    Assembly and structural analysis of a covalently closed nano-scale DNA cage

    The inherent properties of DNA as a stable polymer with unique affinity for partner molecules determined by the specific Watson–Crick base pairing makes it an ideal component in self-assembling structures. This has been exploited for decades in the design of a variety of artificial substrates for investigations of DNA-interacting enzymes. More recently, strategies for synthesis of more complex two-dimensional (2D) and 3D DNA structures have emerged. However, the building of such structures is still in progress and more experiences from different research groups and different fields of expertise are necessary before complex DNA structures can be routinely designed for the use in basal science and/or biotechnology. Here we present the design, construction and structural analysis of a covalently closed and stable 3D DNA structure with the connectivity of an octahedron, as defined by the double-stranded DNA helices that assembles from eight oligonucleotides with a yield of ∌30%. As demonstrated by Small Angle X-ray Scattering and cryo-Transmission Electron Microscopy analyses the eight-stranded DNA structure has a central cavity larger than the apertures in the surrounding DNA lattice and can be described as a nano-scale DNA cage, Hence, in theory it could hold proteins or other bio-molecules to enable their investigation in certain harmful environments or even allow their organization into higher order structures

    Die UniversitĂ€t Siegen beschĂ€ftigt sich seit ĂŒber 15 Jahren wissenschaftlich und im Bereich der anwendungsorientierten Forschung mit diesem Thema und hat dazu mittlerweile fĂŒnf Symposien durchgefĂŒhrt. Mit der Veröffentlichung soll die langjĂ€hrige Tradition als etablierte wissenschaftliche Plattform mit einem Wissensaustausch auf europĂ€ischer Ebene fortgesetzt werden. Die Bearbeitung dieser Thematik erfolgt auf der Basis der bewĂ€hrten Kooperation zwischen Geotechnik und Wasserbau an der UniversitĂ€t Siegen. Aktuelle Ereignisse, wie z.B. die aus England oder Australien im Februar des Jahres 2014, machen uns aber auch deutlich, dass ein absoluter Schutz gegen Extremereignisse nicht möglich ist. Sie zeigen aber auch, dass dort wo technischer Hochwasserschutz konsequent umgesetzt wurde SchĂ€den vermieden werden konnten. Wir sind nach den Ereignissen in den vergangenen Jahren aufgefordert wissenschaftlich noch leistungsfĂ€higere und duktilere Systeme zu entwickeln. Weiter ist die Wissenschaft in der Pflicht, die Zivile Sicherheit im Hochwasser-schutz permanent zu bewerten, zu bearbeiten und ganzheitliche-interdisziplinĂ€re und lĂ€nderĂŒbergreifende Lösungen fĂŒr die Zivilgesellschaft einzufordern

    The main objective of the ACRIDICON-CHUVA campaign in September 2014 was the investigation of aerosol-cloud-interactions in the Amazon Basin. Cloud properties near cloud base of growing convective cumuli were characterized by cloud droplet size distribution measurements using a cloud combination probe and a cloud and aerosol spectrometer.     Polygenic Risk Scores for Prediction of Breast Cancer and Breast Cancer Subtypes

    Stratification of women according to their risk of breast cancer based on polygenic risk scores (PRSs) could improve screening and prevention strategies. Our aim was to develop PRSs, optimized for prediction of estrogen receptor (ER)-specific disease, from the largest available genome-wide association dataset and to empirically validate the PRSs in prospective studies. The development dataset comprised 94,075 case subjects and 75,017 control subjects of European ancestry from 69 studies, divided into training and validation sets. Samples were genotyped using genome-wide arrays, and single-nucleotide polymorphisms (SNPs) were selected by stepwise regression or lasso penalized regression. The best performing PRSs were validated in an independent test set comprising 11,428 case subjects and 18,323 control subjects from 10 prospective studies and 190,040 women from UK Biobank (3,215 incident breast cancers). For the best PRSs (313 SNPs), the odds ratio for overall disease per 1 standard deviation in ten prospective studies was 1.61 (95%CI: 1.57-1.65) with area under receiver-operator curve (AUC) = 0.630 (95%CI: 0.628-0.651). The lifetime risk of overall breast cancer in the top centile of the PRSs was 32.6%. Compared with women in the middle quintile, those in the highest 1% of risk had 4.37- and 2.78-fold risks, and those in the lowest 1% of risk had 0.16- and 0.27-fold risks, of developing ER-positive and ER-negative disease, respectively. Goodness-of-fit tests indicated that this PRS was well calibrated and predicts disease risk accurately in the tails of the distribution.     Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements

    Zur Messung der Wirksamkeit von Thienopyridinen gibt es zahlreiche Messverfahren. Ziel dieser Arbeit war es, die klassische Impedanzaggregometrie (WBA) mit der durchflusszytometrisch gemessenen P-Selectin Expression, der In-vitro Blutungszeit (IVBT) und der Mehrelektrodenaggregometrie (MEA) zu vergleichen. Bei allen Messverfahren wurde ADP als Thrombozytenaktivator verwendet. Messungen an gesunden Probanden dienten zur Ermittlung von Referenzwerten. Der direkte Vergleich der Messverfahren erfolgte anhand von Messungen an kardiologischen Patienten unter Thienopyridinmedikation. Es zeigte sich, dass IVBT und P-Selectin Expression nicht sicher zwischen gesunden Probanden und Patienten unter Therapie differenzieren konnten. Die Messwerte von WBA, IVBT und P-Selectin Expression korrelierten nur schwach miteinander. Die Messwerte von WBA und MEA hingegen zeigten eine gute Korrelation. Beide Verfahren scheinen zur Messung der Wirksamkeit antithrombozytÀrer Medikamente geeignet zu sein

    International audienceAbstract. Aerosol–cloud interactions contribute to the large uncertainties in current estimates of climate forcing. We investigated the effect of aerosol particles on cloud droplet formation by model calculations and aircraft measurements over the Amazon and over the western tropical Atlantic during the ACRIDICON–CHUVA campaign in September 2014. On the HALO (High Altitude Long Range Research) research aircraft, cloud droplet number concentrations (Nd) were measured near the base of clean and polluted growing convective cumuli using a cloud combination probe (CCP) and a cloud and aerosol spectrometer (CAS-DPOL). An adiabatic parcel model was used to perform cloud droplet number closure studies for flights in differently polluted air masses. Model input parameters included aerosol size distributions measured with an ultra-high sensitive aerosol spectrometer (UHSAS), in combination with a condensation particle counter (CPC). Updraft velocities (w) were measured with a boom-mounted Rosemount probe. Over the continent, the aerosol size distributions were dominated by accumulation mode particles, and good agreement between measured and modeled Nd values was obtained (deviations â‰Č 10 %) assuming an average hygroscopicity of Îș∌0.1, which is consistent with Amazonian biomass burning and secondary organic aerosol. Above the ocean, fair agreement was obtained assuming an average hygroscopicity of Îș∌0.2 (deviations â‰Č 16 %) and further improvement was achieved assuming different hygroscopicities for Aitken and accumulation mode particles (ÎșAit=0.8, Îșacc=0.2; deviations â‰Č 10 %), which may reflect secondary marine sulfate particles. Our results indicate that Aitken mode particles and their hygroscopicity can be important for droplet formation at low pollution levels and high updraft velocities in tropical convective clouds
