    Intact procedural motor sequence learning in developmental coordination disorder

    The purpose of the present study was to explore the possibility of a procedural learning deficit among children with developmental coordination disorder (DCD). We tested 34 children aged 6–12 years with and without DCD using the serial reaction time task, in which the standard keyboard was replaced by a touch screen in order to minimize the impact of perceptuomotor coordination difficulties that characterize this disorder. The results showed that children with DCD succeed as well as control children at the procedural sequence learning task. These findings challenge the hypothesis that a procedural learning impairment underlies the difficulties of DCD children in acquiring and automatizing daily activities. We suggest that the previously reported impairment of children with DCD on the serial reaction time task is not due to a sequence learning deficit per se, but rather due to methodological factors such as the response mode used in these studies.Peer reviewe

    Analysis of Endocytic Pathways in Drosophila Cells Reveals a Conserved Role for GBF1 in Internalization via GEECs

    In mammalian cells, endocytosis of the fluid phase and glycosylphosphatidylinositol-anchored proteins (GPI-APs) forms GEECs (GPI-AP enriched early endosomal compartments) via an Arf1- and Cdc42-mediated, dynamin independent mechanism. Here we use four different fluorescently labeled probes and several markers in combination with quantitative kinetic assays, RNA interference and high resolution imaging to delineate major endocytic routes in Drosophila cultured cells. We find that the hallmarks of the pinocytic GEEC pathway are conserved in Drosophila and identify garz, the fly ortholog of the GTP exchange factor GBF1, as a novel component of this pathway. Live confocal and TIRF imaging reveals that a fraction of GBF1 GFP dynamically associates with ABD RFP (a sensor for activated Arf1 present on nascent pinosomes). Correspondingly, a GTP exchange mutant of GBF1 has altered ABD RFP localization in the evanescent field and is impaired in fluid phase uptake. Furthermore, GBF1 activation is required for the GEEC pathway even in the presence of Brefeldin A, implying that, like Arf1, it has a role in endocytosis that is separable from its role in secretion

    Is procedural memory enhanced in Tourette syndrome? evidence from a sequence learning task

    Procedural memory, which is rooted in the basal ganglia, underlies the learning and processing of numerous automatized motor and cognitive skills, including in language. Not surprisingly, disorders with basal ganglia abnormalities have been found to show impairments of procedural memory. However, brain abnormalities could also lead to atypically enhanced function. Tourette syndrome (TS) is a candidate for enhanced procedural memory, given previous findings of enhanced TS processing of grammar, which likely depends on procedural memory. We comprehensively examined procedural learning, from memory formation to retention, in children with TS and typically developing (TD) children, who performed an implicit sequence learning task over two days. The children with TS showed sequence learning advantages on both days, despite a regression of sequence knowledge overnight to the level of the TD children. This is the first demonstration of procedural learning advantages in any disorder. The findings may further our understanding of procedural memory and its enhancement. The evidence presented here, together with previous findings suggesting enhanced grammar processing in TS, underscore the dependence of language on a system that also subserves visuomotor sequencing

    Apprentissage procédural chez l'enfant: études développementales et cliniques

    Nous avons étudié les capacités d'apprentissage procédural (verbal, moteur et cognitif) chez l'enfant normal, l'enfant avec atteinte des noyaux gris centraux et l'enfant avec trouble spécifique de langage (TSDL). Les résultats mettent en évidence 1) des capacités similaires d'apprentissage procédural dans les trois domaines chez les enfants de 8 à12 ans, suggérant une invariance développementale 2) une atteinte globale des apprentissages procéduraux chez les enfants avec pathologie des noyaux gris centraux, sans évidence de plasticité et 3) un déficit procédural verbal chez les enfants avec TSDL, associé à un déficit procédural moteur lors de la co-morbidité avec un trouble de la coordination motrice, et combiné avec un déficit procédural cognitif lors de la présence de mouvements choréiformes suggestifs d'une dysfonction striatale. Les résultats chez l'enfant indiquent la présence de sous-systèmes fonctionnellement distincts au sein du système procédural et une indépendance des systèmes mnésiques procédural et déclaratif, comme chez l'adulte
