    Nonnative Ungulate Impacts on Greater Sage-grouse Late Brood-rearing Habitat in the Great Basin, USA

    Domestic livestock grazing is the dominant land use on much of the current range inhabited by greater sage-grouse (Centrocercus urophasianus; sage-grouse) in the western United States. Nonnative feral horses (Equus ferus caballus) also inhabit important sage-grouse seasonal habitats. Overabundant feral horse populations and improper grazing by domestic cattle (Bos taurus) can impact the health of sagebrush (Artemisia spp.) and desert shrub rangeland communities and native wildlife. These impacts to sage-grouse can be exacerbated when they affect late brood-rearing habitat, which provide the forbs and arthropods required to fledge broods. Managers require better information regarding the extent of these impacts. In 2020, we assessed the potential impact of feral horses and domestic cattle on sage-grouse late brood-rearing habitats in western Utah and eastern Nevada, USA. We acquired late brood-rearing location data from sage-grouse marked with global positioning system and very-high frequency radio-transmitters from 2016 to 2020 for North Utah data, 2017 to 2018 for South Utah data, and 1961 to 2017 for both east and west Nevada data to delineate late brood-rearing habitats. Using these location data, we compared 8 sites (4 pairs) within horse and non-horse use areas to assess sage-grouse habitat quality characteristics between areas that have been predominantly horse and cattle grazed versus sites that have been predominantly cattle grazed. For each pairing, 1 site was located within and the other outside of a Bureau of Land Management herd management area boundary, and both sites shared similar habitat characteristics (i.e., topography, dominant vegetation, soils, and climate) and selection probability for broods. We collected vegetation and dung count data at each site to assess characteristics related to habitat quality for sage-grouse brood-rearing, based on ungulate presence. We used a mixed model analysis of variance to detect differences between each paired site comparison (α \u3c 0.01). Horses or evidence of horse presence (i.e., dung) were not detected at our non-horse sites allowing for an unbiased comparison between paired sites. Cattle presence was noted at all our paired sites. Average annual grass frequency was 0.74 in horse and 0.17 in non-horse use areas (P = 0.20), and average annual grass cover was 4.0% compared to 0.2% in horse use areas (P = 0.32). Average annual grass biomass was 0.45 kg/ha in horse and 0.04% in non-horse use areas (P = 0.34). Vegetation height was 44.2 cm in non-horse compared to 34.5 cm in horse use areas (P = 0.23). These results suggest that increased ungulate grazing and year-long use of late brood-rearing habitat by feral horses coupled with livestock grazing may impair habitat suitability, particularly considering ecological impacts from invasive plant species. Our results suggest that managing late brood-rearing habitats to reduce the frequency and intensity of year-long grazing by feral horses can be best accomplished by reducing horse numbers and the seasonal distribution of grazing by domestic livestock

    Practice Patterns and Preferences Among Hematopoietic Cell Transplantation Clinicians

    Hematopoietic cell transplantation can cure many high-risk diseases but is associated with complexity, cost, and risk. Several areas in transplantation practice were identified in the 2014 Blood and Marrow Transplant Clinical Trials Network State of the Science Symposium (BMT CTN SOSS) as high priorities for further study. We developed a survey for hematopoietic cell transplantation clinicians to identify current practices in BMT CTN SOSS priority areas and to understand, more generally, the variation in approach to transplantation and estimation of transplantation benefit in current medical practice. Of 1439 transplantation clinicians surveyed, 305 responded (20% response rate). Clinicians were well represented by age, experience, geography, and size of practice. We found that several techniques identified in the BMT CTN SOSS, such as maintenance therapy for acute myeloid leukemia or myelodysplastic syndromes after allogeneic transplantation, were already being utilized in practice on and off study, with higher rates of use in higher-volume centers. There was significant variation among clinicians in use of transplantation technologies and approaches to common transplantation scenarios. Appraisals of risks and benefits of transplantation appeared to converge upon similar estimates despite the presentation of different hypothetical scenarios. These results suggest overall equipoise in several BMT CTN SOSS high-priority areas and support the need for better data to inform clinical practice

    Validation of Walk Score® for Estimating Neighborhood Walkability: An Analysis of Four US Metropolitan Areas

    Neighborhood walkability can influence physical activity. We evaluated the validity of Walk Score® for assessing neighborhood walkability based on GIS (objective) indicators of neighborhood walkability with addresses from four US metropolitan areas with several street network buffer distances (i.e., 400-, 800-, and 1,600-meters). Address data come from the YMCA-Harvard After School Food and Fitness Project, an obesity prevention intervention involving children aged 5–11 years and their families participating in YMCA-administered, after-school programs located in four geographically diverse metropolitan areas in the US (n = 733). GIS data were used to measure multiple objective indicators of neighborhood walkability. Walk Scores were also obtained for the participant’s residential addresses. Spearman correlations between Walk Scores and the GIS neighborhood walkability indicators were calculated as well as Spearman correlations accounting for spatial autocorrelation. There were many significant moderate correlations between Walk Scores and the GIS neighborhood walkability indicators such as density of retail destinations and intersection density (p < 0.05). The magnitude varied by the GIS indicator of neighborhood walkability. Correlations generally became stronger with a larger spatial scale, and there were some geographic differences. Walk Score® is free and publicly available for public health researchers and practitioners. Results from our study suggest that Walk Score® is a valid measure of estimating certain aspects of neighborhood walkability, particularly at the 1600-meter buffer. As such, our study confirms and extends the generalizability of previous findings demonstrating that Walk Score is a valid measure of estimating neighborhood walkability in multiple geographic locations and at multiple spatial scales

    Proteasome subunit variants cause neurosensory syndrome combining deafness and cataract due to proteotoxic stress

    The ubiquitin–proteasome system degrades ubiquitin‐modified proteins to maintain protein homeostasis and to control signalling. Whole‐genome sequencing of patients with severe deafness and early‐onset cataracts as part of a neurological, sensorial and cutaneous novel syndrome identified a unique deep intronic homozygous variant in the PSMC3 gene, encoding the proteasome ATPase subunit Rpt5, which lead to the transcription of a cryptic exon. The proteasome content and activity in patient\u27s fibroblasts was however unaffected. Nevertheless, patient\u27s cells exhibited impaired protein homeostasis characterized by accumulation of ubiquitinated proteins suggesting severe proteotoxic stress. Indeed, the TCF11/Nrf1 transcriptional pathway allowing proteasome recovery after proteasome inhibition is permanently activated in the patient\u27s fibroblasts. Upon chemical proteasome inhibition, this pathway was however impaired in patient\u27s cells, which were unable to compensate for proteotoxic stress although a higher proteasome content and activity. Zebrafish modelling for knockout in PSMC3 remarkably reproduced the human phenotype with inner ear development anomalies as well as cataracts, suggesting that Rpt5 plays a major role in inner ear, lens and central nervous system development

    Alcohol, Volatile Fatty Acid, Phenol, and Methane Emissions from Dairy Cows and Fresh Manure

    There are approximately 2.5 million dairy cows in California. Emission inventories list dairy cows and their manure as the major source of regional air pollutants, but data on their actual emissions remain sparse, particularly for smog-forming volatile organic compounds (VOCs) and greenhouse gases (GHGs). We report measurements of alcohols, volatile fatty acids, phenols, and methane (CH4) emitted from nonlactating (dry) and lactating dairy cows and their manure under controlled conditions. The experiment was conducted in an environmental chamber that simulates commercial concrete-floored freestall cow housing conditions. The fluxes of methanol, ethanol, and CH4 were measured from cows and/or their fresh manure. The average estimated methanol and ethanol emissions were 0.33 and 0.51 g cow−1 h−1 from dry cows and manure and 0.7 and 1.27 g cow−1 h−1 from lactating cows and manure, respectively. Both alcohols increased over time, coinciding with increasing accumulation of manure on the chamber floor. Volatile fatty acids and phenols were emitted at concentrations close to their detection limit. Average estimated CH4emissions were predominantly associated with enteric fermentation from cows rather than manure and were 12.35 and 18.23 g cow−1 h−1 for dry and lactating cows, respectively. Lactating cows produced considerably more gaseous VOCs and GHGs emissions than dry cows (P \u3c 0.001). Dairy cows and fresh manure have the potential to emit considerable amounts of alcohols and CH4 and research is needed to determine effective mitigation

    Risk portfolio on modern finance has become increasingly technical, requiring the use of sophisticated mathematical tools in both research and practice. Since companies cannot insure themselves completely against risk, as human incompetence in predicting the future precisely that written in Al-Quran surah Luqman verse 34, they have to manage it to yield an optimal portfolio. The objective here is to minimize the variance among all portfolios, or alternatively, to maximize expected return among all portfolios that has at least a certain expected return. Furthermore, this study focuses on optimizing risk portfolio so called Markowitz MVO (Mean-Variance Optimization). Some theoretical frameworks for analysis are arithmetic mean, geometric mean, variance, covariance, linear programming, and quadratic programming. Moreover, finding a minimum variance portfolio produces a convex quadratic programming, that is minimizing the objective function ðð¥with constraintsð ð 𥠥 ðandð´ð¥ = ð. The outcome of this research is the solution of optimal risk portofolio in some investments that could be finished smoothly using MATLAB R2007b software together with its graphic analysis

    Search for heavy resonances decaying to two Higgs bosons in final states containing four b quarks

    A search is presented for narrow heavy resonances X decaying into pairs of Higgs bosons (H) in proton-proton collisions collected by the CMS experiment at the LHC at root s = 8 TeV. The data correspond to an integrated luminosity of 19.7 fb(-1). The search considers HH resonances with masses between 1 and 3 TeV, having final states of two b quark pairs. Each Higgs boson is produced with large momentum, and the hadronization products of the pair of b quarks can usually be reconstructed as single large jets. The background from multijet and t (t) over bar events is significantly reduced by applying requirements related to the flavor of the jet, its mass, and its substructure. The signal would be identified as a peak on top of the dijet invariant mass spectrum of the remaining background events. No evidence is observed for such a signal. Upper limits obtained at 95 confidence level for the product of the production cross section and branching fraction sigma(gg -> X) B(X -> HH -> b (b) over barb (b) over bar) range from 10 to 1.5 fb for the mass of X from 1.15 to 2.0 TeV, significantly extending previous searches. For a warped extra dimension theory with amass scale Lambda(R) = 1 TeV, the data exclude radion scalar masses between 1.15 and 1.55 TeV
