    Characterization of Chlorella vulgaris and Chlorella protothecoides using multi-pixel photon counters in a 3D focusing optofluidic system

    Analysis of microparticle size and fluorescence intensity can be used to classify microparticles. We designed and fabricated an optofluidic system that characterizes microparticles, including fluorescent microparticles and microalgae. A new type of multi-pixel photon counter (MPPC) was employed to miniaturize the device, lower its power consumption, and make it insensitive to magnetic fields. The system uses a 635 nm laser for excitation of the microparticles\u27 fluorescence. The scattered light from the fluorescent microparticles, as well as Chlorella vulgaris and Chlorella protothecoides, were measured. Additionally, we analyzed the width and height of the measured signals generated as a result of microparticles and microalgae passing through the interrogation region. Using this device, we distinguished fluorescent microparticles of sizes of 3.2 μm and 10.2 μm. Additionally, different types of microalgae were distinguished using this high-throughput 3D focusing system. The analysis of the experimental results also confirms the capability of the device for monitoring of algae growth over time. The unique properties of MPPC as optical detector for both laboratory and field applications are demonstrated

    Proteomic Analysis of Anti-Tumor Effects of 11-Dehydrosinulariolide on CAL-27 Cells

    The anti-tumor effects of 11-dehydrosinulariolide, an active ingredient isolated from soft coral Sinularia leptoclados, on CAL-27 cells were investigated in this study. In the MTT assay for cell proliferation, increasing concentrations of 11-dehydrosinulariolide decreased CAL-27 cell viability. When a concentration of 1.5 μg/mL of 11-dehydrosinulariolide was applied, the CAL-27 cells viability was reduced to a level of 70% of the control sample. The wound healing function decreased as the concentration of 11-dehydrosinulariolide increased. The results in this study indicated that treatment with 11-dehydrosinulariolide for 6 h significantly induced both early and late apoptosis of CAL-27 cells, observed by flow cytometric measurement and microscopic fluorescent observation. A comparative proteomic analysis was conducted to investigate the effects of 11-dehydrosinulariolide on CAL-27 cells at the molecular level by comparison between the protein profiling (revealed on a 2-DE map) of CAL-27 cells treated with 11-dehydrosinulariolide and that of CAL-27 cells without the treatment. A total of 28 differential proteins (12 up-regulated and 16 down-regulated) in CAL-27 cells treated with 11-dehydrosinulariolide have been identified by LC-MS/MS analysis. Some of the differential proteins are associated with cell proliferation, apoptosis, protein synthesis, protein folding, and energy metabolism. The results of this study provided clues for the investigation of biochemical mechanisms of the anti-tumor effects of 11-dehydrosinulariolide on CAL-27 cells and could be valuable information for drug development and progression monitoring of oral squamous cell carcinoma (OSCC)

    Sustained proliferation in cancer: mechanisms and novel therapeutic targets

    Proliferation is an important part of cancer development and progression. This is manifest by altered expression and/or activity of cell cycle related proteins. Constitutive activation of many signal transduction pathways also stimulates cell growth. Early steps in tumor development are associated with a fibrogenic response and the development of a hypoxic environment which favors the survival and proliferation of cancer stem cells. Part of the survival strategy of cancer stem cells may manifested by alterations in cell metabolism. Once tumors appear, growth and metastasis may be supported by overproduction of appropriate hormones (in hormonally dependent cancers), by promoting angiogenesis, by undergoing epithelial to mesenchymal transition, by triggering autophagy, and by taking cues from surrounding stromal cells. A number of natural compounds (e.g., curcumin, resveratrol, indole-3-carbinol, brassinin, sulforaphane, epigallocatechin-3-gallate, genistein, ellagitannins, lycopene and quercetin) have been found to inhibit one or more pathways that contribute to proliferation (e.g., hypoxia inducible factor 1, nuclear factor kappa B, phosphoinositide 3 kinase/Akt, insulin-like growth factor receptor 1, Wnt, cell cycle associated proteins, as well as androgen and estrogen receptor signaling). These data, in combination with bioinformatics analyses, will be very important for identifying signaling pathways and molecular targets that may provide early diagnostic markers and/or critical targets for the development of new drugs or drug combinations that block tumor formation and progression

    Whitebark Pine Stand Condition, Tree Abundance, and Cone Production as Predictors of Visitation by Clark's Nutcracker

    Accurately quantifying key interactions between species is important for developing effective recovery strategies for threatened and endangered species. Whitebark pine (Pinus albicaulis), a candidate species for listing under the Endangered Species Act, depends on Clark's nutcracker (Nucifraga columbiana) for seed dispersal. As whitebark pine succumbs to exotic disease and mountain pine beetles (Dendroctonus ponderosae), cone production declines, and nutcrackers visit stands less frequently, reducing the probability of seed dispersal.We quantified whitebark pine forest structure, health metrics, and the frequency of nutcracker occurrence in national parks within the Northern and Central Rocky Mountains in 2008 and 2009. Forest health characteristics varied between the two regions, with the northern region in overall poorer health. Using these data, we show that a previously published model consistently under-predicts the proportion of survey hours resulting in nutcracker observations at all cone density levels. We present a new statistical model of the relationship between whitebark pine cone production and the probability of Clark's nutcracker occurrence based on combining data from this study and the previous study.Our model clarified earlier findings and suggested a lower cone production threshold value for predicting likely visitation by nutcrackers: Although nutcrackers do visit whitebark pine stands with few cones, the probability of visitation increases with increased cone production. We use information theoretics to show that beta regression is a more appropriate statistical framework for modeling the relationship between cone density and proportion of survey time resulting in nutcracker observations. We illustrate how resource managers may apply this model in the process of prioritizing areas for whitebark pine restoration

    Partonic flow and ϕ\phi-meson production in Au+Au collisions at sNN\sqrt{s_{NN}} = 200 GeV

    We present first measurements of the ϕ\phi-meson elliptic flow (v2(pT)v_{2}(p_{T})) and high statistics pTp_{T} distributions for different centralities from sNN\sqrt{s_{NN}} = 200 GeV Au+Au collisions at RHIC. In minimum bias collisions the v2v_{2} of the ϕ\phi meson is consistent with the trend observed for mesons. The ratio of the yields of the Ω\Omega to those of the ϕ\phi as a function of transverse momentum is consistent with a model based on the recombination of thermal ss quarks up to pT4p_{T}\sim 4 GeV/cc, but disagrees at higher momenta. The nuclear modification factor (RCPR_{CP}) of ϕ\phi follows the trend observed in the KS0K^{0}_{S} mesons rather than in Λ\Lambda baryons, supporting baryon-meson scaling. Since ϕ\phi-mesons are made via coalescence of seemingly thermalized ss quarks in central Au+Au collisions, the observations imply hot and dense matter with partonic collectivity has been formed at RHIC.Comment: 6 pages, 4 figures, submit to PR

    The energy dependence of ptp_t angular correlations inferred from mean-ptp_{t} fluctuation scale dependence in heavy ion collisions at the SPS and RHIC

    We present the first study of the energy dependence of ptp_t angular correlations inferred from event-wise mean transverse momentum fluctuations in heavy ion collisions. We compare our large-acceptance measurements at CM energies $\sqrt{s_{NN}} =$ 19.6, 62.4, 130 and 200 GeV to SPS measurements at 12.3 and 17.3 GeV. $p_t$ angular correlation structure suggests that the principal source of $p_t$ correlations and fluctuations is minijets (minimum-bias parton fragments). We observe a dramatic increase in correlations and fluctuations from SPS to RHIC energies, increasing linearly with $\ln \sqrt{s_{NN}}$ from the onset of observable jet-related fluctuations near 10 GeV.Comment: 10 pages, 4 figure

    Diversity, distribution and conservation status of island conifers: a global review

    Aim Conifers comprise an ancient and diverse group of plants showing a wide distribution range. To better understand the general patterns of species successfully established on islands, this review compiles information about the distribution, diversity, dispersal potential and conservation status of insular conifers, with special emphasis on those inhabiting remote oceanic islands. Location Global. Methods An exhaustive survey was made of world-wide databases and literature. We registered information on island distribution (including ocean region, extension and geological origin of the island), endemism and threat status for each insular conifer. Results 285 of the 547 conifer species considered in this review show an insular distribution (i.e. their distribution encompass insular territories). The family Podocarpaceae is best represented, with 40% of the insular species. The importance of endozoochory for long-distance dispersal is clear, because it was the most frequent dispersal syndrome among oceanic conifers. A high proportion of the total threatened conifers occur on islands (52%), and many of them are insular endemics (72%). Among conifer families, Araucariaceae is the most threatened in insular territories. Main conclusions Our results highlight the wide diversity of insular conifers, as well as the key role of oceanic islands in catalysing speciation mechanisms. Pacific islands in particular harbour the greatest diversity levels, constituting a major centre of diversification. The wide distribution of conifers reflects their great potential for dispersal and colonization, endozoochory being the most favourable dispersal syndrome for reaching remote islands.     Measurement of Transverse Single-Spin Asymmetries for Di-Jet Production in Proton-Proton Collisions at s=200\sqrt{s} = 200 GeV

    We report the first measurement of the opening angle distribution between pairs of jets produced in high-energy collisions of transversely polarized protons. The measurement probes (Sivers) correlations between the transverse spin orientation of a proton and the transverse momentum directions of its partons. With both beams polarized, the wide pseudorapidity (1η+2-1 \leq \eta \leq +2) coverage for jets permits separation of Sivers functions for the valence and sea regions. The resulting asymmetries are all consistent with zero and considerably smaller than Sivers effects observed in semi-inclusive deep inelastic scattering (SIDIS). We discuss theoretical attempts to reconcile the new results with the sizable transverse spin effects seen in SIDIS and forward hadron production in pp collisions.Comment: 6 pages total, 1 Latex file, 3 PS files with figure

    Energy and system size dependence of \phi meson production in Cu+Cu and Au+Au collisions

    We study the beam-energy and system-size dependence of \phi meson production (using the hadronic decay mode \phi -- K+K-) by comparing the new results from Cu+Cu collisions and previously reported Au+Au collisions at \sqrt{s_NN} = 62.4 and 200 GeV measured in the STAR experiment at RHIC. Data presented are from mid-rapidity (|y|<0.5) for 0.4 < pT < 5 GeV/c. At a given beam energy, the transverse momentum distributions for \phi mesons are observed to be similar in yield and shape for Cu+Cu and Au+Au colliding systems with similar average numbers of participating nucleons. The \phi meson yields in nucleus-nucleus collisions, normalised by the average number of participating nucleons, are found to be enhanced relative to those from p+p collisions with a different trend compared to strange baryons. The enhancement for \phi mesons is observed to be higher at \sqrt{s_NN} = 200 GeV compared to 62.4 GeV. These observations for the produced \phi(s\bar{s}) mesons clearly suggest that, at these collision energies, the source of enhancement of strange hadrons is related to the formation of a dense partonic medium in high energy nucleus-nucleus collisions and cannot be alone due to canonical suppression of their production in smaller systems.Comment: 20 pages and 5 figure
