights for the development of effective treatments for leishmaniasis
Mitotic index of epithelia cells in gills of Guppy (Poecilia vivipara) exposed to fractions of the leaf and bark of pequi (Caryocar brasiliensis)
Many wild plants of the Cerrado bioma are used as medicinal plants. The pequi (Caryocar brasiliensis) is representative of those equipped with molluscicidal action against schistosomiasis. This paper sets out to verify the toxic action of pequi fractions in other aquatic animals before they are used in rivers or lakes. Analysis were made of the alterations to the mitotic index of epithelia cells in the gills of guppy (Poecilia vivipara) exposed to ethyl-acetate fraction of leaf pequi and bark. Animals exposed to ethyl-acetate fraction (leaf), no significant change was observed in the mitotic index when compared with the control group, whereas in fish exposed to ethyl-acetate fraction (bark) we detected an increase in mitotic index of the epithelia cells in two regions of the branchial filaments. Thus, ethyl-acetate fractions (leaf) could be used against schistosomiasis, given their high efficacy and low action as piscicide.VĂĄrias plantas nativas do bioma Cerrado sĂŁo utilizadas como plantas medicinais. Constatamos que nenhuma das fraçÔes se mostrou letal aos peixes. Os animais expostos Ă fração acetato de etila da folha nĂŁo apresentaram modificaçÔes significativas no Ăndice mitĂłtico em relação ao grupo controle, mas os animais expostos Ă fração acetato de etila da casca do caule apresentaram aumento do Ăndice mitĂłtico das cĂ©lulas epiteliais em duas regiĂ”es dos filamentos branquiais. Desta forma, a fração acetato de etila da folha poderia ser utilizada como moluscicida em mananciais, enquanto que a fração acetato de etila da casca do caule necessitaria passar por outros testes mais especĂficos.22122
Study of the production of and hadrons in collisions and first measurement of the branching fraction
The product of the () differential production
cross-section and the branching fraction of the decay () is
measured as a function of the beauty hadron transverse momentum, ,
and rapidity, . The kinematic region of the measurements is and . The measurements use a data sample
corresponding to an integrated luminosity of collected by the
LHCb detector in collisions at centre-of-mass energies in 2011 and in 2012. Based on previous LHCb
results of the fragmentation fraction ratio, , the
branching fraction of the decay is

Evidence for the strangeness-changing weak decay
Using a collision data sample corresponding to an integrated luminosity
of 3.0~fb, collected by the LHCb detector, we present the first search
for the strangeness-changing weak decay . No
hadron decay of this type has been seen before. A signal for this decay,
corresponding to a significance of 3.2 standard deviations, is reported. The
relative rate is measured to be
, where and

Measurements of long-range near-side angular correlations in TeV proton-lead collisions in the forward region
Two-particle angular correlations are studied in proton-lead collisions at a
nucleon-nucleon centre-of-mass energy of TeV, collected
with the LHCb detector at the LHC. The analysis is based on data recorded in
two beam configurations, in which either the direction of the proton or that of
the lead ion is analysed. The correlations are measured in the laboratory
system as a function of relative pseudorapidity, , and relative
azimuthal angle, , for events in different classes of event
activity and for different bins of particle transverse momentum. In
high-activity events a long-range correlation on the near side, , is observed in the pseudorapidity range . This
measurement of long-range correlations on the near side in proton-lead
collisions extends previous observations into the forward region up to
. The correlation increases with growing event activity and is found
to be more pronounced in the direction of the lead beam. However, the
correlation in the direction of the lead and proton beams are found to be
compatible when comparing events with similar absolute activity in the

flavour tagging using charm decays at the LHCb experiment
An algorithm is described for tagging the flavour content at production of
neutral mesons in the LHCb experiment. The algorithm exploits the
correlation of the flavour of a meson with the charge of a reconstructed
secondary charm hadron from the decay of the other hadron produced in the
proton-proton collision. Charm hadron candidates are identified in a number of
fully or partially reconstructed Cabibbo-favoured decay modes. The algorithm is
calibrated on the self-tagged decay modes and using of data collected by the LHCb
experiment at centre-of-mass energies of and
. Its tagging power on these samples of

Search for direct production of charginos and neutralinos in events with three leptons and missing transverse momentum in âs = 7 TeV pp collisions with the ATLAS detector
A search for the direct production of charginos and neutralinos in final states with three electrons or muons and missing transverse momentum is presented. The analysis is based on 4.7 fbâ1 of protonâproton collision data delivered by the Large Hadron Collider and recorded with the ATLAS detector. Observations are consistent with Standard Model expectations in three signal regions that are either depleted or enriched in Z-boson decays. Upper limits at 95% confidence level are set in R-parity conserving phenomenological minimal supersymmetric models and in simplified models, significantly extending previous results
Measurement of the polarisation of W bosons produced with large transverse momentum in pp collisions at sqrt(s) = 7 TeV with the ATLAS experiment
This paper describes an analysis of the angular distribution of W->enu and
W->munu decays, using data from pp collisions at sqrt(s) = 7 TeV recorded with
the ATLAS detector at the LHC in 2010, corresponding to an integrated
luminosity of about 35 pb^-1. Using the decay lepton transverse momentum and
the missing transverse energy, the W decay angular distribution projected onto
the transverse plane is obtained and analysed in terms of helicity fractions
f0, fL and fR over two ranges of W transverse momentum (ptw): 35 < ptw < 50 GeV
and ptw > 50 GeV. Good agreement is found with theoretical predictions. For ptw
> 50 GeV, the values of f0 and fL-fR, averaged over charge and lepton flavour,
are measured to be : f0 = 0.127 +/- 0.030 +/- 0.108 and fL-fR = 0.252 +/- 0.017
+/- 0.030, where the first uncertainties are statistical, and the second
